Immunity parameters in mice of different strains.

Quantitative composition and functional activity of immunocompetent cells differ in mice of different strains. The counts of T cells in the bone marrow and spleen, proliferative activity of T cells in the spleen, levels of IL-2 and IL-10 production by splenic T cells, number of antigen-specific T cells and their functional activity are low in C57Bl/6, BALB/c, and CC57W mice and high in CBA/CaLac, DBA/2, and C3H animals. Low phagocytic activity of peritoneal macrophages was detected in BALB/c and CC57W mice and high activity in C3H animals. The content of antibody-producing cells in the spleens of C57Bl/6, BALB/c, and CC57W mice is higher than in CBA/CaLac, DBA/2, C3H, A/SN, and AKR/JY mice. Functional activity of B cells is lower in BALB/c and CC57W compared to CBA/CaLac and DBA/2 mice.

Morphological and functional peculiarities of lymphoid cells in mice of different strains.

We revealed differences in quantitative composition and functional activity of lymphoid cells in intact mice of different strains. Cellularity and counts of lymphoid elements in hemopoietic and lymphoid organs, proliferative activity of T and B lymphocytes, and counts of CD4+ and CD8+ lymphocytes in the spleen were minimum in CC57W and Balb/c mice and maximum in CBA/CaLac and DBA/2 mice. The highest absolute content of lymphoid elements in the spleen was detected in Balb/c mice, while CC57W mice had the highest content of these elements in the thymus.

Sensitivity of immunocompetent cells of DBA/2 and C57Bl/6 mice to cyclophosphamide.

T cells were most sensitive to cyclophosphamide in DBA/2 mice, while in C57Bl/6 mice both T and B cells were sensitive. The formation of antibody-producing cells and the production of specific antibodies were delayed in DBA/2 mice immunized after pretreatment with antitumor drug. Accumulation of antibody-producing cells in the spleen was more active in immunized C57Bl/6 mice treated with cyclophosphamide compared to animals not treated with cyclophosphamide.

Immunotropic effects of potentiated antibodies to human interferon-gamma.

We studied immunotropic properties of antibodies to human interferon-gamma in homeopathic doses. The effects of these antibodies on the ratio between the counts of T helper (CD4+) and T suppressor cells (CD8+), production of interleukin-1 and interleukin-2, and functional activity of natural killer cells were evaluated. Potentiated antibodies slightly decreased the number of CD8+ cells in the spleen of experimental animals. After the addition to cultured human mononuclear cells the preparation produced a moderate comitogenic effect, stimulated proliferation of T and B lymphocytes, increased spontaneous and induced production of interleukin-1, and enhanced functional activity of natural killer cells (under certain conditions).

Ultralow doses of anti-idiotypic antibodies to human gamma-interferon: immunotropic properties.

We studied the effects of ultralow doses of anti-idiotypic antibodies to human gamma-interferon on humoral immune response in experimental animals immunized with a thymus-dependent antigen, delayed-type hypersensitivity, and phagocytic activity of neutrophils in the peritoneal exudate. This preparation stimulated the humoral immune response and activated phagocytosis, but had no effect on functional activity of T cells.

Reactions of immune system to immobilization stress in inbred mice of different strains.

Mice of different strains can be classified by the intensity of humoral immune reaction to thymus-dependent antigen as high, moderate, and low responders. High and moderate responders (CBA/CaLac, DBA/1, BALB/c) are characterized by high sensitivity of the productive phase of humoral immune response and phagocytic activity of macrophages to immobilization stress. In low responders (C57BL/6, CC57W) stress only slightly affected the productive phase of the humoral immune response, but activity of peritoneal macrophages decreased. These differences in the reactions of the immune system of inbred mice after immobilization stress reflect different reactions of the immune system to extreme factors.

Immune reactions in different mouse strains.

The responses of immunocompetent cells to thymus-dependent antigen differ in mice of different strains. Immunization stimulated phagocytic activity of peritoneal macrophages in CBA/CaLac, DBA/2, and BALB/c mice and suppressed it in CC57W mice. By the formation of antibody-producing cells in the spleen in response to thymus-dependent antigen DBA/2 and CBA/CaLac mice can be classified as high responders, BALB/c mice as medium-responders, and C57Bl/6 and CC57W mice as low responders.

Study on conformational changes in hemoglobin protoporphyrin in essential hypertension.

Changes in protoporphyrin conformation, partial pressures of O2 and CO2, and the mechanisms responsible for regulation of pCa and pH in erythrocytes were studied in essential hypertension (EH). Changes in protoporphyrin conformation in EH were accompanied by a decrease in the partial pressure of O2 and an increase in the partial pressure of CO2. This was associated with increased activities of Na+/H+-exchange and Ca2+-dependent K+-channels and with a decreased activity of Ca2+-ATPase. The changes in protoporphyrin conformation in EH are suggested to decrease the efficiency of O2 metabolism in hemoglobin and increase the values of intracellular pCa and pH of erythrocytes.

Complexes produced by associated forms of porphyrin bound with hydrophobic-hydrophilic copolymer and transition metal ions.

Properties of protonated dimeric forms of meso-tetraphenylporphine (TPP) and meso-tetra(p-aminophenyl)porphine (TAPP) bound with copolymer and also complexes produced by associated TAPP bound with copolymer, Mn2+, and Fe3+ are investigated by absorption, luminescence, and Raman spectroscopy. According to absorption spectra of protonated dimers of TPP, three dimeric forms of the porphyrin are observed in the ground state. However, selective excitation of these forms according to the fluorescence spectra reveals only two dimeric forms in the excited state. In contrast, similar selective excitation of TAPP bound with copolymer in aqueous-dioxane solution results in weak changes in the fluorescence spectra, nevertheless, there is strong interaction between porphyrin and macromolecular carboxyl groups in the ground state. In the case of the formation of the complexes between associated TAPP bound with copolymer, Mn2+ and Fe3+, a new band in the near IR region with a maximum at 840 nm is built up in the fluorescence spectrum. However, this near IR emission is completely quenched when new strong vibrational bands at approximately 1800 and 1900 cm-1 are revealed in the resonance Raman spectra of the complexes. The observed effects are explained in terms of direct participation of water molecules involved in the water-porphyrin dimeric complex in the processes of transformation of excitation energy. The involvement of water in this dimeric complex can lead to redistribution of flows of the energy degradation when transition metal ions play a role of the agent which enhances the trapping properties of the porphyrin-metal-ions complexes.

Transition metal ion complexes with associates of porphyrin bound to a hydrophobic-hydrophilic copolymer.

The interaction of dimeric forms of meso-tetraphenylporphine with Mn2+ as well as the interaction of associated forms of meso-tetra(p-aminophenyl)porphine bound to a hydrophobic-hydrophilic copolymer with Mn2+ and Fe3+ were studied by absorption, luminescence, Raman, and EPR spectroscopies. Both dimeric and associated forms of these porphyrins produced Mn2+ complexes. Manganese ions in these complexes undergo clusterization, which is accompanied by transformation of the six-line EPR signal of Mn2+ into a broad single-line signal. The EPR signal of Mn2+ in these clusters is characterized by a g-factor value typical of a free electron with half-width DeltaHpp = 50 mT. The interaction of the two-component complex with Fe3+ produces a donor-acceptor complex. The electronic spectrum of the donor-acceptor complex contains a broad band with a maximum at 760 nm. The molar extinction coefficient of the complex at 760 nm is 9.1.104 M-1.cm-1, and the rate constant for its formation is Kdac = (3.9 +/- 0.6).106 M-1. The constants for Mn2+ binding to the organic compounds used in this work were also determined.

Electrochromic behaviour of carotenoid molecules in nerve cell membranes: a resonance Raman study.

It was found that, following the depolarization of cell membranes of frog sciatic nerve, the 1521 and 1156 cm-1 resonance Raman (RR) bands of carotenoids showed an increase in intensity of 20%, 15% and 4% at excitation wavelengths of 488, 496.5 and 514.5 nm respectively. None of the bands revealed any change with excitation at 441.6, 457.9, 472.7 and 476.5 nm. These results can be explained by an electrochromic shift of the excitation profile of the RR bands by about 0.5 nm to the red. A dependence of the 1521 cm-1 band intensity on transmembrane potential was obtained. Under the effect of lanthanum ions, which are known to modify the surface charge of the membrane, the intensity of the Raman bands decreased by 15%-20% (excitation, 488.0 nm). The results obtained indicate that RR spectroscopy of carotenoids can be used to monitor changes in the transmembrane potential and surface charge of nerve cell membranes.

[The mechanisms of the porphyrin conformation of normal blood hemoglobin and in pathology].

The spectra of resonance Raman scattering of blood in norm and under pathology (myocardial infarction and sepsis), as well after artificial hemotransfusion or UV photomodification have been studied. It has been shown that under heart pathology the structure of hemoglobin porphyrin macrocycle of erythrocytes changes, the size of porphyrin "nucleus" increases. The opposite conditions are observed at blood sepsis. It has been found that the traditional methods in tissue restoration, hemotransfusion and UV photomodification of blood don't result in complete restoration of hemoporphyrin molecule.

[Raman spectra of somatic nerves under different functional conditions].

The light scattering spectra (LS) of different kind of somatic nerves during excitation and its block has been investigated. In the LS spectra of frog nerves the spectra of C40-carotenoids was dominated by the following bands at 1008 cm-1, 1160 cm-1 and 1526 cm-1. The peaks of LS were decreased during the excitation and under the action of ouabain and increased under the action of ether. The changes of LS spectra can be explained by shift of maximum of light intensity of C40-carotenoids uptake and positions of side CH3-radical in polyene chain.
