[Affinity modification of the 40S subparticle from human placenta with derivatives of pAUG and pAUGU3].

Affinity labeling of 40S subunits from human placenta with 4-(N-2-chloroethyl-N-methylamino)benzylmethyl-[32P]phosphoamide s of oligoribonucleotides pAUG and pAUGU3 was studied. Covalent attachment of these derivatives to 40S subunits within the complexes with 40S subunits, formed in the presence of Met-tRNAf.eIF-2.GTP, was detected. Both rRNA and ribosomal proteins were modified. Fragments of 18S rRNA, containing sites of the reagent attachment were identified: 1058-1164 for pAUG derivative and 976-1057--for pAUG and pAUGU3 ones. The data obtained allowed to conclude that the presence of the neighbouring codon at the A-site, regardless of the presence of the tRNA in it, affects significantly the arrangement of the trinucleotide template in the codon-anticodon interaction region. The large subunit does not cause significant alterations in the structural organization of the codon-anticodon interaction region.

[Interaction of human S26 ribosomal protein with fragments of mRNA and pre-mRNA for this protein in Hela nuclear cell extracts].

As shown by nitrocellulose filtration assays with RNA fragments transcribed from various regions of the human ribosomal protein (rp) S26 gene, recombinant rpS26 binds to the first intron of the rpS26 pre-mRNA (apparent association constant (Ka) approximately 5.0 x 10(7) M-1) and, to a lesser extent, to the rpS26 mRNA (Ka approximately 2.0 x 10(7) M-1). The binding was specific, since human rpS19 had an order of magnitude lower Ka with the first intron and did not bind with the rpS26 mRNA. Immunoassays with specific antibodies showed that rpS26 contained in the nuclear extract of HeLa cells binds to the first intron of its pre-mRNA and, less efficiently, to its mRNA. In either case, RNA binding substantially increased in the presence of recombinant rpS26. Along with other (48 K, 59 K) nuclear proteins, rpS26 was assumed to form complexes, the functional role of which is storage of pre-mRNAs inactive in splicing.

[Approach to identifying the functionally important segments of RNA, based on complementation-addressed modification].

An approach based on complementation-addressed modification of nucleic acids by oligodeoxyribonucleotide derivatives was proposed for changing the spatial structure of particular RNA sites in order to study their role in the biological activity of the total RNA molecule. Hepatitis C virus (HCV) IRES was used as a model. Oligodeoxyribonucleotide derivatives contained a 4-[N-(2-chloroethyl)-N-methylamino]benzylamino group at the 5'-P and were complementary to various RNA sites located in regions of hairpins II, IIId, or IIIe. Covalent adducts resulting from RNA alkylation with the derivatives were isolated by denaturing PAGE and tested for binding with the 40S subunit of human ribosomes. Structural alteration of hairpin II had no effect, whereas alteration of hairpin IIIe substantially reduced the binding. The RNA with modified hairpin IIId showed virtually no binding with the 40S subunit. Hairpin IIId was assumed to play a critical role in the binding of HCV IRES with the 40S subunit.

[Human ribosomal protein S26 inhibits splicing of its own pre-mRNA].

In vitro splicing was studied for a human ribosomal protein (rp) S26 pre-mRNA fragment containing the first exon, first intron, and a part of the second exon. Splicing yielded two products, the first was corresponded to a fragment of the mature rpS26 mRNA and another was retained the 19 3'-terminal nucleotides of the first intron between the first and second exons. Recombinant rpS26 inhibites generation of both splicing products in vitro. The inhibition was specific, because another recombinant human rp, S19, had no effect on the splicing of the pre-mRNA fragment. Toe-printing was used to map the spS26-binding sites of the per-mRNA within the regions of the conventional and alternative 3' splicing sites of the first intron. On the strength of the rusults, rpS26 was assumed to regulate the expression of its own gene at the level of pre-mRNA splicing via a feedback mechanism.

[Binding of human ribosomal protein S5 with the 18S rRNA fragment 1203-1236/1521-1698].

Human ribosomal protein S5 is a homologue of prokaryotic ribosomal protein S7 that binds to the 16S rRNA region formed by helices H28-30 and H41-43 and contacts this site within the 30S ribosomal subunit. To date, one of the most available approaches to study the structure of the eukaryotic ribosome is based on studying binding of rRNA transcripts with recombinant ribosomal proteins. In the present work, we report studying interaction of human recombinant protein S5 with an RNA transcript "corresponding to the region 1203-1236/1521-1698 (helices H28-30 and H41-43) of human 18S rRNA. The homologous region of 16S rRNA is known to bind protein S7. The apparent association constant of S5 with the transcript was determined. Nucleotide residues of the transcript changing their accessibility for cleavage with RNases T1 and T2 in the presence of S5 were determined by the enzymatic footprinting. It was shown that these nucleotides are located preferentially in the internal loop of the fragment formed by basal parts of the helices H29, H30 and H41 that agrees with the data on the S7 binding to 16S rRNA and on its location within the 30S ribosomal subunit. Besides, we also found nucleotides in the transcript regions corresponding to 16S rRNA regions that do not contact S7 in the 30S subunit.

[Eukaryotic ribosomal proteins: interaction with their own pre-mRNAs and involvement in the splicing regulation].

The present review summarizes data concerning regulation of eukaryotic ribosomal protein genes expression at the splicing step, including own results. In particular, roles of the ribosomal proteins in regulation of splicing of their coding pre-mRNAs are considered. Special attention is devoted to discussion of the molecular mechanisms that underlie the process and to the analysis of interactions of ribosomal proteins with their own pre-mRNAs and mRNAs. Besides, critical consequences arising by disturbances in the mechanisms of regulation of ribosomal proteins biosynthesis in the cell are noted in the review. Special role of autoregulation in the maintenance of the normal level of ribosomal protein biosynthesis is underlined in the conclusion.

[Characteristics of nonenzymatic binding of oligoribonucleotides-- analogs of mRNA and Phe-tRNA Phe with 80S ribosomes from human placenta].

Binding of labelled oligouridylates--mRNA analogs--to human placenta 80S ribosomes in the presence of Phe-tRNAPhe has been studied. The single site for (pU)n (n = 6, 9, 13) binding on the ribosome was found; association constants for their tRNA-dependent binding were evaluated. In the presence of oligouridylates as templates [14C]Phe-tRNAPhe was found to be able to bind simultaneously at acceptor and donor ribosomal sites which resulted in diphenylalanine formation. The observed maximum Phe-tRNAPhe binding level was considerably lower than for the corresponding oligouridylate binding; the longer oligouridylate the higher Phe-tRNAPhe maximum binding level. To explain the results obtained we have proposed that (i) (Phe)2-tRNA produced from transpeptidation dissociates from the ribosomal A site to a significant extent and (ii) when oligouridylate length increases its interaction with 3'-side of mRNA binding center results in allosteric stabilization of the complex of peptidyl-tRNA with the ribosome at A site.

[Functional value of 980-1061 sequences of human 18S ribosomal RNA using complementary DNA probes].

Region 980-1061 in human 18S rRNA was chosen on the basis of our previous results indicating, that the cross-linking sites of alkylating mRNA analogs are located within this region. In the present study, we have used 10 DNA 15-mers complementary to various overlapping sequences within the 18S rRNA positions 980-1061. Their ability to bind selectively at the desired rRNA sequences was proved by hydrolysis of 18S rRNA within heteroduplexes with the corresponding probes by RNase H. Only four of the probes were able to bind to 40S subunits indicating, that the corresponding 18S rRNA sequences 980-994, 987-1001, 1025-1039 and 1032-1046 are exposed within the subunits. None of the probes inhibited tRNA-dependent binding of oligo(U) messengers to 40S subunits. Nevertheless, two probes (complementary to 18S rRNA sequences 987-1001 and 1025-1039) being covalently attached to 40S subunits, inhibited translation of poly(U) by human 80S ribosomes in a cell-free system. The binding of messenger trinucleotide in the complex pAUG.40S.Met-tRNA.eIF-2.GTP was strongly affected by the same oligomers. Thus 987-1001 and 1025-1039 18S rRNA sequences are supposed to be involved in interaction with mRNA in the course of translation.
