[New data on the leukocyte functional morphology in pyo-septic processes].

It is established by electron microscopical radioautography that sepsis results in the lowering of neutrophils bacterial capacity while their absorptive capacity is retained. Living bacteria may be released from neutrophils undergoing degradation into the environment. The same is observed when phagocytized but still living bacteria are located in phagosomes which are fused with one another and with neutrophil plasma membrane; after the destruction of the latter bacteria are released into the extracellular space. This is a negative issue for the host of the incomplete phagocytosis. Neutrophils participate more actively in the lysis of the necrotic tissues and wound cleaning than in the bacteria phagocytosis.

[An electron microscopic radioautographic study of the viability of human cells after death. The postmortem activation of RNA synthesis in neutrophils].

RNA synthesis in human neutrophils isolated from blood 2.5-67 hours after death of patients was studied by means of light and electron microscopy radioautography. The rate of RNA synthesis in neutrophils isolated from blood of cadavers was higher in comparison with the same parameter in neutrophils isolated from survivor's blood. The significance of this observation for resuscitation and transplantology is discussed.

[Role of bacterial agglutination in wound infection].

Method of study of bacterial agglutination by means of serum taken from patients with wound infections was suggested. It was found out that agglutination suppresses viability of microbial cells in the center of agglutinate and reduces the risk of bacterial spreading from the wound over the body.

[A comparative radioautographic analysis of DNA synthesis by enterocytes of the small intestine in in vivo and in vitro experiments].

A method of radioautography was used to study DNA synthesis in mice enterocytes as an index of their proliferative activity. Cells were labelled by means of incorporation [3H]thymidine in vivo and in vitro. The program was worked out to computed analysis of the reflected light intensity of obtained radioautographs and estimation of reliability data. Our experiments showed that thymidine diffused very slowly.

[Method of radioautographic study of cell cultures].

The method of radioautographic study of cell cultures is suggested. It is based on culture embedding into epoxy resin in situ. After this manipulation the photolayer on the lower surface of the polymerized epoxy unit with the cell culture in it is applied. The advantages of this method are discussed.

[Correlations of intracellular and cellular regeneration of the epithelium of the kidney tubules in necronephrosis].

The study of radioautographs in ultrathin and semithin sections extracted from animal tissues and following the impulsive as well as long-term injection of 3H-thymidine was made. Intracellular regeneration processes (external cell membrane synthesis, DNA reparation, restoration of the lost albumin-synthesizing complex and cytoskeleton) turned to develop in proximal canals of epithelial cells. The processes were aimed at the liquidation of distortions preventing cell from entering mitosis.

[Electron microscopic-radioautographic study of RNA synthesis in the epithelial cells of the kidney urinary tubules of white rats in corrosive sublimate poisoning].

Radioautographic study of ultrathin selections of proximal white rat renal canals labeled with 3H-uridine revealed diffuse transition of peripheral and paranuclear heterochromatin to be observed in nuclei of undamaged epithelia cells and those affected by partial necrosis in case of sublimate nephrosis. Nucleus degranulation as well as decrease in its size accompanied by inhibition of RNA synthesis is also observed. Nucleus reparation being observed 48 hours after subcutaneous injection of 0.4/0.1 kg corrosive sublimate in some cells and characterized by the appearance of granular component as well as the increase of nucleolar vacuole count is usually revealed in case of decondensation form of peripheral and paranuclear chromatin in the enlarged nuclei.

[Electron microscopic-radioautographic study of cultured human epidermocytes].

A split thickness skins of burned patients were treated by trypsin and received the epidermocyte suspension. In that suspensions the content of basal keratinocytes was 10 to 70% in dependence from the treating conditions. As a rule, cultivation was a success only in those cases, when among basal keratinocytes of initial suspension were cells, synthesized DNA. Cultures were grown more rapidly under decrease of ion calcium concentration in nutritional medium up to 0.14 mM and in the presence of cell-free surface of substratum. At such conditions connections between cells were weakened, desmosomes disappeared, stratification and colony formation were broken.

Plastic operations of burns using cultured epithelium.

Autoradiographic and electron-microscopic methods were used for the study of cultures of human skin epidermocytes in media with a low calcium content. It has been demonstrated that the culturing of epidermocytes in a medium containing 0.15 mM of calcium suppresses differentiation and stimulates division of cells. The considerable number of dividing cells in the cultures permits their use both for grafting on wound surfaces and further cultivation.

[Electron-radioautographic study of phagocytosis of Pseudomonas aeruginosa by neutrophils].

RNA synthesis was studied in the bacteria and neutrophils from healthy persons having phagocytosis. Elevation of RNA synthesis occurred in the neutrophils with phagocytic activities. The rate of labelled RNA precursor incorporation into the bacteria was used as a measure of their viability. This measure made it possible to judge the bactericidal action displayed by neutrophils, to follow the progressive suppression of bacterial vital activity at the stages of phagocytosis, and to detect the fusion of phagosomas varying in their life duration. The phagosomal fusion is a factor that promotes the retention of viability shown by some bacteria following phagocytosis. It was demonstrated that even ultrastructural morphological characterization of bacterial cells is not always a reliable criterion for their viability.

[The effect of the seeding density on the growth of a culture of human epidermocytes].

The growth of the culture of epidermocytes of man has been studied. Regardless of the initial density of seeding the authors revealed a considerable difference in the structure of the cellular layer and the rate of the cell proliferation at separate parts of the fundus. In seedings of diluted cellular suspensions the authors observed insignificant increase in the rate of proliferation.

[Effects of gentamycin on interactions of neutrophils with Staphylococcus].

The authors studied the effect of gentamicin on abilities of neutrophils to uptake and killing of extracellular Staphylococcus. Gentamicin had not changed naturally the ability of healthy men and burn patients neutrophils to uptake Staphylococcus, but intensified the ability of neutrophils to killing of these bacteria.

[Various mechanisms of reducing the vascular system of granulation tissue in the process of its maturation].

Rat granulation tissues were histologically and electron radioautographically examined at Days 7, 20, 30, 40, 53, and 60 after making a 3 X 4-cm2 wound in the back. During the entire period of cicatrization, alterations were shown to occur in the capillaries, as manifested by destructive changes in endotheliocytes, break in their contacts, thinning of the capillary wall with occasional lacerations, disintegrity of the basal membrane and, finally, vessel-composing cell dissociation and departure, as well as unusual arrangement among other cells and fibrous structures. On the basis of the findings it is proposed that the known diminished numbers of vessels during granulation tissue maturation involves not only disappeared vessels due to their progressive dystrophic changes, but the described unusual capillary alterations whereby the vascular wall cells are gradually converted to the cellular elements freely located in the interstitial tissue.

[Radioautographic research on human epidermocytes cultured in a medium with a low calcium content].

The process of human skin epidermocyte culturing in low calcium medium has been studied with radioautographic and electron microscopic methods. Epidermocyte culturing in 0.15 mM calcium medium has been proved to suppress the differentiation inside as well as to stimulate cell dividing process. Considerable amount of dividing cells in cultures makes it possible to use them both for grafting on wound surfaces and for the further cultivation.

[Characteristics of the cultivation of epidermocytes on collagen substratum].

Epidermocytes obtained from human split-thickness skin were cultured in hypocalcium medium (ion calcium content was 0.14 mM). The half bottoms of culture plastic dishes were coated with collagen film before bringing cells in these dishes. It was proved that epidermocytes attach more quickly to collagen, than to plastic substratum, incorporate H3-thymidine (at early periods of cultivation) with larger rate and form and monolayer more quickly, than on the plastic substratum.

[Specificities of bacterial death in culture and suppurative wounds].

Electron microscopic autoradiographic study has been performed. In tissue sections, obtained from suppurative wounds, diploforms of cocci and bacilli have been observed. In the former cells DNA and RNA synthesis was normal, however, in the latter cells no synthesis was detected. Such diploforms were not observed in fresh and exhausted cultures. Possible existence in the body of a factor directly injuring bacterial genome is discussed.

[Radioautographic demonstration of the intracellular and extracellular bactericidal action of the neutrophils].

The authors described a method of quantitative radioautographic analysis of RNA synthesis level in the bacteria. The method allows to define by inhibition of RNA synthesis a bactericidal action of neutrophils during phagocytosis. The essence of a method consists in the introduction for a short period of a label and development of radioautographs at semithin section by paraphenylenediamine which form a small and numerous silver grains above bacteria which retained an ability to RNA synthesis. Preparations obtained in this way is studied in a light optical microscope using a double illumination: from below through a condenser and from above through an objective.
