[Phagocytosis of bacteria by polymorphonuclear leukocytes suspended in liquid or adhering to a surface].

Phagocytosis of Staphylococcus aureus by polymorphonuclear leukocytes (PNL) was studied in healthy men. PNL suspended in nutrient medium did not practically ingest bacteria. The intake of bacteria got considerably intensified if leukocytes and bacteria ran into each other by turning over the test tubes during incubation. A still greater rise of phagocytic activity was discovered under the conditions favouring the attachment of PNL to the surface and the possibility of chemotaxis. These conditions were created by introducing a gelatin-coated film into the test tube.

[Vascular cellular elements as the source of development of desmoid fibroma].

Electron microscopic radioautography was used to investigate DNA and RNA syntheses in desmoid fibroma pieces ablated at operation. The DNA precursor 3H-thymidine was discovered to be incorporated only into vascular wall cells. RNA synthesis was the most overt in endotheliocytes, pericytes and fibroblasts adjacent to the vessels. At a distance from the vessels the majority of the cells were in a state of destruction. Based on the data obtained the authors arrive at the conclusion that the proliferation of vascular wall cells gives rise to the development of desmoid fibroma providing for the replenishment of tumorous cells. The latter ones, while making the cycle of differentiation and specific function, move away from the vascular lumen and then get destroyed.

[Several regularities of the interrelation of blood leukocytes and microbes in the infectious process].

The rate of absorption by polymorphonuclear leukocytes of different strains of staphylococci present in a homogeneous mixture was studied. The cells of one of the strains were radiolabeled and this made it possible to differentiate them from the cells of another strain by means of electron microscopic autoradiography. Different rates of absorption of cocci belonging to different strains by leukocytes of normal subjects and by those from patients with sepsis resulting from burn or wound were observed. The seizing capacity of leukocytes of such patients in relation to the autostrains of S. aureus and epidermal staphylococcus cultivated from the blood did not differ significantly from normal in numerous tests. The electron-autoradiographic examinations of the synthesis of nucleic acids in cultures of S. aureus and epidermal staphylococcus revealed different functional status of individual cells.

[Electron microscopic and autoradiographic study of keloid scars].

The skin surrounding postburn keloid cicatrices showed high proliferative and functional activity of the cell elements comprising the vessel wall or located near it. Keloid cicatrices differed from the surrounding skin by the lower content and lower activity of cell elements in general and mast cells in particular. Here too, however, the highest intensity of biosynthesis was found in blood vessels and perivascular zones. Both in the keloid and the skin surrounding it, more frequently distally from the blood vessels, there was a large number of cells in which RNA and protein synthesis could not be detected autoradiographically, and destructive and necrotic lesions were observed. Not infrequently such cells were in immediate proximity to biosynthetically active cells.

[Electron microscopic and autoradiographic study of the interaction of human polymorphonuclear leukocytes and staphylococci].

Leukocyte suspensions of normal subjects and patients with burns and wounds were incubated with Staphylococcus epidermidis and incorporation of 3H-uridine into leukocytes and bacteria was examined by electron microscopic autoradiography. As the maturation of the precursor cells of polymorphonuclear leukocytes (PMNL) progressed the level of RNA synthesis in them was found to decrease gradually and in PMNL RNA synthesis ceased completely. If PMNL take part in phagocytosis, however, RNA synthesis is found in a considerable number of them. The results indicate that RNA synthesis in PMNL is associated with the formation and functional condition of primary and secondary granules. Certain conditions affecting the viability of staphylococci ingested by phagocytes are described.

[Characteristics of RNA production in the body of the nucleolus studied by an analysis of serial electron microscopic radioautographs].

Electron microscopic radioautography was used to examine the changes in the density of silver grains over the serial sections of neurons of the fifth layer of the rat cortex after injection of 3H-uridine. In the majority of the neurons under study, the changes in the density of silver grains over the sections of nucleoli did not correlate with those in the density over the sections of the extranucleolar part of the nucleus. On the basis of these findings the conclusion is drawn that essential differences in the grain density in the neighbouring sections of the nucleolus are caused by non-uniformity of distribution of newly synthesized RNA in the nucleolus rather than by the errors of the method described. Possible reasons for such a non-uniformity are discussed.

[Development method providing fine-grain electron microscopic radioautographs].

A modification of the development of electron microscopy autoradiographs with p-phenylenediamine is suggested. The development in the mode described was found fairly convenient for bacteriological studies. The method provides small silver grains specific in shape, which leave open the greater part of a bacterium even in the presence of considerable accumulation of silver grains. The grains can be readily counted. The development results are reasonably reproducible.

[Periodicity of RNA synthesis in human adipocytes].

Electron microscopy and autoradiography were used to disclose remarkable differences in the RNA synthesis rate by morphologically related human adipocytes. These differences might be accounted for by the fact that the RNA synthesis rate in individual adipocytes does not remain at the same level but undergoes periodic changes. The phasic changes are unlikely to coincide in different adipocytes. The evidence obtained is discussed in relation to the problems of the periodicity of cell functional activity.

[Relationship between the intensity of protein synthesis in fibroblasts and the width of the channels of the cytoplasmic network].

The relationship between the rate of protein and RNA synthesis in fibroblasts and different width of the rough endoplasmic reticulum canals was studied in electron autoradiography experiments by the double labeling method. It was found that protein synthesis in fibroblasts with narrow canals of the rough endoplasmic reticulum proceeds faster than in the cells with wide canals. A possibility of periodic changes in protein synthesis and secretion by fibroblasts is discussed.

[Electron autoradiographic study of RNA synthesis in cerebral cortex neurons in burns].

RNA synthesis in neuronal nucleoli of the rat cerebral cortex was studied by electron microscopic autoradiography in rats before and 1, 12 h, 3 and 6 days after burn injury. It was ascertained that the rate of RNA synthesis begins increasing rapidly after burn injury (1 h) and reaches high level after 12 h. The decrease in the rate of RNA synthesis proceeds more slowly than the increase. This rate does not return to normal even by the close of day 6 following injury.

[Madelung's lipoma (an electron microscopic and autoradiographic study)].

In two cases of Madelung lipoma DNA and RNA synthesis in the material removed at operation was studied by electron-radioautographic techniques. Lipoma adipocytes were found to differ normal ones by incompleteness of the differentiation process. This is manifested by the retention of DNA and RNA synthesis in lipoma adipocytes. A faster rate of proliferation of stromal-vascular cells of lipoma which may be the source of formation of new adipocytes was observed. Intensified DNA and RNA synthesis in patients with Madelung lipoma was observed not only in the cells of the tumor itself but also in the adjacent fatty tissue.

[Electron autoradiographic study of the distribution of 3H-adrenaline in the central nervous system].

3H-Adrenaline was injected into the rat lateral brain ventricle. Half an hour after injection the brain tissue was fixed and the distribution pattern of labeled adrenaline was studied by electron microscopic autoradiography. Silver granules were located mainly above the neuropile. The label content was infrequently different in adjacent synapses. Reasons for dissimilar labeling of adjacent synapses and possibility of using labeled exogenous mediators for studying function of the central nervous system are discussed.

[Sequence of RNA and protein synthesis in wound fibroblasts].

RNA and protein synthesis was studied in the mouse granulation tissue fibroblasts on the 3d and 7th days after wound production. An electron-radioautographic modification of the double marker method, based on the supposition that the two biosynthetic processes investigated occur, mainly, in different parts of the cell, was worked out. Synthesis rates were determined in radioautographs obtained after administration of a mixture of labelled RNA and protein precursors to the animals. There was but a slight difference in density of the label over the nucleus and cytoplasm, nevertheless, in some cells either nucleus or cytoplasm were predominantly labelled and, as the investigation of serial sections demonstrated, this phenomenon was not occasional. Existence of similar fibroblasts should be explained by the suggestion that RNA and protein syntheses increase in the cell not simultaneously but successively.
