[The enzyme-linked immunosorbent assay (ELISA) test in the immunodiagnosis of urinary schistosomiasis. Comparison with the indirect haemagglutination (IH) and counterimmunoelectrophoresis (CIEP) (author's transl)].

Sera from patients with urinary schistosomiasis were used to compare the sensitivity and specificity of the ELISA, IH and CIEP tests for the detection of antibodies to schistosomes. Sera from persons with parasitic infections other than schistosomiasis, and sera from healthy persons were used as negative controls. The ELISA test was more sensitive than the IH and CIEP, and slightly more specific than the CIEP. This simple test may be suitable for use in large seroepidemiological surveys.

Indirect immunoperoxydase test in the serological diagnosis of urinary schistosomiasis.

The outcome of the indirect immunofluorescence test (IIF) and the indirect immunoperoxydase test (IIP) was compared in 60 serum samples of patients affected by haematobium schistosomiasis and in controls. There was a striking agreement (positive correlation in 86.6%). The IIP is a simpler and cheaper test than the IIF.

Fluorescent Antibody Technique↗

[Indirect immunoperoxidase technic in the biological diagnosis of human parasitic disease].

Two serological techniques, immunoperoxidase (IP) and immunofluorescence (IFI), were used for the detection of specific antibodies in 60 sera from patients with schistosomiasis, in 44 sera from patients with hydatidosis, and 28 sera from patients with intestinal amebiasis or liver amebic abscess. The IP test correlated well with IFI. The advantage of IP technique compared to IFI are that it is more rapid, less laborious and is performed without technical equipment.

[Comparative evaluation of three techniques of passive haemoagglutination for the determination of HBsAg (author's transl)].

The AA. studied the reaction of indirect haemoagglutination for the determination of HBsAg in 157 sera from patients with viral hepatitis, with 3 differents commercial tests based on the agglutination by HBsAg, respectively, of sheep, turkey and human erythrocytes sensitized with antibody to HBsAg. All three techniques showed a high sensibility comparable to that of the RIA. Regarding specificity the best results were obtained with the turkey and human erythrocytes, while the sheep erythrocytes because of the presence of some aspecific positivity, need a confirmatory test.

Reinfection of Somali children with Trichuris trichiura after chemotherapy: relevance of immunostimulation.

The intestinal helminth status of an age-stratified sample (6 to 20 years old) from a Somalian community has been assessed and the typical pattern of highly aggregated parasite distribution found. A reinfection study on a sample of 40 children (treated and untreated with a pentapeptide identical to the active site of the thymic hormone thymopoietin) seemed to indicate that immunological factors play a significant role in modulating the population dynamics of infection in endemic communities.

High prevalence of anti-hepatitis delta virus antibody in chronic liver disease in Somalia.

We have assessed the prevalence of hepatitis delta virus (HDV) infection in people with histologically proven chronic liver disease living in Somalia. Among 104 patients studied (14 with chronic persistent hepatitis, 74 with chronic active hepatitis, and 16 with active cirrhosis), 52 were positive for hepatitis B surface antigen; of these, 26 (50%) carried anti-delta antibodies. HDV infection was detected more frequently in sera from hepatitis B e antigen (HBeAg) negative patients (60.9%) than in HBeAg positive patients (9.1%). Using the dot-blot hybridization technique, serum hepatitis B virus deoxyribonucleic acid was revealed in 73.1% of patients without HDV infection, while it was detected in only 7.7% of anti-delta positive patients. It is concluded that HDV is strongly associated with chronic liver disease in Somalia.

[The circulation of the hepatitis A and B viruses in the Somali population].

We report the results of a study carried out to evaluate the extent of hepatitis A virus (HAV) and hepatitis B virus (HBV) circulation in Somalia. Serum samples were collected from 593 subjects (age range 0-83 years) and tested for anti-hepatitis A (HAV) and anti-HAV IgM. Serum samples taken from 1272 individuals (age range 0-83 years) were tested for HBsAg, anti-HBsAg, anti-HBcAg, HBeAg and anti-HBeAg. We confirmed a very high rate of HAV exposure (about 90% of the subjects tested had circulating anti-HAV) as is typical of fecal-orally transmitted infectious agents. The age-specific anti-HAV IgM prevalence suggests that HAV infection is acquired very early in life. Our data also indicate a high rate of HBsAg carriers (range: 10.5%-27.4%) in the Somalian population. When all markers are considered, 60% of the adult population showed evidence of HBV exposure. HBV spreads very subtly: in fact, it is generally transmitted via non-overtly percutaneous routes. In Somalia, hepatitis A virus infection is highly endemic and occurs very early in life. Hepatitis B virus infection is also widespread in this country.

[Serological diagnosis of hydatid disease with indirect immunoperoxidase and immunofluorescence methods (author's transl)].

Indirect immunoperoxidase (IP) method is compared with indirect immunofluorescence (IFI) in 44 cases of active hydatidosis established at operation (30 cysts of the liver, 9 cysts of the lung and 5 extra-hepatic and extra-pulmonary cysts). The sensitivities of the IP and IFI tests were 88.6% and 63.6%, respectively. The IP test is easy, specific and very sensitive.

[Sensitivity and specificity of the RAST (radioallergosorbent test) in biological diagnosis of the hydatidosis (author's transl)].

The radioallergosorbent test (RAST) for specific IgE antibodies to Echinococcus granulosus was compared with other immunological methods in regard to sensitivity, specificity and its use as a diagnostic aid for hydatid disease. Sera used were from patients with active hydatidosis proved surgically, from persons operated for hydatidosis during the previous 2-10 years and from patients with other parasitic diseases or healthy subjects. Results with enzyme-immuno assay (ELISA), indirect haemagglutination (IHA), counter-immunoelectrophoresis (CIEP) and skin test were compared with those with RAST. The percentage of positive RAST reactions (81.2%) among the patients with active hydatidosis was slightly lower than the percentage of positive ELISA (90.6%), IHA (90.6%) and skin test (87.5%) and was superior to CIEP (75%). Among the patients with previous hydatidosis, the RAST was negative in 90% of case, whereas other serological tests were positive in a considerable proportion of cases (ELISA test, 80%; IHA test, 40%; CIEP test, 40%; skin test, 60%). A high percentage (50%) of false positive RAST reactions have been observed in sera from patients infested with other parasites. Results indicated that the RAST for IgE antibodies not be used as the only method for diagnosis of hydatidosis, but it may be employed, if serological data obtained before surgery are available for comparison, for evaluate the results of surgery and to clarify the prognosis.
