Abundance and location of DARPP-32 in striato-tegmental circuits of domestic chicks.

The striatum is reciprocally connected to the brainstem dopaminergic nuclei and receives a strong dopaminergic input. In the present study the spatial relation between the dopaminergic and dopaminoceptive structures of the avian medial striatum (formerly: lobus parolfactorius) was observed by confocal laser scanning microscope in the domestic chick (Gallus domesticus). We also analysed the connections in the area ventralis tegmentalis and the substantia nigra. To label the dopaminergic structures, anti-tyrosine hydroxylase was used and DARPP-32 (dopamine and cAMP regulated phosphoprotein) was a marker of dopaminoceptive elements. The tyrosine hydroxylase positive fibres formed baskets of juxtapositions around the DARPP-32 containing cells of the medial striatum. However, such baskets were also observed to juxtapose DARPP-32 immunonegative cells. In the tegmentum, DARPP-32 was observed in axons descending from the telencephalon via the ansa lenticularis. These varicose fibers innervated the ventral tegmental area and substantia nigra and were often juxtaposed to dopaminergic neurons and dendrites. Approximately 40% of the striatal projection neurons targeting the ventral tegmentum, and 60% of striatal projection neurons targeting the nigra were immunoreactive to DARPP-32, as revealed by retrograde pathway tracing with Fast Blue. Endogenous dopamine may exert a retrograde synaptic effect on the afferent striato-tegmental fibers, apart from the reported extrasynaptic action. The abundance of juxtapositions observed in the avian brainstem and medial striatum corroborates the possibility of reciprocal striato-tegmental circuits, relevant to the reinforcement of behaviour.

Heat-inducible expression of a reporter gene detected by transient assay in zebrafish.

Heat-inducibility of two reporter constructs expressing lacZ gene under the control of mouse and Xenopus hsp70 promoters was tested in zebrafish (Danio rerio) embryos using a transient expression system. Cells expressing beta-galactosidase were stained blue by histochemical staining and their average number per embryo was used as an indicator of the expression level of the reporter gene. Both constructs were heat-inducible in the embryonic tissues and showed similar heat dependence (increasing expression levels from 35-36 degrees C up to 39 degrees C with an apparent decrease at 40 degrees C), resembling that of the zebrafish hsp70 genes. However, their induction kinetics were different, which might be due to differences in their 5' UTRs. Spatial expression patterns of the two hsp/lacZ constructs and an endogenous hsp70 gene were mostly similar on the RNA level. These results indicate that our approach is applicable for in vivo analysis of the heat-shock response and that exogenous heat-shock promoters may be useful for inducible expression of transgenes in fish.

[The place of percutaneous transluminal angioplasty in the treatment of obliterative vascular diseases of the lower limbs. Angioplasty of of the iliac system].

Between 1983-91 386 patients with hemodinamically significant stenosis or short-segment occlusion of iliac arteries were treated by transluminal angioplasty. 487 dilatations and 13 recanalizations were performed. In 353 (70.2%) of these cases were carried out without medicinal preparation continuously after the diagnostic angiography. The technical success rate was 93.4%. The technical success rate did not depend on medicinal preparation. 93% of their patients became symptomless or improved. 28 complications were noted (7 thrombosis, 4 embolisms, 14 inguinal haematomas, 2 false aneurysms, 1 bleeding), 11 of them required surgical intervention. One patient died because of cardiac failure due to retroperitoneal haematoma. Authors' opinion isolated short uni- and bilateral iliacal stenosis can be treated by transluminal angioplasty. Longer or multifocal stenosis required individual consideration.

[Successful intraoperative angioplasty in bilateral fibromuscular dysplasia of the internal carotid arteries].

The authors report the clinical history of a 55-years old woman suffering from the multiplex stenosis of both internal carotid arteries caused by fibromuscular dysplasia. Successful intraoperative angioplasty was performed. In connection with their case the authors review briefly the clinical feature, diagnosis and therapy of fibromuscular dysplasia of the internal carotid artery.

[Characteristics of nosocomial pneumonia].

284 patients with pneumonia treated in non-invasive chest wards were analysed in a prospective study. Out of 284 cases 26 proved to be nosocomial pneumonias. Older age, chronic non specific lung diseases, malignancies, peptic ulcus, chronic alcoholism as risk factors for nosocomial pneumonia should be taken into consideration. The pathogen agent was clearly identified in four cases, meanwhile the pathogenity of the agents was suspected at seven patients. By distribution of all pathogens the widely known bacteria are responsible for nosocomial pneumonia were less than one third of cases (in three cases Staphylococcus aureus, in one-one case E. coli and Enterobacter). The possible causes of their observation were analysed. Out of the 15 pathogens Gram-negative bacilli were found in six cases only. Authors suggest to use this observation at the rational choice of empiric treatment.

[Abscessing pneumonia].

Among the patient of a prospective study aimed to recognize the epidemiological and clinical characteristics of pneumonias, cavitating pneumonia occurred in 12.5%, typical lung abscess in 24 cases and necrotizing pneumonia in 11 cases. The illness was secondary in 4 cases. Predisposing factors were present in the majority of the patients. Assumable pathogen was detected with microbiological examinations in 24 cases. 20 patient recovered--19 by antibiotic therapy and one by surgery--were operated on the average in 7 weeks. Complication occurred in 15 patients 7 of them died. The prognosis of cavitating pneumonias is favourable, however the mortality is high in the secondary cases.

Use of a new erythromycin product--eryc--in the treatment of the lower respiratory tract infections.

Eryc therapy was used in 27 patients admitted with acute exacerbation of chronic obstructive bronchitis, and in 4 patients with pneumonia. Depending on the severity of the condition the dose of the drug was 4 x 500 mg or, in a lower number of cases, 4 x 250 mg. The effectivity of therapy was evaluated on the basis of the changes in the quality of sputum, elimination of facultative pathogens identified from sputum at admittance, clinical symptoms, respiratory function values, blood gas values, and, in case of pneumonia, improvement of chest X-ray finding. Eryc therapy proved to be effective in 78.6% of the cases. Side-effects were observed in 25.8% of the cases but the therapy had to be discontinued because of the severity of unwanted effects only in 3 patients (9.7%). The change of the method of drug administration-intake during meals-did not decrease the frequency of side-effects when compared to Hungarian data. On the basis of our results Eryc therapy was found to be useful for the treatment of the lower respiratory tract, community-acquired infections.
