Local and systemic tolerance of flutrimazole skin creams following single and repeated topical application in healthy volunteers.

A double-blind, randomized phase I study was performed in 21 healthy volunteers to evaluate the dermal tolerance of skin creams containing 1% and 2% 1-[(2-fluorophenyl)(4-fluorophenyl)phenylmethyl]-1H-imidazole (flutrimazole, UR-4056, CAS 119006-77-8) or only the excipient, versus a commercial skin cream containing 1% clotrimazole. The study was carried out using the patch-test procedure performed in three stages: 1. single application in the back skin; 2. induction period (usage test) using three skin areas on the volar side of the forearm of each subject, where skin cream samples were applied once a day for a period of three weeks; and 3. after a wash-out period of two weeks, challenge applications in the back and forearm skin. The systemic tolerance of the formulations was also tested. There was no evidence of allergic sensitization after the application of flutrimazole creams, or their excipients, with signs of mild and doubtful skin reactions being observed in few subjects with all formulations. Furthermore, no systemic side effects after topical administration were detected throughout the study.

The use of levamisole in atopic dermatitis: a prospective study.

Levamisole hydrochloride, a nonspecific immunostimulant, was given to 30 children with atopic dermatitis (AD) in a double-blind manner during an eight-month period. Clinical course findings, including intradermal reactions to tuberculin, candidin, and streptokinase-streptodernase; sensitization to dinitrochlorobenzene; periphal blood T- and B-lymphocytes; in vitro lymphocyte transformation with phytohemagglutinin; and serum levels of IgM, IgG, IgA, and IgE, were noted every two months. Fifteen children completed the study. Neither the clinical course nor the immunologic values showed substantial differences between the levamisole-treated and the placebo group. Our experience with levamisole in AD is disappointing.

[Therapy with transfer factor in a case of recidivating herpes with polymorphous erythema].

A young woman had recidivating herpes with erythema multiforme for several years. When conventional therapy failed to correct this condition she was treated with Transfer Factor (TF). Immunologic anomalies which had been registered previously became normal after the first application of TF. The skin lesions disappeared after the fourth dose. Six months later a new skin manifestation with immunologic alterations became apparent. They disappeared after two doses of TF. A year and a half later the patient has no lesions and the immunologic parameters, which were monitored throughout this period, are normal.

[Chronic urticaria and serum iron].

During the year 1974, 111 patients with urticaria were studied in the Department of Dermatology, of the Saint Paul's Hospital (Autonomous University of Barcelona). Among those, 77 had chronic urticaria, of which 22 a had low blood iron values. After iron treatment the urticaria improved or was cured in some of these patients. The remaining cases in which the cause of their hives was unknown were followed for one year. Some systemic causatives for their urticaria may appear on longer follow-up. At the same time, blood levels of salicylates trypsin and alpha-chymotrypsin were checked in 14 patients, with the object of a possible relationship concerning the persistence of the urticaria.
