Differential frequency of human X and Y chromatin as related to cell density in vitro.

The relationship between cell density in vitro and the frequency of fluorescent X chromatin and/or Y chromatin-positive interphase nuclei was studied in cultured cells from three human male and female subjects. It was found that altough the frequency of fluorescent X chromatin-positive cells was proportional to cell density, the frequency of fluorescent Y chromatin-postitive cells was independent of cell density. Our results are compared with those of other investigators.

Cell cycle analysis and X-chromosome inactivation in the developing mouse.

The duration of different phases of the cell cycle (G1, S, G2, and M), as well as the exact time of initiation of asynchronous DNA replication pattern in one of the X-chromosomes in the female mouse during the early stages of development, have been investigated. It was found that early developmental stages in the mouse are devoid of G1 period, and from the late blastocyst stage the embryonic cells first acquire all the characteristic stages of the cell cycle (G1, S, G2, and M). Additionally, asynchronous DNA replication pattern in one of the X-chromosomes in the female embryos was found to be initiated with the appearance of the G1 period. I have established a correlation between the onset of the G1 period and the initiation of an asynchronous DNA replication pattern in one of the X-chromosomes of the female mammal during embryogenesis.

Fluroescent characteristics of drumsticks, drumstick-like projections, and other nuclear bodies in human blood cells.

The fluorescent properties of drumsticks, drumstick-like appendages, and other nuclear bodies in the polymorphonuclear leukocytes from six human males and females were studied with the aid of the quinacrine-mustard staining technique. Both brightly and weakly fluorescent drumsticks (in females) and drumstick-like bodies (in males) were observed, and they were readily differentiated on the basis of size, shape and, usually, fluorescent intensity. An analysis of the correlation between the extent of nuclear lobulation of the polymorphs and the corresponding fluorescent patterns of the adjoining drumsticks and drumstick-like bodies indicated that a possible change in the state and/or condensation of chromatin in these nuclear bodies might occur with increasing age of the polymorphs. Although the brightly fluorescent regions of the nuclei usually corresponded to the areas darkly stained with Giemsa, much finer patterns of differential staining of drumsticks and other nuclear bodies were obtained only by the fluorescent method.

Inhibition of RNA synthesis by acetyl salicylate and actinomycin D during early development in the mouse.

Experiments were designed to ascertain and compare the effects of acetyl salicylate and actinomycin D on RNA synthesis in mouse oocytes in vitro and in vivo. After exposure to the drugs the effects on RNA synthesis were measured by incorporation of [3H]uridine and autoradiography. The results indicate that acetyl salicylate inhibits RNA synthesis in the treated oocytes as does actinomycin D. The only difference in the effects of these two drugs is that salicylate inhibits RNA synthesis to a much lesser degree than does actinomycin D. Effects from a short exposure to salicylate may be reversible; the same effects with actinomycin D cannot be reversed. In utero exposure of the female fetus may lead to partial or total sterility (depending on the dose and time of exposure) of that fetus and/or abnormal development of the progeny from those mice (F2). These results suggest that RNA synthesis in early oogenesis is a vital part of later development of the oocytes in adult mouse ovary. Inhibition of RNA may be one of the causes of malformations and sterility.
