Oxidative stress markers and antioxidant levels in normal pregnancy and pre-eclampsia.

OBJECTIVE: To compare the levels of 3 oxidative stress markers (glutathione peroxidase [GPX], superoxide dismutase [SOD], and malondialdehyde [MDA]) and 2 antioxidants (vitamin C and lycopene) in healthy and pre-eclamptic pregnant women. METHODS: Circulating levels of GPX, SOD, MDA, vitamin C and lycopene were measured in 50 healthy pregnant women and 50 women with pre-eclampsia (PE) (41 with mild PE and 9 with severe PE) attending the antenatal clinic or admitted to the maternity ward of the All-India Institute of Medical Sciences, New Delhi, India. RESULTS: The levels of GPX, SOD and MDA were significantly higher in women with PE than in controls, and the increase was higher in women with severe PE (P<0.001 using analysis of variance and the Kruskal Wallis test). The levels of vitamin C and lycopene were significantly lower in women with PE than in controls, with a greater decrease in women with severe PE. CONCLUSION: Increased levels of oxidative stress markers and decreased levels of antioxidants in pre-eclamptic women suggest that oxidative stress markers play a significant role in the pathophysiology of pre-eclampsia, and that supplemental dietary antioxidants may have a beneficial role in the prevention of pre-eclampsia in women at high-risk for this condition.

Stability and kinetic behaviour of some enzymes in surfactant environment.

The interaction of alpha-chymotrypsin, invertase, alcohol dehydrogenase and alkaline phosphatase with some ionic and non-ionic surfactants, viz. sodium dodecyl sulphate, dioctyl sodium sulphosuccinate, hexadecyltrimethylammonium bromide, tetradecyltrimethylammonium bromide and Triton X-100, has been examined by studying the effect of varying surfactant concentrations on enzyme activities as well as by determining the time-dependent inactivation and the time-independent inhibition. The kinetic parameters, Km and Vmax, for alpha-chymotrypsin-catalysed reaction in presence of sodium dodecyl sulphate were evaluated. Anionic surfactants markedly decreased enzyme activity, whereas cationic surfactants were less effective. Nonionics showed no effect. This change in enzyme activity was also dependent on the nature of enzyme.

Induction of cell mediated immunity in vitro and in vivo after cis-platin treatment.

Effect of cis-dichlorodiammine platinum II (cis-platin) on Dalton's lymphoma in syngeneic (DBA) and allogeneic (Swiss white mice) system has been analyzed. Cytotoxic spleen cells from normal and tumor bearing mice (already treated with cis-platin at its full fledged state) were mixed in different ratios with the tumor cells (the same number of cells) to induce immunization in healthy mice. Capillary cell migration inhibition assay was also performed to study the development of immunity after chemotherapy. The immunization of mice with the platinum complex results in specific enhancement of the host cellular immune system.

Chemoimmunotherapeutical studies on Dalton's lymphoma with cis-platin.

Effect of cis-platin (cis-dichlorodiamine platinum II) on Dalton's lymphoma in syngeneic DBA and allogeneic Swiss white mice was studied. Normal healthy mice were immunized with cis-platin incubated cells and challenged to check the immunity developed. In vitro assay of cell mediated immunity was investigated by the capillary cell migration inhibition of reticuloendothelial cells from both DBA and Swiss white (treated as well as normal) mice against the specific tumor antigen. Cis-platin treatment results in specific enhancement of host cellular immune system.

Enhanced cell mediated immunity in mice after cisplatin treatment.

Tumor regression in mice after cisplatin treatment and the role of cell mediated immunity were studied through Winn assay (in vivo) and capillary cell migration inhibition assay (in vitro). The immunization of mice with the platinum complex results in specific enhancement of host cellular immune system by stimulating splenocytes and peritoneal exudate cells against the specific tumor antigens.
