Toxic effects of lemon peel constituents on Ceratitis capitata.

A series of experiments were conducted to evaluate the toxicity of lemon peel extracts incorporated into mediterranean fruit fly Ceratitis capitata diet. Extracts were obtained with different solvents: diethyl ether, ethyl acetate, and methanol. All three extracts were toxic to some extent; the diethyl ether extract was selected for further studies. Ether extracts of lemon peel were prepared weekly over a 2-month period, from fruits collected on the 1st d of the bioassay. Weekly GC-MS and UV analyses of the extracts demonstrated that the concentration of citral and coumarins decreased in the peel after harvest. We conducted a series of bioassays to evaluate the toxicity of the ether extract, and mixtures of this extract with citral, 5,7-dimethoxycoumarin. and linalool incorporated to C. capitata larvae's natural diet (lemon slices endocarp) at a concentration of 250 microg/g of diet. Significant larvicidal activity can be obtained from a fresh lemon peel extract; however, when the extract was obtained from stored lemons, toxicity decreased. Addition of small amounts of citral or 5,7-dimethoxycoumarin, and linalool to the stored lemon peel extract would bring back the toxicity to the rates of fresh lemons extracts. Finally, female adults of C. capitata fed on diets containing additional amounts of ether extract, 5,7-dimethoxycoumarin, and linalool, were exposed to different photoperiods to test for phototoxicity. The treatment was toxic and affected the oviposition capacity of females depending on photoperiod.

Melampolides from Enydra anagallis.

The investigation of an Argentine collection of Enydra anagallis afforded sesquiterpene lactones of the melampolide type two of which were previously known. Their structures were elucidated by spectroscopic methods.

Sesquiterpene lactones and other constituents of Disynaphia multicrenulata from Argentina.

Flowers and leaves of Disynaphia multicrenulata from Argentina afforded a large number of known germacradienolides and heliangolides, a new germacradienolide, a known melampolide, a new parthenolide derivative, one known and seven new cronquistiolides, a new 4Z-melampolide, three known and five new eudesmanolides, a new elemadienolide, a known grazielolide, an isoguaiagrazielolide, two diepoxygermacran-8,12-olides, three common kauranoic acids, pinoresinol, jaceosidin and the sesquiterpene oplopanone. Structures were elucidated by high-field 1H NMR spectrometry. The relationship to the known chemistry of other members of the Disynaphiinae is discussed.

New coumarins from Pterocaulon polystachyum.

Aerial parts of Pterocaulon polystachyum from Chaco province, Argentina, afforded the known coumarins ayapin, isoscopoletin, prenyletin, prenyletin methyl ether, virgatenol, obtusinin, 5-methoxy-6,7-methylenedioxycoumarin, 5-(3,3-dimethylallyloxy)-6,7-methylenedioxycoumarin, 5-(2',3'-dihydroxy-3-methylbutanoxy)-6,7-methylenedioxycoumarin, haplopinol methyl ether, 6-(1,1-dimethyl-2-propenyl)-7-hydroxycoumarin and demethylnieshoutin; the last two are new as natural products while the five new coumarins from the collection were isovirgatenol, 3'-deoxyobtusinin, 6-methoxy-7-(2'-hydroxyethoxy)-coumarin, 5-(2'-hydroxyethoxy)-6,7-methylenedioxycoumarin and a substance tentatively identified as 5-hydroxy-6,7-methylenedioxy-8-(3,3-dimethylallyl)-coumarin.

Allelopathic agents from Cyrtocymura cincta.

The sesquiterpenoid fractions from the chloroform extracts of roots and aerial parts of the weed Cyrtocymura cincta (Griseb.) H. Robinson (Compositae) were tested to observe the effects on seed germination and growth of three dicot and four monocot species. Promotion and inhibition effects were observed according to the examined compound, specific seed or cultivar, and concentration.

Germacranolides and a new type of guaianolide from Acanthospermum hispidum.

The aerial parts of an Argentinian collection of Acanthospermum hispidum afforded 26 sesquiterpene lactones, including the two guaianolides (1 and 2) having a novel oxygen bridge between C-4 and C-14, three new cis,cis-germacranolides (4, 7, and 8), and two new melampolides (25 and 26). Guaianolides 1 and 2 seem to derive biosynthetically from the germacranolide 27 having the (1)D(14, 15)D(5) conformation. The structures were elucidated using extensive spectroscopic analysis.

Biochemical changes and endocrine responses in cystic fibrosis in relation to incremental maximal exhaustive exercise.

The purpose of this study was to compare biochemical changes and endocrine responses during an incremental maximal bicycle test in three well-trained 18-year-old patients with cystic fibrosis (CF) and in three healthy controls. Although the blood concentration at rest of the white cell count, haptoglobin, phosphorus, urea, creatinine, and uric acid were somewhat different in the two groups, the CF patients had similar biochemical changes in response to the exercise compared with the healthy men. The endocrine responses to exercise seemed to be different between the two groups with regard to changes in cortisol, growth hormone, and testosterone concentrations. The differences, however, were probably caused by differences in age, training situation, and psychological stress reaction rather than by pathological mechanisms.

Biochemical changes and endocrine responses in cystic fibrosis in relation to a marathon race.

Biochemical changes and endocrine responses during the New York Marathon (42195 m) were investigated in three 18-year-old male adolescents with cystic fibrosis (CF) and three healthy men who accompanied the CFs during the race. The ambient temperature was 20 degrees-28 degrees C and the relative humidity 98%-75% during the run. The CF patients, who had Shwachman scores of 60, 85 and 95 completed the run without major problems in 6.10, 4.42, and 4.32 h, respectively. Serum concentrations of Na and Cl decreased slightly, but the values were still within normal range. Na and Cl excretions in the urine decreased to very low levels in the CF adolescents during the run. All the other biochemical changes were similar to the changes in the controls. Aldosterone concentration increased to a higher level and maintained this increase for a longer time after the race in the CFs. Testosterone concentration decreased more in the CFs during the race compared with the controls. Growth hormone concentration showed individually varying changes in response to the exercise. This study demonstrates that patients with CF may participate in strenuous prolonged exercise even in humid and hot conditions, without untoward effects. The observed differences in hormonal responses to exercise might be explained by differences in age, training status, and relative exercise intensity rather than by hormonal or other disturbances in CF.

Fatty acids in cystic fibrosis in response to a marathon race.

Lipid metabolism was studied in three cystic fibrosis (CF) patients (18 years old) and three healthy controls (35-40 years old) who completed the New York Marathon 1984. Lipase was not detectable in plasma from two of the CF patients, but in one CF patient with a functioning pancreas and in the three controls it increased during and after the run. Cholesterol concentrations were lower in all patients than in the controls. The HDL/total cholesterol ratios increased in all runners throughout the observation period, substantially more in the patients than in the controls. Triglycerides and glycerol values were similar in the two groups. The relative distribution of fatty acids in total plasma, plasma free fatty acid fractions, and total red blood cell membranes, at rest, were similar to those earlier reported both for the CF patients with pancreatic insufficiency and for the controls. Lower relative concentrations of linoleic acid (C18:2n6) and higher relative concentrations of C14:0, C16:0, C16:1, C18:3n6, and C20:3n6 were found in the CF specimens. During and after the race, linoleic acid concentration increased and the other fatty acids decreased, giving a more normalized pattern in the patients. The fatty acid pattern of the CF patient with a functioning pancreas behaved more like the healthy controls. The fatty acid pattern of the red cell membrane did not change during or after the run. Although the observations are few, it is encouraging that both the HDL/total cholesterol ratio and the fatty acid pattern improve in response to prolonged aerobic exercise.

Dopamine and catecholamines in cystic fibrosis in response to a marathon race.

The excretions of 3,4-dihydroxyphenylalanine (DOPA) metabolites and 5-hydroxyindoleacetic acid (5-HIAA) were followed before, during, and after a marathon race in three patients with cystic fibrosis (CF) and three healthy controls. Moderately increased DOPA excretions in CFs compared with controls before, during, and after the race support the idea that there is an altered metabolism of DOPA in CF. Noradrenaline, adrenaline, homovanillic acid, and vanilmandelic acid were increased in CFs compared with controls mainly during and after the race, which might be a reflection of the heavier working intensity that the CF patients had in comparison with their healthy controls. 5-HIAA, the breakdown product of serotonin, was higher in CFs in comparison with controls before and during the run, but equal thereafter. This reflects an ameliorated serotonin metabolism after the run.

Cystic fibrosis-like changes in saliva of healthy persons subjected to anaerobic exercise.

The biochemical composition of saliva secreted by healthy persons and by heterozygotes and homozygotes of cystic fibrosis at rest and by healthy persons subjected to aerobic or anaerobic effort were compared. In the saliva from cystic fibrosis homozygotes at rest substantial increases of the activity of ribonuclease (p less than 0.001) and of the concentrations of protein (p less than 0.001), lactate (p less than 0.001), sodium (p less than 0.001), potassium (p less than 0.01) and calcium (p less than 0.05) were found in comparison with saliva from healthy persons at rest. In the saliva from cystic fibrosis heterozygotes at rest similar but less pronounced changes were seen. After anaerobic exercise these biochemical parameters were increased in the saliva of healthy persons and mimicked the values of cystic fibrosis saliva. However, after aerobic effort no changes other than a slightly increased ribonuclease activity were seen in the saliva of healthy persons. This indicates that salivary glands of cystic fibrosis patients, at rest, are in the same state of lactate acidosis and energy depletion as these glands are in healthy persons after anaerobic work.
