Long lasting IgG subclass and antibacterial polysaccharide antibody deficiency after allogeneic bone marrow transplantation.

Serum IgG subclasses were measured by a competitive indirect immunoassay with monoclonal antibodies in 31 leukemic patients before and after bone marrow transplantation. Antibodies to Hemophilus influenzae type b (Hib) capsular polysaccharide were determined in 28 cases. Abnormally low or borderline subclass (mostly IgG2 and IgG4) levels were found late after transplant in 23 infected and noninfected patients. These levels persisted for as long as 25 months, in association with low or borderline IgA levels in 78% of the cases. IgG2, IgG4, and IgA often showed a parallel evolution, whereas IgG1, IgG3, and IgM often varied together in the opposite way. Class but not subclass deficiencies were more frequent in patients with graft-v-host disease (GVHD). Subclass abnormalities predominated in infected patients, with mean levels correlating with the severity of infections; however, the abnormalities are not clearly predictive of infections in individual cases. Most patients with Hib pneumonia showed virtually no IgG antibody response to Hib, and one-half of the patients had a moderate IgM and IgA response. In the whole series, many sera collected greater than 1 year after graft contained very low or undetectable antibodies. Correlation between anti-Hib antibody and IgG2 levels was significant but weak because of discrepancies that were only partially explained by the subclass distribution of the antibodies.

Isotypic distribution of human anti-thyroglobulin IgG antibodies: methodological difficulties.

The subclass distribution of human anti-thyroglobulin IgG antibodies (TgAb) was studied in 24 sera using an indirect enzyme-linked immunosorbent assay (ELISA) with monoclonal anti-subclass antibodies. An IgG4/IgG1 predominance was found. TgAb were purified from seven sera by affinity chromatography. The antigen-specific indirect ELISA was repeated in the purified TgAb, in which IgG subclasses were measured by a competitive assay with monoclonal antibodies. The latter method showed an IgG2 predominance. Comparison of the results obtained by the three approaches showed discrepancies that probably reflect bias introduced by both the indirect ELISA and the affinity chromatography.

Systemic and ocular effects of alpha 2-adrenergic stimulating and beta 2-blocking agents.

The effects of alpha 2-adrenergic stimulating agents (clonidine and guanabenz) and beta-adrenolytic agents (atenolol and propranolol) on the systemic arterial pressure, heart-rate, blood glucose level and intraocular pressure, as well as on the concentrations of glucose, Na+, K+ and Ca++ in the lens and aqueous and vitreous humours, are evaluated in the rabbit. alpha 2-adrenergic stimulating agents cause a significant increase of the glucose concentration in the blood, lens, aqueous and vitreous humours; whereas the beta-adrenolytic agents cause insignificant effects. Neither of the two drugs alters the concentration of Na+, K+ and Ca++ in the lens and aqueous and vitreous humours.

A truncated form of teratocarcinoma-derived growth factor-1 (cripto-1) mRNA expressed in human colon carcinoma cell lines and tumors.

The human teratocarcinoma-derived growth factor (TDGF)-1 gene encodes a 188-amino acid protein, cripto-1. The TDGF-1 gene is overexpressed in the majority of human primary colorectal carcinomas and hepatic metastases, in breast carcinomas and in testicular nonseminoma germ cell embryonal carcinomas. In the human embryonal carcinoma cell line NTERA-2 clone D1, a 2-kb TDGF-1 mRNA transcript is expressed. The present study shows that a 1.7-kb mRNA transcript lacking the first two exons of the TDGF-1 gene is expressed in the human colon carcinoma cell line GEO. This shorter mRNA is the only TDGF-1 transcript that is present in the majority of primary human colorectal carcinomas and hepatic metastases and in adult human tissues such as the pancreas, heart, stomach, mammary gland, skeletal muscle, liver and placenta. In contrast, in the kidney, brain, testis, ovary and spleen, the longer 2-kb TDGF-1 mRNA transcript is expressed. The putative shorter protein starts at a CUG codon 129 nucleotides downstream of the starting AUG codon of the longer protein. These data indicate the potential for differential transcriptional regulation of the TDGF-1 gene in different normal and tumor tissues.
