[Fenofibrate: chemical development and differences with clofibrate (author's transl)].

Since its discovery by Thorp and Waring in 1962, the clofibrate molecule has undergone various modifications. The all-important phenoxy-2-methyl-2-propionic acid chain was preserved, but it seemed reasonable to try and improve pharmacological activity by substituting for the Cl atom a group with different hydrophobic profile. However, none of the phenylketone derivatives produced had satisfactory antilipaemic properties, except for the benzoyl derivative. Grafting on this derivative a Cl atom in position 4 by an original method based on a vinylogy principle, and esterifying with propanol-2 yielded the most active and best tolerated compound: fenofibrate. Fenofibrate resembles clofibrate in that it has the same phenoxy-2-methyl-2-propionic chain but differs from it by the way electrons are distributed, by its lipophilia and by its supple structure, the mobile carbon links between the two rings allowing for spatial changes and facilitating adaptation to receptors. The strong individuality of the molecule is undoubtedly responsible for the significant pharmacokinetic and pharmacological differences between fenofibrate and clofibrate.

Secondary products of itanoxone.

Itanoxone synthesis by Friedel-Crafts reaction between itaconic anhydride and 2-chlorobiphenyl was studied. Five isomers corresponding to possible impurities were prepared and studied to perfect a reliable and practical method to detect these impurities in itanoxone.

[Application of Verloop parameters. Comparison with other steric parameters and problems of choice of parameter].

Verloop's steric parameters. L, B1, B2, B3, B4, were tested in five groups of molecules, generally successfully, These were compared with classical steric parameters: indices of molecular connectivity, Taft's Es parameter, Van der Waals radii and molecular refraction. Some rules have been developed to decide which parameters to use for various types of substituent.

GABA transaminase inhibitors.

In summary, several branched-chain fatty acids appeared to be competitive inhibitors of GABA-T and non-competitive inhibitors of SSADH. These compounds produce an increase in brain GABA level, and for two of these it was shown that the increase differs among various brain areas. An increase of GABA cannot be obtained by inhibition of SSADH. The increase in brain GABA seems to correlate with the anticonvulsant activity of branched-chain fatty acids.

[Qualitative and quantitative determination of amino-2 benzimidazole in aqueous medium by polarography (author's transl)].

Amino-2 Benzimidazole, a residue of the decomposition of some fungicides (Benomyl, Carbendazine, Thiophanate, Methylthiophanate), is measured by polarography in aqueous medium, with a platinium rotating electrode, for the exploration in potentials from +0.5 v to 1.4 v. The best conditions for the quantitative determination are investigated, the study according to pH is performed and the standard curve is given for concentrations from 10(-5) to 10(-3) M/l. The interaction of elementts found in tap water and in aquarium water is also investigated.
