Use of monoclonal antibodies to follow the phylogenic distribution of human renal antigens.

Nine human differentiation antigens have been defined by monoclonal antibodies (M. Abs) developed from mice immunized with embryonic kidney cells (mesonephros or metanephros of 7 week-developmental ages). Their spatial and temporal distributions during human kidney organization were previously studied [3]. In this paper we have attempted to follow by immunofluorescence their phylogenic location, from fish to mammals. Six of them recognized the same structures as in humans: proximal convoluted tubules (PCT) (EG9.11, EG19.6, E116.1), glomerular basement membrane (GBM) (EG14.1) and extracellular matrix (EK8.1, EK17.1). However, staining was limited to certain mammals. EK17.1 has been characterized as an anti-fibronectin. These antibodies revealed the same histological structures in the human mesonephros and metanephros. The three other antibodies revealed epitopes appearing earlier in evolution and whose histological distribution varied according to species. These antibodies stained different structures in the mesonephros and metanephros. Thus, the staining particularities observed during human renal ontogenesis were found again in the phylogenetical study.

17 beta-Hydroxysteroid dehydrogenase activity in human breast epithelial cell and fibroblast cultures.

Primary cultures of human breast cells prepared from surgical specimens of reduction mammoplasty were used to study the activity of the enzyme 17 beta-hydroxysteroid dehydrogenase (E2DH) which converts estradiol (E2) into its less active metabolite estrone. This study was performed in both epithelial and stromal cells separated, after collagenase digestion of the tissue, on a Percoll gradient, and then cultured as monolayers in Ham's F 10 medium supplemented differently for epithelial cells and fibroblasts. E2DH activity was strikingly higher in epithelial cells than in fibroblasts, since after [3H]E2 incubation (2 nM), 600 fmol/micrograms DNA were metabolized to estrone in epithelial cells after 1 h, whereas an equivalent amount was hardly obtained in fibroblast cultures after 24 h. The affinity and capacity of E2DH were greater in epithelial cells with apparent Michaelis-Menten constant (Km) = 0.6 +/- 0.1 microM and maximum velocity (Vmax) = 250 to 360 pmol/micrograms DNA/h, whereas they were 10 +/- 1 microM and 50 to 70 pmol/micrograms DNA/h, respectively, in fibroblast cultures. Moreover, the E2DH activity was 2 to 5 times higher in epithelial cells cultured in the presence of the progestin medroxyprogesterone acetate, whereas it remained unchanged in fibroblasts cultured under the same conditions. This increase in E2DH activity was dose dependent from 10(-10) to 10(-7) M medroxyprogesterone acetate and inhibited by both actinomycin D and cycloheximide. This system of differential breast cell culture appears to be a fruitful tool for the study of the hormone dependence of normal breast growth and differentiation. Due to the presence of E2DH, epithelial cells are more apt to undergo and to moderate E2 action. Moreover, epithelial cells are a possible site of progesterone modulation of E2DH activity. Therefore, E2DH could be a good marker both for epithelial cells and their hormone dependence.

[Prospective study of cytomegalovirus infections in 211 children with kidney transplants].

A prospective serologic study was conducted to determine the incidence of primary Cytomegalovirus (CMV) infection after a renal transplant and to evaluate the consequences on the graft. Serologic tests using the indirect hemagglutination technique with CMV-sensitized erythrocytes were performed serially over 6 years in 370 children initially awaiting renal transplantation. 211 children received a graft during the study period. Transplantation was followed by primary CMV infection in 48 (35.3%) of the 136 children with negative serologic tests at the time of the procedure. The percentage of graft failures is identical in the 75 children with positive tests before transplantation and in the 88 children with negative tests before and after transplantation (24 and 23% respectively). Conversely, only 6 (12.5%) graft failures were recorded among the 48 children who experienced primary CMV infection after transplantation. It can be speculated that these results are due to the immunosuppressive effect of CMV infection.

Prenatal diagnosis of congenital enzymopenic methaemoglobinaemia with mental retardation due to generalized cytochrome b5 reductase deficiency: first report of two cases.

Prenatal diagnosis of congential enzymopenic methaemoglobinaemia (CEM) with mental retardation was performed in two fetuses at risk for generalized NADH-cytochrome b5 reductase deficiency. In the first case the enzyme activity of cultured amniotic cells was in the heterozygous to normal range. The mother delivered a normal baby with normal enzyme activity in cord blood cells. In the second case, the amniotic cells were almost completely enzyme deficient. The pregnancy was terminated, and the diagnosis of homozygous NADH-cytochrome b5 reductase deficiency was confirmed in cord blood cells, in several different tissues and in cultured fibroblasts from the aborted fetus.

Prenatal diagnosis of congenital adrenal hyperplasia (21-OH deficiency type) by HLA typing.

The close genetic linkage between HLA-B and congential adrenal hyperplasia due to 21-hydroxylase deficiency permits prenatal diagnosis of an affected fetus by HLA typing of amniotic fluid cells in pregnancies at risk. Some families at risk, especially those with an affected girl with ambiguous genitalia, will only plan another pregnancy if a prenatal diagnosis is possible. After HLA typing of the index case, parents and eventually grandparents, the family were informed of the possibility of a prenatal diagnosis. Fibroblast cell lines were initiated from skin biopsies of the index cases and parents and were used as controls in the tests. HLA typing of the fetus was done on amniotic fluid cells grown in vitro using first, a microcytotoxicity test and second quantitative microabsorption test. Ten prenatal diagnoses are reported. In two cases the HLA genotype indicated an affected fetus, examination of the aborted fetuses was in agreement with the diagnosis. In one case an affected male fetus was diagnosed, the pregnancy is in progress. In seven cases an unaffected infant was predicted (four carriers and three homozygous normal infants).

Determination of type 1 poliovirus subtype classes with neutralizing monoclonal antibodies.

Hybridomas secreting neutralizing monoclonal antibodies (MA) against type 1 poliovirus were developed by fusion of mouse myeloma cells with splenocytes of immunized mice. Strain-specific MA selected for their capacity to neutralize wild (Mahoney) or attenuated (Sabin, LSc 2ab) poliovirus strains were used for identification of field isolates. A simple and reliable neutralization-index-assay was developed and used to test for intratypic serodifferentiation of poliovirus with the selected MA. Three subtype classes of type 1 poliovirus have so far been identified by testing 4 reference strains and 96 field isolates: Mahoney-like, LSc-like, and strains not recognized by either anti-Mahoney or anti-LSc MA. Neutralizing MA proved an excellent tool for unequivocal antigenic characterization of different poliovirus strains.

[Trophoblastic gestational diseases. Triploid syndrome, perivillous trophoblastic hyperplasia, trophoblastic pseudotumor, trophoblastic microcarcinoma and carcinoma].

Comparing the cytogenetic and morphological data on cases of hyperplasia and neoplasia of the trophoblast allows some conditions to be better defined and separated. The term partial mole or embryonic mole should be replaced by the term triploid syndrome because of the especially strong correlation between the triploid caryotype and the special aspect of the placenta. Triploidy is usually the result of fecundation of a haploid ovule by two spermatozoids, probably due to the similarity of the haploids of the two parents, as is suggested by the study of HLA antigens. Classical hydatidiform mole, or perivillous trophoblastic hyperplasia, is usually discovered earlier, during an ultrasound examination or abortion and its histological diagnosis is easy. The caryotype of complete hydatidiform mole is diploid, nearly always XX, with the two sex chromosomes coming from the father (the maternal sex chromosome being eliminated). The caryotype of vesicular dystrophia without trophoblastic hyperplasia is, on the other hand, normal. Trophoblastic microcarcinomas have a better known presentation. And trophoblastic carcinomas are distinct from non-trophoblastic neoplasms which secrete HCG and from benign trophoblastic pseudo-tumors which are often non-secretory.

The repair of DNA single strand breaks in human cells with genetical disorders.

The aim of this work was to make a preliminary examination of some hereditary diseases, related to precocious aging in order to determine whether the defects involved resulted from an impairment of the DNA repair capacity. To test for an impairment in DNA single strand capacity, in cells from patients with different diseases, we have followed the rate of the DNA after X-irradiation by sedimentation on alkaline sucrose gradients. It was found that all cells observed from Lesh Nyhan, cystic fibrosis, trisomy 15 et 21, retinoblastoma, were able to repair the single strand breaks in their DNA to the same extent and at the same initial rate.

[Sero-epidemiology of infections due to cytomegalovirus by indirect hemagglutination technic].

Applied to cytomegalovirus, the technique of indirect hemagglutination offers a good sensitivity and reliable specificity for serological testings. Improvements in the standardization are however still required. Sero-epidemiological studies performed in different groups of children and carried out with this method led to the following results: mother-child contagium, early infection in young infants, and influence of socio-economical and ethnical factors. Studies performed on pregnant women showed that the risk of infection decreased after the first pregnancy.
