Compartment-specific regulation of phosphoinositide 3-kinase by platelet-derived growth factor and insulin in 3T3-L1 adipocytes.

To understand how the stimulation of phosphoinositide 3-kinase (PI 3-kinase) by different growth factors can activate different subsets of downstream responses, growth-factor regulation of PI 3-kinase activity at different intracellular locations was investigated in 3T3-L1 adipocytes. Insulin caused a large stimulation of glucose transport and stimulated recruitment of transferrin receptors to the plasma membrane (PM) in these cells, whereas platelet-derived growth factor (PDGF)-bb was virtually without effect on these responses. Subcellular fractionation studies after stimulation with PDGF-bb or insulin revealed a differential effect of these growth factors on subcellular localization of PI 3-kinase activity. PDGF was more effective than insulin in stimulating PI 3-kinase activity and recruiting the p85 alpha PI 3-kinase adaptor subunit in the fraction containing the PM. However, in the microsomal fraction insulin significantly increased PI 3-kinase activity and p85 alpha levels, whereas PDGF was almost without effect. In the microsomal membrane fraction the insulin-stimulated recruitment of p85 alpha closely matched the increase PI 3-kinase activity, indicating that insulin stimulation of PI 3-kinase in this fraction is largely due to recruitment of PI 3-kinase enzyme rather than alterations in specific activity. Insulin-stimulated recruitment of p85 alpha to the microsomal membranes was not inhibited by wortmannin, indicating that PI 3-kinase activity was not required for this process. A further level of compartment-specific regulation of PI 3-kinase in response to PDGF was revealed by the finding that tyrosine phosphorylation of the p85 alpha adaptor was restricted to the PM-containing fraction. Insulin had no effect on p85 tyrosine phosphorylation in either fraction. In summary, these results suggest a basis by which insulin and PDGF could both use PI 3-kinase signalling cascades but achieve different signalling outcomes.

Tuberculosis in England and Wales 1982-1993: notifications exceeded predictions.

About 8000 more cases of tuberculosis were notified in the years 1988 to 1993 than would have been expected if previous trends had continued (95% confidence interval (CI) 3000-12,000). Tuberculosis notifications were analysed to determine the population groups and geographical areas in which increases occurred. Increases were greatest in regional health authorities (RHAs) with large conurbations, while rates generally continued to decline in RHAs that serve more rural areas. The rate of notification rose significantly in males aged 15 to 34 years and fell in men over 35. In females, the rate increased only in the 35 to 64 year age group. Non-respiratory tuberculosis accounted for 21% of notifications in 1987 and 27% in 1993. In most age, sex, and geographical groups significant increases were confined to non-respiratory tuberculosis, but the rate of both respiratory and non-respiratory tuberculosis rose in males aged 15 to 34 years in North Thames RHAs. A number of factors may have contributed to the observed increase. Reporting of cases may have improved following the appointment of consultants in communicable disease control in 1988, whose role includes improving notification practices. Such improvements might explain the increased proportion of notifications of non-respiratory tuberculosis, as this may have been undernotified to a greater extent than respiratory tuberculosis in the past. Other factors associated with recent increases are discussed.

Construction of species-specific primers for Pseudomonas andropogonis based on 16S rDNA sequences.

Approximately 94% of the total 16S rDNA of Pseudomonas andropogonis strain ACH 01053A was sequenced and compared with that of strain ATCC 23061 obtained from the GenBank database. The two sequences were highly homologous with 1.3% difference. Alignment of sequences with those from closely related bacteria revealed two possible regions for design of a specific primer suitable for detection of Ps. andropogonis. Using primers designed to these variable regions in a PCR test, an amplification product of approximately 410 bp was specifically produced by 40 strains of Ps. andropogonis. No other bacterial species showed an amplification product under optimized PCR conditions. As few as 1000 cells per reaction were detected.

Bacterial whole cell protein profiles of the rRNA group II pseudomonads.

Studies on bacterial whole cell protein profiles showed that members of the rRNA group II pseudomonads were distinct from other non-fluorescent and fluorescent pseudomonads, including Pseudomonas aeruginosa, the type species of the genus Pseudomonas. Strains of Ps. andropogonis, Ps. caryophylli, Ps. gladioli pv. gladioli, Ps. pickettii, Ps. pseudomallei and Ps. rubrisubalbicans showed uniform and distinct protein patterns, while strains of Ps. solanacearum and Ps. cepacia displayed differences within species. Numerical analysis of their protein profiles with GelManager and Taxan programs generated dendrograms comprising 16 clusters at 89% similarity. Each cluster included strains belonging to the same species with the exception of Ps. solanacearum, which fragmented into three clusters. Pseudomonas solanacearum showed different protein patterns correlating with different biovars and the two divisions of Cook et al. (1989), as well as the results of 16S rRNA gene sequencing. The whole cell protein profiles of a total of 83 strains belonging to 14 bacterial species were numerically analysed.

Telluria mixta (Pseudomonas mixta Bowman, Sly, and Hayward 1988) gen. nov., comb. nov., and Telluria chitinolytica sp. nov., soil-dwelling organisms which actively degrade polysaccharides.

Pseudomonas mixta (type strain, ACM 1762 [= ATCC 49108], an actively dextranolytic species that possesses both lateral and polar flagella, was compared with the strictly aerobic, rod-shaped, chitinolytic bacterium "Pseudomonas chitinolytica" ACM 3522T (= CNCM I-804) (T = type strain), which has a similar flagellation pattern, by performing phenotypic characterization and DNA-DNA hybridization studies and by analyzing DNA base compositions and 16S rRNA sequences. Our results indicated that "P. chitinolytica" ACM 3522T was phenotypically and genotypically distinct from P. mixta and other phenotypically analogous Pseudomonas spp., Xanthomonas maltophilia, and other aerobic chitin degraders. The 16S rRNA sequences of strains ACM 1762T and ACM 3522T were found to be very similar (97%) to each other and indicated that these organisms are proteobacteria that belong to the beta subclass. The strains were deeply branched in the beta subclass and were distinct from other pseudomonads, including Pseudomonas cepacia, and from Comamonas testosteroni. On the basis of phenotypic, genotypic, and phylogenetic evidence, it is proposed that P. mixta and "P. chitinolytica" ACM 3522T represent two distinct species in a new genus called Telluria. Thus, the genus Telluria gen. nov. contains Telluria mixta comb. nov. and Telluria chitinolytica sp. nov., which are strictly aerobic, rod-shaped, soil-dwelling bacteria that are active polysaccharide degraders.

Heterogeneity, persistence, and distribution of Pseudomonas aeruginosa genotypes in cystic fibrosis patients.

A collection of 222 isolates of Pseudomonas aeruginosa was obtained from the respiratory tract of 16 patients with cystic fibrosis over a 4- to 9-month period. Fourteen of these patients were unrelated, while the remaining two were siblings. Isolates were typed by conventional pyocin typing and also by the use of a DNA probe containing 741 bp immediately upstream of the exotoxin A structural gene and the initial 732 bp of the exotoxin A structural gene. By pyocin typing, 69% (11 of 16) of the patients were shown to harbor a single type that persisted in the lung throughout the study. By genotyping (DNA probe typing), all but three patients (13 of 16, 81%) harbored a single persistent genotype in their lungs. Six patients other than the sibling pair (6 of 14, 43%) shared a common genotype in their lungs as judged by DNA probing, and the pyocin type of these isolates was also identical. In four of these six patients, the shared genotype was also the persistent genotype. The sibling pair studied also carried a common genotype in their lungs as indicated by DNA probing, even though the pyocin type of these isolates varied. Results presented suggest that the majority of patients harbor a persistent strain in their lungs and that cross-colonization may occur.

Phenotypic conversion of Pseudomonas aeruginosa in cystic fibrosis.

Pseudomonas aeruginosa strains isolated from cystic fibrosis patients were tested for production of exoenzymes, sensitivity to pooled normal human serum, and colony morphology. Strains isolated from patients exhibiting a severe form of the disease were seen to produce a decreased range of exoenzymes, to show an increase in their serum sensitivity, and to be predominantly mucoid in colonial character compared with strains isolated from patients with a milder form of the disease. These results suggest that P. aeruginosa undergoes phenotypic changes with respect to exoenzyme secretion, serum sensitivity, and colony form as the clinical condition of the cystic fibrosis patient changes.

Numerical taxonomy of proteolytic psychrotrophs from Queensland raw milks.

Eighty-seven proteolytic psychrotrophic micro-organisms were isolated from 11 bulk milk supplies of two Queensland factories from different climatic regions, before and after storage at 4 degrees C for 7 d. These isolates together with 15 reference strains formed the basis of a numerical taxonomic study involving 81 attributes. All but six isolates were pseudomonads. The strains clustered into nine groups, of which one group consisted of four yeasts. One group, containing 39 isolates, was designated as Pseudomonas fluorescens biovar 1; three groups, containing 27 isolates, as Ps. fluorescens biovar 5; and one group, containing 10 isolates, as Ps. putida biovar A. This study showed that the proteolytic psychrotrophic microflora of the 11 milks supplying the two factories was substantially different and that the proteolytic flora of 7 d refrigerated milk could not be estimated by examining the flora before storage.

Nutritional requirements and biochemical activities of pineapple pink disease bacterial strains from Hawaii.

Bacteria which cause pink disease of pineapple, identified on the basis of their nutritional and biochemical activities, were found to belong to three genera. These bacteria include the following species: Gluconobacter oxydans, Acetobacter aceti, and Erwinia herbicola. Several pink disease strains required one to three vitamins for growth. Both G. oxydans strains 303D and 180 required biotin, nicotinic acid, and pantothenic acid for growth; E. herbicola 189 required only nicotinic acid; however, A aceti 295 was able to grow without any added supplements in glucose mineral salts medium. Optimal vitamin concentrations for maximal growth and optimal pH for the maximal number of generations per hour was established for a few pink disease strains.

Could a tuberculosis epidemic occur in London as it did in New York?

In early 1999, more than 160 senior physicians, public health officials, and nurses met to discuss London's tuberculosis (TB) control program. The program was examined against the public health response of New York City's Bureau of Tuberculosis Control during a 1988 to 1992 epidemic. This article outlines TB epidemiology and control in New York City 10 years ago and in London today to assess whether the kind of epidemic that occurred in New York could occur in London.
