Flow cytometric analysis of lymphocytes isolated from nasal polyps.

We analyzed by flow cytometry the expression of IL-2 receptors (alpha subunit-CD25) and ICAM-1 adhesion molecules (CD54) on T cells and subsets (CD4, CD8) isolated from nasal polyp tissue in allergic and non-allergic patients. We found a significant increase in IL-2 receptor and ICAM-1 molecule expression on T cells isolated from nasal polyp tissue compared to peripheral blood lymphocytes. We also found a significantly increased expression of ICAM-1 molecules on CD8+ cells in non-allergic compared to allergic patients. The latter may reflect a difference in cytotoxic immune response between allergic and non-allergic patients, but the result should be confirmed in a more extensive study including cytokine and immunoglobulin analysis. We hope that it would enable us to obtain a deeper insight into the local immune events and further to clarify the etiology and pathogenesis of nasal polyps and their relation to allergy.

Astereological analysis of the FRTL-5 cells up to four days after trypsinisation.

FRTL-5 cells grow as a monolayer culture in vitro and they must be detached from the solid surface by trypsin before reseeding. The aim of this study was to examine by light microscopy and by the astereological method the changes that occur in FRTL-5 cells one, two, three or four days after trypsinisation. The most pronounced qualitative and quantitative changes were seen from the first to the second day after trypsinisation, as the area of the cells increased by 119%, their nuclei by 71% and the cytoplasm by 150%, while the nuclear-cytoplasmic ratio decreased by 42%. Significant increases in the average area of the cells, their average nuclear diameter and perimeter were seen also on the third and fourth days, while the average nuclear-cytoplasmic ratio decreased significantly on the first three days after trypsinisation and then remained constant.

Expression of Ki-67 antigen and proliferative cell nuclear antigen in benign and malignant epithelial lesions of the larynx.

In an attempt to analyse the proliferative activity in benign and malignant laryngeal epithelial lesions, and to determine the relationship to their histologic grade, we studied the expression of proliferative cell nuclear antigen (PCNA) and Ki-67 antigen on 20 squamous carcinomas, and on 30 biopsies of epithelial hyperplasia categorized according to the Kambic-Lenart classification into simple, abnormal, and atypical hyperplasias. In simple hyperplasia, both antibodies stained the nuclei of the occasional cells in the basal layer. In abnormal hyperplasia (mild dyplasia), positive cells occupied up to a third, and in atypical hyperplasia (moderate and severe dysplasia) they occupied from two-thirds to the entire epithelial thickness. In squamous carcinoma, we have found a statistically significant correlation between its grade and the percentage of Ki-67-(p < 0.01) and PCNA-(p < 0.00001) positive cells. Our results suggest that the proliferative fraction progressively increases with the degree of epithelial hyperplasia and the grade of carcinoma. We conclude that the patterns of immunoreactivity to PCNA and Ki-67 antigen correspond to the histologic grade of both benign and malignant epithelial lesions of the larynx. This method should be regarded as a useful adjunct to traditional histological techniques allowing more objective grading of benign and malignant epithelial lesions.

Ag-NOR counting for differentiation of follicular thyroid tumours.

According to the histological typing system of thyroid tumours adapted by the World Health Organisation follicular tumours are classified as follicular adenomas and follicular carcinomas. The most important diagnostic criterion for follicular carcinoma is the evidence of vascular and/or capsular infiltration. The aim of this work was to verify the Ag-NOR counting method for the diagnosis of thyroid tumours. From the material of the experiment on mice that drunk thyrostatic (1.2% NaCIO4) and were irradiated 10 histologically normal thyroids, 10 showing hypertrophy and hyperplasia, 8 follicular adenomas and 10 follicular carcinomas were used. Thyroid glands were impregnated with the silver to present Ag-NORs. The average numbers of Ag-NORs per nucleus (Ag-NOR/nucleus) for each group were determined. The results showed that the method of counting Ag-NOR can be used to differentiate among thyroid tumours.

Measurement characteristics of stereological and histochemical methods in the diagnosis of thyroid tumours.

Seventy-two female mice of BALB/c strain were divided into three control and three perchlorate groups. The perchlorate groups drank 1.2% sodium perchlorate solution. After 8 or 32 weeks of the 46-week experiment one control and one perchlorate group were totally irradiated with 0.8 Gy on 5 consecutive days. Ten normal thyroid glands, 10 with hypertrophy and hyperplasia, 8 follicular adenomas and 10 follicular carcinomas were histologically and stereologically analysed. The results showed significant differences in the average nuclear volume (p < 0.05) and in the average number of the silver-stained nucleolar organiser regions (AgNORs) per nucleus (p < 0.005) between the follicular adenomas and the follicular carcinomas. The method of counting Ag-NORs per nucleus was more valid, more accurate and more economical than the method of measuring the average nuclear volume.

[The nucleolar organizer region in malignant non-Hodgkin's lymphoma].

Nucleolar organizer region-associated proteins (AgNORs) have been studied in parafin sections of 15 non-Hodgkin's lymphomas by a silver staining technique. On the basis of the Working Formulation of malignant non-Hodgkin's lymphomas all studied cases were divided into three groups. A statistically significant difference was found between number of AgNORs in the nuclei of low-grade lymphomas (mean 2.654 per nucleus), and also between intermediate-grade (mean 1.588 per nucleus) and high-grade malignant lymphomas. Significant difference was also observed between AgNOR area in high-grade and low-grade malignant lymphomas and between high-grade and intermediate-grade malignant lymphomas. However no statistically significant differences between all three studied groups with regard to the size of nucleus were found. It is suggested that this method can be very useful in the diagnosis of malignant non-Hodgkin's lymphomas.
