Early modifications of nucleic acid metabolism and nuclear lipid biosynthesis associated with antiproliferative activity of interferon-alpha on Daudi lymphoma cells.

The effect of human recombinant DNA interferon-alpha type A (rIFN alpha A) on nuclear lipid biosynthesis and on in vitro nucleic acid synthesis was investigated in Daudi lymphoma cells sensitive (Daudi Is) or resistant (Daudi Ir) to the antiproliferative activity of this glycoprotein. In the Daudi Is studied at 90 min and up to 8 h, relative proportions of 3H-labeled nuclear lipids were reproducibly altered as compared to controls: phosphatidylcholine increased while phosphatidylserine and total neutral lipids decreased. These changes were not detected in parallel studies of the whole cell extracts. No significant changes in the profiles of nuclear lipids were observed in Daudi Ir. Decreased rates of alpha DNA polymerase and of RNA transcription were evident within 90 min in the Daudi Is nuclei but not in untreated controls or nuclei from rIFN alpha A-treated Daudi Ir cells, thus suggesting a possible relationship of the rapid alterations of nuclear lipid biosynthesis in Daudi Is cells to the rIFN alpha A antiproliferative activity.

Influence of suckling on plasma concentrations of somatostatin, insulin and gastrin in lactating women.

The effects of suckling on plasma somatostatin, insulin and gastrin values were evaluated in eight nursing women on the 3rd to 4th day postpartum. Plasma prolactin levels were also determined. Prolactin levels increased in response to suckling, as expected. Insulin levels also rose, whereas somatostatin and gastrin concentrations did not change after suckling. It is possible that the suckling-induced hyperinsulinemia blunts the somatostatin response to suckling in humans.

Construction and characterization of a protective antigen-deficient Bacillus anthracis strain.

The pag gene coding for protective antigen (PA), one of the three toxin components of Bacillus anthracis, has been cloned into the mobilizable shuttle vector pAT187 and transferred by conjugation from Escherichia coli to B. anthracis. Using this strategy, an insertionally mutated pag gene constructed and characterized in E. coli, was introduced into B. anthracis Sterne strain. This transconjugant was used to select a recombinant clone (RP8) carrying the inactivated pag gene on the toxin-encoding plasmid, pXO1. Strain RP8 was deficient for PA while still producing the two other toxin components, i.e. lethal factor (LF) and edema factor (EF). In contrast to spores from the wild-type Sterne strain, spores prepared from RP8 were totally non-lethal in mice. These results clearly establish the central role played by PA in B. anthracis pathogenicity.

Twenty-four-hour secretory pattern of vasoactive intestinal polypeptide in the elderly.

A chronobiological study was carried out in 8 elderly male subjects (74-85 years) to evaluate the 24-hour vasoactive intestinal polypeptide (VIP) secretory pattern. Eight young adult males (21-32 years) made up the control group. Blood samples were drawn from each subject every 2 h during the day and hourly during the night for a 24-hour period. Mean 24-hour VIP values in elderly (21.1 +/- 0.3 pg/ml) and young adults (19.1 +/- 0.3 pg/ml) did not differ statistically, but daytime VIP values observed in elderly subjects (21.4 +/- 0.5 pg/ml) were higher (p less than 0.05) than those recorded in young adults (17.5 +/- 0.5 pg/ml). The young adults showed significant (p less than 0.05) circadian VIP fluctuations with highest values during the nighttime, while the elderly subjects did not. An age-related decreased activity of the peripheral neuronal VIP-ergic network is hypothesized.

Gamma heavy chain disease associated with Hodgkin's disease. Clinical, pathologic, and immunologic features of one case.

This report describes the clinical, pathologic, and immunologic features of a patient with gamma heavy chain disease (gamma-HCD) associated with Hodgkin's disease (HD). The diagnosis of gamma-HCD was established by serum electrophoresis and immunoelectrophoresis and confirmed by biochemical analysis of patient's serum showing the presence of an incomplete gamma chain, with an approximate molecular weight of 40 kilodaltons. The diagnosis of HD rested upon the presence of systemic lymphadenopathy, the typical histologic pattern and reactivity of Reed-Sternberg cells with the LeuM1-CD15 monoclonal antibody. The two diseases developed independently, in the absence of any immunosuppressive treatment. Furthermore, there was some evidence suggesting that HD tissue was not responsible for the production of the incomplete gamma chain. This and similar cases may provide a model for a better understanding of the events leading to the simultaneous outgrowth of two lymphoid neoplasias in the same patient.

Plasma beta-endorphin, ACTH and cortisol secretion in man after nasal spray administration of calcitonin.

Beta-endorphin, ACTH and cortisol secretion were measured in twelve healthy adult males after nasal spray administration 200 IU salmon calcitonin. A significant increase in plasma beta-endorphin, from 19.2 ng/l under basal conditions to a peak of 27.1 ng/l at 30 min was recorded. Plasma ACTH and cortisol were not affected. In individual subjects the beta-endorphin level was increased in eight of the twelve, ACTH rose in three and cortisol did not change in any of them. The data indicate that calcitonin induced a beta-endorphin increase independent of enhanced corticotrophin-cortisol release.

Influence of phosphatidylcholine on two protein mRNA levels in interferon treated Daudi cells.

Evidence related to nuclear structure and metabolism have shown the presence of a chromatin lipid fraction which may be involved in the events of cell proliferation and differentiation (Maraldi et al., 1984, Cocco et al., 1987). The relative amounts of these molecules have been reported to be modified in different metabolic conditions of normal and neoplastic cells (Coetzee et al., 1975; Manzoli et al., 1982). Phospholipids have also been found among the components of the nuclear matrix where they seem to play a role in the interactions between nucleic acids and proteins (Berezney, 1984; Cocco et al., 1987). In this paper we report that the transcriptional induction of two genes (pIFN-IND-1 and pIFN-IND-2) produced by interferon in Daudi lymphoma cells is influenced by addition, in the transcriptional assay, of phosphatidylcholine (PC) liposomes which have been reported to be able to enter nuclei (Maraldi et al., 1984; Maraldi et al., 1982).

Plasma beta-endorphin concentrations during suckling in lactating women.

Beta-endorphin appears to be involved in the hormonal response to suckling in some animals. The peripheral secretory patterns of beta-endorphin, prolactin and cortisol were investigated in serial venous blood samples taken during suckling from eight healthy women who were breast-feeding on the third or fourth day of the puerperium. Plasma levels of prolactin and beta-endorphin increased significantly during suckling reaching a peak after 20 min, levels of cortisol remained unaffected. It is suggested that the increased beta-endorphin derives from an extra-hypophyseal source.

Plasma somatostatin and vasoactive intestinal polypeptide responses to an oral mixed test meal in obese patients.

Ten obese and 10 control subjects were studied in basal conditions and after ingestion of a standard mixed test meal. Blood glucose, insulin, somatostatin (SLI) and vasoactive intestinal polypeptide (VIP) concentrations were determined before and 30, 60, 90, 120, 180 and 240 min after the start of the meal. Basal SLI levels in the obese (14.4 +/- 0.7 ng/l) were not significantly different from those in the controls (15.5 +/- 0.8 ng/l), whereas after the meal a blunted secretory response was recorded. Baseline plasma VIP levels were higher in the obese (29.7 +/- 1.5 ng/l) than in the control subjects (19.8 +/- 1.3 ng/l) and, similarly to the controls, were unaffected by meal ingestion. Data suggest that in the course of obesity an enhanced VIP secretion in association with a diminished SLI responsiveness to meals occurs.

Evidence for a reduction of the replicative activity of matrix-bound DNA polymerase alpha following treatment with phospholipase C.

Since phospholipids have been suggested to play some role in the molecular organisation of the nuclear matrix, the effect of their removal by means of phospholipase C has been investigated in regenerating rat liver nuclear matrix. The matrix-bound DNA polymerase alpha shows an almost complete loss of activity following the digestion with phospholipase C. Since the polymerase activity is restored by adding exogenous DNA, we suggest that the effect is due to the removal of matrix residual DNA, which is in some way linked to the nuclear matrix by means of hydrophobic interactions.

Diurnal changes of plasma and cerebrospinal fluid somatostatin and 24-h growth hormone secretory pattern in man. A study in hydrocephalic patients.

Somatostatin concentration was determined in plasma and in cerebrospinal fluid during a 24-h period in 7 male patients suffering from hydrocephalus of differing aetiologies. Blood and ventricular cerebrospinal fluid samples were taken every 2 h during the day (08.00-22.00 h) and every hour during the night (24.00-07.00 h). Simultaneously, plasma growth hormone levels were also evaluated. Plasma SRIH levels showed significant circadian variations with highest values in the daytime and lowest values during the night. Cerebrospinal fluid SRIH did not show any significant time-related circadian changes. Plasma GH levels showed the well-known circadian pattern in the majority of patients. No significant correlation was found between the plasma GH and plasma or cerebrospinal fluid SRIH values recorded during the 24-h period. Results suggest that peripheral SRIH does not play any major role in the control of the 24-h GH secretory pattern in man.

Prolactin release induced by physical exercise is independent from peripheral vasoactive intestinal polypeptide secretion.

The plasma concentrations of vasoactive intestinal polypeptide and prolactin were measured before and after an exhaustive and a submaximal exercise test in 7 male marathon runners. A significant increase of vasoactive intestinal polypeptide was recorded after both tests, whereas the prolactin increase was observed only after the exhaustive exercise test. No significant correlation was found between the plasma vasoactive intestinal polypeptide and the plasma prolactin values recorded during the two exercise tests. Data suggest that the exercise-induced prolactin release occurs independently from variations of the vasoactive intestinal polypeptide levels in peripheral circulation.

Somatostatin in the elderly: diurnal plasma profile and secretory response to meal stimulation.

Plasma somatostatin levels were determined during a 24-hour period and after a meal test in 7 and 5 elderly subjects (76-90 years), respectively. The data obtained were compared with those recorded in young adult subjects (22-30 years). Increased basal somatostatin values were found in elderly subjects (20.0 +/- 1.5 pg/ml) when compared to young adults (14.1 +/- 0.6 pg/ml; p less than 0.01). Also, the mean 24-hour somatostatin levels were higher in the elderly (21.3 +/- 0.8 pg/ml) than in young adults (16.7 +/- 0.5 pg/ml; p less than 0.01), and minor diurnal variations were found in the former group. The response to the meal test was less evident in the elderly than in the control group. The data obtained indicate an increased basal somatostatin production associated with a diminished variability throughout the 24-hour period and in relation to meals.

Pharmacokinetic profile of fosfomycin trometamol (Monuril).

The kinetics of fosfomycin in serum and in urine was studied in 5 healthy volunteers after intravenous administration of disodium fosfomycin and after oral administration of trometamol fosfomycin at 50 mg/kg. The presence of secondary peaks in serum kinetics, more evident after oral administration, requires the use of a compartmental model with enterohepatic recirculation to which all the data of each subject were simultaneously fitted. The following values of the various pharmacokinetic parameters were calculated: central volume 10.6 +/- 0.92 liters; bioavailability 0.58 +/- 0.04; delay time in the recirculation 2.00 +/- 0.92 h; half-life 2.43 +/- 0.31 h; total clearance 8.3 +/- 1.6 l/h; urinary clearance 7.0 +/- 0.9 l/h; peak level 32.1 +/- 3.0 micrograms/ml; time of the peak 2.2 +/- 0.44 h. The fosfomycin concentration in urine remains above 1,000 micrograms/ml for 12 h and above 100 micrograms/ml for 48 h. The oral kinetics of fosfomycin is dose-dependent, as shown by serum and urine kinetics in other 4 volunteers after oral administration of 2, 3, 4 and 5 g; the fraction of the dose excreted by urine goes from 50% for 2 g to 22% for 5 g.

Twenty-four-hour beta-endorphin secretory pattern in the elderly.

A chronobiological study was carried out in seven elderly male subjects (78-84 years) to evaluate the 24-h beta-endorphin secretory pattern. Seven young adult males (28-37 years) made up the control group. Blood samples were drawn every four hours from 08.00 to 20.00 h and every two hours from 24.00 to 06.00 h. ACTH and cortisol levels were also determined in the same plasma samples. Mean 24-h beta-endorphin values in the elderly (32.6 +/- 1.1 ng/l) and in the young adult male subjects (29.5 +/- 1.4 ng/l) did not differ statistically, but the circadian rhythm was absent in the elderly subjects. In the elderly, plasma ACTH and cortisol concentrations showed a circadian rhythm similar to that observed in the adult subjects. However, in the elderly patients, in contrast to that in the adult subjects, the multilinear regression analysis did not show any statistically significant correlation between the beta-endorphin, ACTH and cortisol 24-h plasma concentrations.

Phospholipids as components of the nuclear matrix: their possible biological significance.

The phospholipid involvement in the regulation of the functional and structural properties of the nuclear matrix is discussed analysing the results obtained with the enzymic removal of these molecules. Namely phospholipids seem to mediate hydrophobic interactions between nucleic acids and matrix fibrils either directly or indirectly through an association with the non-histone proteins of the matrix.

Positive inotropic effects of ibopamine in patients with congestive heart failure. A multicenter investigation.

A multicenter double-blind placebo-controlled investigation was carried out in 42 patients with severe congestive heart failure who were given ibopamine (SB-7505), the 3,4-diisobutyryl ester of N-methyldopamine, for a period of 10 days in addition to previous treatment with digitalis and diuretics. Symptoms were evaluated by using a suitable score; some patients were submitted to an exercise test along with other clinical tests. Compared with placebo, ibopamine induced no significant changes in heart rate and arterial blood pressure. Clinical scores were lower after ibopamine than after placebo (p less than 0.01). Functional capacity, defined as the duration (in s) of a maximum symptom-limited exercise test using the ergometric bicycle with a constant 25-W workload, was significantly higher after ibopamine than after placebo (p less than 0.05). So a clear symptomatic improvement was observed after ibopamine in cardiac patients with prolongation of exercise time and reduced clinical scores. In conclusion, the results obtained, along with the slight improvement in the systolic time intervals, indicate that ibopamine is capable of improving cardiac performance in patients with advanced heart failure who display symptoms despite treatment with digitalis and diuretics.
