Multiphoton fluorescence microscopy.

Multiphoton fluorescence microscopy has now become a relatively common tool among biophysicists and biologists. The intrinsic sectioning achievable by multiphoton excitation provides a simple means to excite a small volume inside cells and tissues. Multiphoton microscopes have a simplified optical path in the emission side due to the lack of an emission pinhole, which is necessary with normal confocal microscopes. This article illustrates examples in which this advantage in the simplified optics is exploited to achieve a new type of measurements. First, dual-emission wavelength measurements are used to identify regions of different phase domains in giant vesicles and to perform fluctuation experiments at specific locations in the membrane. Second, we show how dual-wavelength measurements are used in conjunction with scanning fluctuation analysis to measure the changes in the geometry of the domains and the incipient formation of gel domains when the temperature of the giant vesicles is gradually lowered.

Synergistic anti-proliferative effects of vitamin D derivatives and 9-cis retinoic acid in SH-SY5Y human neuroblastoma cells.

This study examines the effect of 1,25-dihydroxyvitamin D(3) [1,25(OH)(2)D(3)], 24,25-dihydroxyvitamin D(3) [24,25(OH)(2)D(3)], two vitamin D analogues (KH 1060 and EB 1089, which are 20-epi-22-oxa and 22,24-diene-analogues, respectively), 9-cis retinoic acid and all-trans retinoic acid on proliferation of SH-SY5Y human neuroblastoma cells, after treatment for 7 days. Cell number did not change when the cells were incubated with 1, 10 or 100 nM 1,25(OH)(2)D(3) or its derivatives, but significantly decreased in the presence of the two retinoids (0.001--10 microM final concentration). A synergistic inhibition was observed, when SH-SY5Y cells were treated combining 0.1 microM 9-cis retinoic acid and 10 nM 1,25(OH)(2)D(3) or 10 nM KH 1060, and 1 microM 9-cis retinoic acid and 10 nM 1,25(OH)(2)D(3) or 10 nM EB 1089. Acetylcholinesterase activity showed a significant increase, in comparison with controls, after treatment of the cells for 7 days with 0.1 or 1 microM 9-cis retinoic acid, alone or combined with 10 nM 1,25(OH)(2)D(3) or 10 nM KH 1060 or 10 nM EB 1089. This increase was synergistic, combining 1 microM 9-cis retinoic acid and 10 nM 1,25(OH)(2)D(3) or EB 1089. The levels of the c-myc encoded protein remarkably decreased after treatment of SH-SY5Y cells for 1, 3, 7 days with 0.1 and 1 microM 9-cis retinoic acid, alone or combined with 10 nM 1,25(OH)(2)D(3) or 10 nM KH 1060 or 10 nM EB 1089. In particular, the association of 1 microM 9-cis retinoic acid and 10 nM 1,25(OH)(2)D(3) or 10 nM EB 1089 resulted in a synergistic c-myc inhibition, in comparison with that obtained in the presence of the retinoid alone. These findings may have therapeutic implications in human neuroblastoma.

The nu(2), nu(4) + nu(5), nu(6) + nu(9), and nu(8) Band System in 1,1,1-Trifluoroethane.

High-resolution FTIR spectra of 1,1,1-trifluoroethane (HFC-143a) have been recorded in the region from 1370 to 1470 cm(-1) with an unapodized resolution of 0.0016 cm(-1) at room temperature and of 0.004 cm(-1) at 183 and 100 K. The two main infrared active bands of A(1) symmetry have been shown to be nu(2) at 1407.5 cm(-1) and nu(4) + nu(5) at 1440.5 cm(-1). With the aid of Raman spectra, the two infrared inactive bands of E symmetry in this spectra region have been shown to be nu(8) at 1457.5 cm(-1) and nu(6) + nu(9) at 1446.2 cm(-1). The nu(2) band was analyzed as an isolated band, whereas the nu(4) + nu(5) band was analyzed as part of the triad nu(4) + nu(5), nu(6) + nu(9), and nu(8). Copyright 2000 Academic Press.

Cholangiocarcinomas express Fas ligand and disable the Fas receptor.

Cholangiocarcinoma is a highly-malignant adenocarcinoma originating from cholangiocytes. Current concepts support escape from immune surveillance using aberrant expression of Fas ligand (FasL) and dysregulation of receptor (FasR) signaling as a potential mechanism for tumor progression. Our aims were to determine if altered expression of FasR and FasL or changes in expression of FLICE inhibitor (I-FLICE) allow cholangiocarcinoma cells to escape immune surveillance. Human cholangiocarcinoma cell lines were evaluated for the functional expression of FasR and FasL by (1) quantitating apoptosis after incubation of cells with agonistic antibodies and (2) an in vitro cell death assay involving coculture of cholangiocarcinoma cells with Fas-sensitive thymocytes. I-FLICE antisense treatment was performed by stable transfection with complementary DNA (cDNA) for I-FLICE in the reverse orientation. We found that normal cholangiocytes in vivo express FasL. Human cholangiocarcinoma cell lines express both FasL and FasR and I-FLICE. FasL expressed by cholangiocarcinomas in vitro induced lymphocyte cell death (70% after 24 hours). Despite the expression of FasR, exposure of the cells to agonistic antibodies (500 ng/mL) induced only minimal apoptosis in the Jurkat cells. Antisense treatment of cholangiocarcinomas in vitro with I-FLICE reduced protein expression of I-FLICE by 90% to 95% and increased Fas-mediated apoptosis 2-fold. We concluded that cholangiocarcinomas escape immune surveillance either by disabling FasR signaling through the expression of I-FLICE and/or increased FasL expression to induce apoptosis of invading T cells. Reduction of I-FLICE expression in cholangiocarcinoma cells restored Fas-mediated apoptosis. Therapeutic maneuvers to inhibit expression of I-FLICE may aid in the treatment of cholangiocarcinoma.

Vitamin D receptor in SH-SY5Y human neuroblastoma cells and effect of 1,25-dihydroxyvitamin D3 on cellular proliferation.

This study examines the effect of 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] on proliferation of SH-SY5Y human neuroblastoma cells and demonstrates, for the first time, the presence of vitamin D receptor (VDR) in this cell line. Cell number showed a significant decrease, when the cells were incubated with 1 or 10 nM 1,25(OH)2D3 for 24, 48, 72, 96 and 144 h, while 100 nM 1,25(OH)2D3 was ineffective after 24 and 96 h incubation. The highest inhibition (about 35%) was observed after 72 h treatment with the hormone at the three concentrations used. Protein content per cell increased, in comparison with controls, after treatment of SH-SY5Y neuroblastoma cells with 1,25(OH)2D3, at the three concentrations, up to 96 h incubation. 1, 10 or 100 nM 1,25(OH)2D3 positively affected [3H]thymidine incorporation after treatment of the cells for 48 and 72 h, while, after 24 h, only 10 nM 1,25(OH)2D3 exerted a stimulatory action. To study the expression of the VDR gene, Northern blot analysis was performed. Subconfluent SH-SY5Y neuroblastoma cells were treated for 24 h with medium containing 10 nM 1,25(OH)2D3 or vehicle alone. A main transcript of an approximate size of 4.5 kb, present either in controls or in cells incubated with the hormone, was revealed. A limited increase in VDR mRNA levels was observed in the cells treated with 1,25(OH)2D3, fetal bovine serum or forskolin. Only slight differences in morphology were perceived between SH-SY5Y cultures maintained with or without 10 nM 1,25(OH)2D3 for 7 days.

Role of 1,25-dihydroxyvitamin D3 and extracellular calcium in the regulation of proliferation in cultured SH-SY5Y human neuroblastoma cells.

This study examines the effects of 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] on SH-SYSY human neuroblastoma cells cultured in the presence of medium containing varying concentrations of calcium (0.1, 0.9, 1.4, 1.8 mM). Pyruvate kinase activity was assayed in SH-SY5Y cells incubated in variable calcium medium with or without 1, 10 or 100 nM 1,25(OH)2D3 for 48 h. The enzyme levels showed a significant increase in comparison with control, when the cells were incubated with 100 nM hormone in the presence of 0.1 mM calcium, while pyruvate kinase activity decreased, when the cells were treated with 100 nM 1,25(OH)2D3 in the presence of 1.8 mM calcium. The proliferative activity of SH-SY5Y was dependent on the extracellular concentration of calcium, being the highest at 1.8 mM calcium and completely absent at 0.1 mM calcium. In the presence of 1,25(OH)2D3, at the three concentrations used and after 48 h incubation, a significant decrease in cell number was always observed, without a direct correlation between 1,25(OH)2D3 effect and calcium concentration in the medium. [3H]Thymidine incorporation in SH-SY5Y cells significantly increased in comparison with control, when the 48 h incubation with 1, 10 or 100 nM 1,25(OH)2D3 was carried out in the presence of 0.1 mM calcium, while, at the other calcium concentrations, the hormone did not cause any significant change in this parameter. The treatment of SH-SYSY cells with 1 nM 1,25(OH)2D3 for 48 h did not affect cell morphology, when 0.1 mM calcium was present, while, in the medium containing 1.8 mM calcium, the treated cells showed a slight trend to differentiation. The differentiating effect of 10 microM all-trans retinoic acid, even if incomplete after 48 h treatment, was only observed in the cultures grown in 1.8 mM calcium, in comparison with those maintained in 0.1 mM calcium.

Dysregulation of apoptosis in the cholangiopathies and cholangiocarcinoma.

The importance of cholangiocytes, the epithelial cells lining bile ducts, in the biology and pathobiology of biliary epithelia is rapidly growing due to the advent of suitable experimental models and techniques to study these cells. However, the role of cholangiocytes as a major cellular target in a variety of severe hepatobiliary diseases or cholangiopathies remains unanswered. As the biology of cholangiocyte death evolves, apoptosis has emerged as a key player in the development of ductopenia in these cholangiopathies. Cholangiocytes are continuously exposed to a variety of genotoxic insults, such as chronic inflammation and hydrophobic bile acids. This chronic exposure may predispose cholangiocytes to oncogenic mutations and the further progression to malignancy (or cholangiocarcinoma [CC]), due, in part, to failure to activate apoptosis and delete cells with genetic damage.

Glutathione depletion is associated with decreased Bcl-2 expression and increased apoptosis in cholangiocytes.

Cholangiocytes are the target of a group of liver diseases termed the cholangiopathies that include conditions characterized by periductal inflammation and cholangiocyte apoptosis. Because inflammation is associated with oxidative stress, we developed the hypothesis that cholangiocytes exposed to oxidative stress will be depleted of endogenous cytoprotective molecules, leading to cholangiocyte apoptosis. To begin to test this hypothesis, we explored the relationships among glutathione (GSH) depletion, expression of Bcl-2 (a protooncogene that inhibits apoptosis), and apoptosis in a nonmalignant human cholangiocyte cell line. Monolayers of human bile duct epithelial cells, derived from normal liver and immortalized by SV40 transformation, were depleted of GSH using buthionine sulfoximine (BSO). Bcl-2 expression was assessed by quantitative immunoblot analysis, and apoptosis quantified by fluorescence microscopy using the DNA binding dye 4', 6'-diamidino-2-phenylindole. Bcl-2 message was assessed by RNase protection assay, and Bcl-2 protein synthesis and half-life by pulse-chase analysis. Exposure of human cholangiocytes in culture to BSO reduced GSH levels by 93 +/- 3% (P < 0.01). In addition, treatment of cholangiocytes with BSO reduced Bcl-2 levels by 87 +/- 2% (P < 0.01) and was associated with a time-dependent increase in the number of cells undergoing apoptosis; approximately 11 +/- 1% of cultured cells demonstrated morphological changes of apoptosis by 72 h compared with 1.5 +/- 0.1% in untreated cholangiocytes (P < 0. 01). Maintenance of GSH levels by addition of glutathione ethyl ester in the presence of BSO blocked the BSO-associated increase in apoptosis in BSO-treated cholangiocytes and also prevented the decrease in Bcl-2 protein. BSO treatment of cholangiocytes did not change steady-state levels of bcl-2 mRNA or Bcl-2 protein synthesis. However, Bcl-2 protein half-life decreased 57% in BSO-treated vs. untreated cells. Our results using a human cholangiocyte cell line demonstrate that reduction in the cellular levels of an antioxidant such as GSH results in increased degradation of Bcl-2 protein and an increase in apoptosis. These data provide a mechanistic link between the consequences of oxidative stress and cholangiocyte apoptosis, an observation that may be important in the pathogenesis of the inflammatory cholangiopathies.

[The malariology centenary (1898-1998). 1898].

This editorial note presents the journal initiative to celebrate the Malariology Centenary. It includes the reprint of the first annual report of the Society for Malaria Studies (founded in 1898), presented by Angelo Celli on December 3rd, 1898, to the first meeting of the Society and published in the Journal of the Royal Italian Society of Hygiene (volume 20, issue 12, 31st December 1898). The text is followed by explanatory footnotes and relevant references. An exhaustive historical essai, including the english version of the Celli's report, is scheduled to appear as supplement to volume 41, and the same volume will host the contributions issued from the international conference. "The malaria challenge after one hundred years of malariology", held in Roma at the Accademia Nazionale dei Lincei on November 16-19, 1998.

Bcl-2 is overexpressed and alters the threshold for apoptosis in a cholangiocarcinoma cell line.

Cholangiocarcinoma is a malignant neoplasm originating from cholangiocytes. The mechanisms responsible for oncogenesis of cholangiocytes are unknown. Resistance to apoptosis, especially by altered expression of B-cell lymphoma/leukemia 2 (Bcl-2) family members, has been implicated as a mechanism contributing to malignant transformation. Thus, our aim was to test the hypothesis that altered expression of Bcl-2 family members by cholangiocarcinoma cells renders them resistant to apoptosis. We compared the apoptotic threshold and expression of the Bcl-2 protein family members, Bcl-2, Bcl-XL, and Bax, in two human cell lines: 1) nonmalignant human cholangiocytes immortalized by transfection with the simian virus 40 (SV 40) large T antigen; and 2) a malignant human cholangiocarcinoma cell line. Apoptosis was induced pharmacologically using beauvericin. Bcl-2, Bcl-x long, and Bax protein expression were evaluated by immunoblot analysis, and Bcl-2 expression was modulated using antisense technology. The cholangiocyte and malignant/nonmaligant phenotype of both cell lines was verified using both in vitro and in vivo approaches. Beauvericin induced apoptosis of nonmalignant cholangiocytes in a concentration- (0 to 25 micromol/L) and time- (0 to 6 hours) dependent manner. In contrast, malignant cholangiocytes were resistant to apoptosis. Although expression of Bcl-x long and Bax protein were similiar in the two cell lines, Bcl-2 protein expression was 15-fold greater in malignant than in nonmalignant cholangiocytes. An 18 mer bcl-2 antisense oligonucleotide reduced expression of Bcl-2 protein by 50% and increased the rate of beauvericin-induced apoptosis more than threefold in the malignant cells. Our results support the hypothesis that resistance to apoptosis by overexpression of Bcl-2 may be a feature of cholangiocarcinoma.

Vitamin D receptor in IMR-90 human fibroblasts and antiproliferative effect of 1,25-dihydroxyvitamin D3.

This study examines the effect of 1,25-dihydroxyvitamin D3 [1,25(OH)2D3], serum or forskolin on the proliferation of IMR-90 fetal lung fibroblasts and demonstrates, for the first time, the presence of 1,25(OH)2D3 receptor (VDR) in this cell line. In quiescent, subconfluent cultures neither the treatment with 100 nM 1,25(OH)2D3 nor that with 50 microM forskolin influenced proliferation, while a significant increase was observed after incubation of the cells with 10% fetal bovine serum. Either cell number, determined on growing IMR-90 human fibroblasts after 48 or 72 h incubation with 100 nM 1,25(OH)2D3 or [3H]thymidine incorporation (24, 48 or 72 h incubation) significantly decreased, while protein content per cell increased. Northern blot analysis revealed the expression of the VDR gene, the VDR mRNA bands being prominent in 1,25(OH)2D3, serum or forskolin treated fibroblasts. VDR mRNA levels slightly decreased, when growing fibroblasts were exposed to 100 nM 1,25(OH)2D3 for 48 or 72 h.

Effect of 1,25-dihydroxyvitamin D3 on proliferation in senescent IMR-90 human fibroblasts.

The response of IMR-90 human fetal lung fibroblasts at high population doubling level (PDL > 42) to 1,25-dihydroxyvitamin D3[1,25(OH)2D3] was investigated to clarify whether some metabolic and molecular parameters of senescent cells are affected by the hormone treatment. Pyruvate kinase, lactate dehydrogenase and glucose-6-phosphate dehydrogenase activity significantly increased after treatment of confluent-phase cells with 10 nM 1,25(OH)2D3 for 24 h. Steroid specificity was established by the failure of 10 nM levels of 25-hydroxyvitamin D3 to affect the enzyme activities, while estradiol-17 beta and progesterone produced a slight increase in glucose-6-phosphate dehydrogenase and lactate dehydrogenase levels, respectively. 1,25(OH)2D3 also affected fibroblast proliferation, protein content/cell and DNA synthesis. The cell number significantly decreased after a 48 h incubation with 1,25(OH)2D3 at various concentrations (0.01-1 nM) when compared with control fibroblasts, while an increase in the protein content/cell was demonstrated. The same experiment, carried out by protracting the incubation with the hormone for 72 h, showed a similar trend, but 10 nM 1,25(OH)2D3 was also able to inhibit cell proliferation and to stimulate protein synthesis. The incorporation of [3H]thymidine into DNA increased after the treatment of high PDL fibroblasts with 0.01-1 nM of hormone for 48 h in comparison with controls.

1,25-dihydroxyvitamin D3 inhibits proliferation of IMR-90 human fibroblasts and stimulates pyruvate kinase activity in confluent-phase cells.

This study tested the hypothesis that 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) plays a role in regulating some aspects of metabolism in IMR-90 normal human fetal lung fibroblasts. Among the enzymes studied, only pyruvate kinase showed a significant increase after treatment of confluent-phase cells with 1,25(OH)2D3 at various concentrations (0.1-100 nM range) for 24 h. A parallel increase in lactate output was observed. Steroid specificity was established by the failure of 10 nM levels of 25-hydroxyvitamin D3, estradiol-17 beta and progesterone to affect pyruvate kinase activity. The determination of the time course of [3H]-2-deoxy-D-glucose transport indicated that the hormone did not influence the transmembrane transport system of D-glucose. The addition of the inhibitors cycloheximide and actinomycin D to the culture medium abolished, at least in part, the 1,25(OH)2D3 stimulation of pyruvate kinase activity, suggesting the probable dependence of the hormone effect on cellular RNA and protein synthesis. 1,25(OH)2D3 also affected fibroblast growth and DNA synthesis. Cell number significantly decreased after 2-5 days treatment with 10 nM hormone in comparison with control fibroblasts, and also the incorporation of [3H]thymidine into DNA decreased after treatment of the cells with 1 and 10 nM hormone for 48 h. In conclusion, these data demonstrate that 1,25(OH)2D3 stimulates pyruvate kinase activity in confluent-phase IMR-90 human fibroblasts by a mechanism probably dependent on de novo protein synthesis, and also affects cell growth and DNA synthesis in sub-confluent-phase cells.

Vitamin D--related modification of enzyme activities in synaptosomes and mitochondria isolated from rat cerebral cortex.

We previously demonstrated that feeding rats the Steenbock and Black rickets-inducing diet produces remarkable changes in the metabolic pattern of intestinal mucosa, kidney, liver, cerebral cortex and heart. We have now determined the levels of calcium, phosphorus and citrate in cerebral cortex and the activity of some enzymes in synaptosomes and cerebral cortex mitochondria of three rat groups: control (Group A), fed a vitamin D-deficient diet (Group B), fed a vitamin D-deficient diet and treated with 1,25-dihydroxyvitamin D3 (Group C). While calcium content increased in Groups B and C, phosphorus concentration increased only in Group C and citrate in Group B in comparison with control. The increase in acetylcholinesterase and citrate synthase registered in Group B was restored to control values by 1,25-dihydroxyvitamin D3 treatment, while, neither the decrease in cytochrome c oxidase, nor the increase in glucose-6-phosphate dehydrogenase, acid phosphatase and NADP+(-)isocitrate dehydrogenase observed in Group B were corrected by 1,25-dihydroxyvitamin D3 supply. Acyl phosphatase showed a remarkable increase in consequence of 1,25-dihydroxyvitamin D3 administration.
