Zinc content in selected tissues in streptozotocin-diabetic rats after maximal exercise.

The Zn metabolism in experimental diabetic rats after maximal exercise was investigated. Forty male wistar rats were used, weighing 240 +/- 10 g at the beginning of this experiment. The animals were assigned to one of four experimental groups (n = 10): control at rest (CR), control plus exercise (CE), diabetic at rest (DR), and diabetic plus exercise (DE). Experimental diabetes was produced by a single intraperitoneal injection of streptozotocin (STZ) (60 mg/kg). Thirty days after injection of streptozotocin, the animals of groups CE and DE were forced to acute exercise (swimming) until exhaustion. Glucose, rectal temperature (RT), pH, swimming time (ST), hematocrit (Hct), serum, and tissue (heart, liver, kidney, and muscle) Zn concentrations were measured. The streptozotocin treated animals used in the current experiment were diabetic. Increases in hepatic, renal, muscle, and serum levels Zn at rest and after exercise until exhaustion were found in normal and diabetic rats. ST decreased (-180%) in the diabetic rat group. In conclusion, the results of the present study indicate that STZ-induced diabetes was associated with altered tissue Zn concentration, both at rest and after exercise.

Effect of hypoxia and exercise on the distribution of 65Zn in rat muscles.

The aim of this study was to determine the influence of hypoxia and exercise in 65Zn distribution in various rat muscles. A total of 80 male wistar rats weighting 250 +/- 10 g were randomly separated into eight groups: four groups in air, two at rest, and the other two after exercise; four groups in hypoxia (3 days exposure), two at rest, and the other two after exercise. Two groups (n = 10) in air and two other groups under hypoxia were injected with 65Zn. The 65Zn uptake by blood, pH, and PaO2 decreased significantly during hypoxia. The Zn concentration increased significantly in serum, diaphragm, gastrocnemius, and heart Zn after exercise in air. After hypoxia, at res, Zn increased in serum and in diaphragm. After chronic hypoxia, at rest, radioactivity of 65Zn increased in heart, gastrocnemius, and bone, and it was similar in air condition after exercise with respect to rest. In both cases (air and hypoxia), the uptake of 65Zn by the tissues was higher at rest. After exercise in hypoxia, endogenous Zn decreased in heart and in diaphragm.

Vaginal reconstruction with bilateral island "extended" groin flaps: description of a personal technique.

We propose a vaginal reconstruction method using two island "extended" groin flaps designed over the iliac crests and in axis with the superficial circumflex iliac arteries. After a surgical revision of the pelvic organs and dissection of the area between the bladder and the rectum, the two flaps pass, extraperitoneally by way of the abdomen, through an incision made along the lateral margin of the rectus muscle and, from before to behind, through the fibrovascular lamina lateral to the bladder, and finally are placed in the area between the bladder and the rectum, where they are sutured. The technique has been applied to one patient and allowed reconstruction of a vaginal canal with very suitable dimensions and in order to allow sexual intercourse. This seems a safe technique in terms of flap vitality because the operation is carried out extraperitoneally. Finally, the appearance of the external genitalia has not been altered and residual suture wounds have been well hidden.

The effect of exercise and zinc supplement on the hematological parameters in rats.

This study evaluates the consequences of a session of intensive short-duration exercise and Zn supplementation on different hematological variables. Forty male Wistar rats were divided into four groups (n = 10): the first nonsupplemented, maintained at rest (R); the second nonsupplemented, undergoing exercise (E); the third supplemented with Zn, kept at rest (ZnR); and the fourth supplemented with Zn, undergoing exercise (ZnE). Zinc supplements (200 ppm) were given in drinking water. The exercise consisted of a single session of swimming until exhaustion. At rest, RBC, Hb, and Hto fell (p < 0.05), whereas red cell indices, MCV, and MCH rose (p < 0.05) in +ZnR compared with R; MCHC remained unchanged (ZnR vs R). After exercise, RBC, Hb, and Hto increased significantly in E and in ZnE compared with R and ZnR, respectively. In addition, RBC and Hb were lower (p < 0.01) in ZnE compared with E; however, MCV and MCH were higher (p < 0.05) in the group ZnE vs E. With respect to white blood cells--leukocytes (WBC), lymphocytes (LYMPH), and neutrophils (NEUT)--no significant differences were observed between groups at rest (ZnR vs R). WBC and LYMPH increased significantly in E with respect to the rest situation (E vs R), but this did not happen in supplemented animals (ZnE vs ZnR). Level of pH decreased after exercise both in E and in ZnE, but the fall was lower in the latter. We believe that a single session of swimming until exhaustion leads to an increase in RBC, Hb, and Hto without causing changes in MCV, MCH, and MCHC.(ABSTRACT TRUNCATED AT 250 WORDS)

Lower eyelid reconstruction by multiple subcutaneous pedicle flaps: a new method.

In this paper we present an original technique for the reconstruction of the full thickness of the lower eyelid after excision of neoplasms. The method is based on the mobilisation of multiple pedicle subcutaneous flaps outlined in the same eyelid and supported by a chondro-mucosal graft. This technique is used for central and off-central losses which involve the outer rim or up to 75% of the width of the whole eyelid. This method is an alternative to the Hughes (1945), Mustardé (1981), Tenzel and Stewart (1978) and McGregor (1973) techniques which we have previously used for this type of reconstruction. The follow-up is satisfactory in terms of the oncological, functional and aesthetic aspects, which represent an inseparable triad in reconstructive eyelid surgery.

Changes on plasmatic and erythrocytic magnesium levels after high-intensity exercises in men.

The aim of this paper was investigate the effects of high-intensity exercise on plasmatic and erythrocytic Mg levels in men. Twelve normal male volunteers participated in this study. The tests were performed on a cycle-ergometer (Monark). The exercise programs consisted of: 1) triangular progressive test (TPT); 2) interval endurance test (IET), of 45 min duration; and 3) maximal subtained test (MST), consisting of a maximal level, at 100% of MTP, for at least 7 min. The percent change in plasma volume (%PV) after exercise was calculated. Magnesium levels in plasma and erythrocytes were measured by atomic absorption spectrophotometry. No significant differences in the absolute Mg concentration of plasma were noted for the three maximal exercises. The percentage increase in plasma Mg concentrations was about 5-7%; however, percent decrease of PV was more important (13-16%). Erythrocytic Mg levels were increased after all maximal exercises (TPT, IET, and MST), although absolute Mg concentrations were not significantly different.

Calcitonin and metabolic alkalosis.

Calcitonin is used in the treatment of osteoporosis and several adverse effects, such as rash, antibody presence, hypocalcemia, etc have been reported with the therapeutic use of large quantities of this hormone. The results found in this paper show that 7 h after administration of large doses of calcitonin to osteoporotic patients on hormone treatment, the acid-base status shifts to metabolic alkalosis. This represents another adverse effect of the use of pharmacological doses of calcitonin.

Effect of swimming to exhaustion, at low temperatures, on serum Zn, Cu, Mg and Ca in rats.

In order to study the response of plasma trace minerals during exercise in cold water at varying temperatures, three groups of ten male Wistar rats each were forced to swim until exhaustion in water at different constant temperatures (10, 20 and 32 degrees C), while another 3 groups were kept at rest in water at the same temperatures and another group at rest out of the water, at room temperature (21 degrees C). Rectal temperature (RT) was measured before and after the exercise. Samples of arterial blood were obtained under anaesthesia (pentobarbital) from the abdominal aorta after the exercise, and serum concentrations of Cu, Zn, Mg and Ca were determined by flame atomic absorption spectrophotometry (FAAS). The swimming time (ST) decreased in cold water, the values obtained at 10 and 20 degrees C being 3.3% and 8.5%, respectively, of those observed at 32 degrees C. The RT of the animals swimming at 32 degrees C was 32.5 +/- 1.8 degrees C (mean +/- SD) and fell to 25.0 +/- 2.0 and to 19.1 +/- 1.5 degrees C in those swimming, respectively, at 20 and 10 degrees C. The concentrations of Mg, Cu and Ca were significantly higher (+50%, +36% and +7%, respectively) in animals subjected to exercise with respect to the control groups in water, although there were no differences among the three groups. On the contrary, Zn rose progressively with the increase of WT (+32% at 10 degrees C, +60% at 20 degrees C and +76% at 32 degrees C).(ABSTRACT TRUNCATED AT 250 WORDS)

Changes of plasma zinc and copper at various times of swimming until exhaustion, in the rat.

To characterise the response in the plasma concentration of Zn a total of 60 male Wistar rats (250 g) were divided into six groups of ten rats each. One group did not exercise; the five other groups swam at constant temperature (32/34 degrees C) and were respectively studied after 30, 60, 90, 120 min and at exhaustion. After anesthesia, rectal temperature (RT) was measured and arterial blood was obtained from the abdominal aorta. Acidbase balance, arterial blood gases and plasma concentration of Zn, Cu, Mg, Ca and Hb were measured. The swimming time at exhaustion was (mean +/- SD) 284 +/- 66 min. RT only changed after exhaustion. (H+) increased significantly at 30 min then did not change until exhaustion. (Hb) increased slightly at 90 and 120 min of swimming and after exhaustion. No changes of Mg and Ca except at exhaustion were observed (+7 and +50% respectively). On the other hand, Cu (30 min = +16%; 60 min = +29% exhaustion = +36%) and Zn (30 min = +26%; 60 min = +39%; 120 min = +66% and exhaustion = +76%) increased significantly through the two first hours of exercise. Haemoconcentration, acidosis and RT did not explain the progressive increase of Zn, which seems to be time-dependent and probably related to some degree of stress.

Prevention of stress-induced involution of the thymus in rats by substance P (SP1-11) and its N-terminal fragment SP1-4.

Selye found that in response to different stressors the body reacts with a characteristic stress syndrome: adrenal enlargement, gastrointestinal ulcera, and thymicolymphatic involution. In this paper we demonstrate that i.p. injected Substance P (SP) can prevent stress-induced involution of thymus in Wistar rats. This protecting effect on the immune system of SP, in addition to the "normalizing" effects of this peptide on stress-induced disorders in vegetative and central functions first described by Oehme and co-workers and Hecht and co-workers is a new hint to the function of SP as an "anti-stress" agent. The mode of action of the anti-stress effect of SP in adrenals as well as in the thymus is discussed. It is suggested that the anti-stress effect of SP1-11 and its N-terminal fragment SP1-4 is mediated by specific receptors in adrenals and/or thymus which are different from the so called SP-P and SP-E receptors.

A site-specific mutation within the active site of ribulose-1,5-bisphosphate carboxylase of Rhodospirillum rubrum.

In vitro mutagenic techniques have generated an asp-->glu substitution at residue 198 adjacent to the carbamate-divalent metal ion binding site of Rhodospirillum rubrum ribulose 1,5-bisphosphate carboxylase. A single C-->A nucleotide change in the coding strand created the mutant and introduced a new EcoRI restriction site on the expression plasmid pRR2119. Although the carboxylase:oxygenase ratio remained the same, the mutant enzyme had slightly altered kinetic properties. The e.p.r. spectra of the quaternary complexes enzyme.activator carbamate.Mn.2-carboxyarabinitol 1,5-bisphosphate and enzyme.activator carbamate.Mn.4-carboxyarabinitol 1,5-bisphosphate for mutant and wild-type enzymes were different, indicating that the metal ion was in a slightly altered environment. These findings are consistent with the hypothesis that, besides the carbamate at lys 201, the carboxyl group of asp 198 contributes to the formation of the divalent metal ion binding site.

The Mongolian spot: a study of ethnic differences and a literature review.

Four hundred thirty-seven consecutively born full-term neonates, delivered at Jackson Memorial Hospital, were examined for the presence of mongolian spots (MS). The spots were found in 96% of the Negro children, 46% of the Hispanic children, 9.5% of the Caucasian children, and in both of the Asian children in the series. The sacro-gluteal region was the most frequent site of pigmentation, with the shoulders next in frequency. Almost all the spots on the extremities were located on the extensor surfaces. The color was most frequently blue-green, but was also commonly greenish-blue, blue-gray, or brown. In a concurrent review of 124 newborn records, MS was not described by house officers in any, although the probability of its occurrence would have been approximately 90 cases, based on the results of the study. A comprehensive review of the history of the description of MS and theories of its development, as well as a review of the clinical aspects, is presented.

[Conformal radiotherapy of brain tumors].

Conformal irradiation of brain tumours is based on the three-dimensional reconstruction of the targeted volumes and at-risk organ images, the three-dimensional calculation of the dose distribution and a treatment device (immobilisation, beam energy, collimation, etc.) adapted to the high precision required by the procedure. Each step requires an appropriate methodology and a quality insurance program. Specific difficulties in brain tumour management are related to GTV and CTV definition depending upon the histological type, the quality of the surgical resection and the medical team. Clinical studies have reported dose escalation trials, mostly in high-grade gliomas and tumours at the base of the skull. Clinical data are now providing a better knowledge of the tolerance of normal tissues. As for small tumours, the implementation of beam intensity modulation is likely to narrow the gap between conformal and stereotaxic radiotherapy.

Changes in serum lipid and lipoprotein measures after maximal exercise in men.

Normolipidaemic, healthy male volunteers (age range 20-23 years) were studied after a session of three maximal exercises and serum lipid and lipoprotein concentrations were measured. The exercise programmes, one of which was performed each week, consisted of a triangular progressive test (TPT), a rectangular sustained test (RST) and an interval endurance test (IET). There was a significant (P less than 0.05) increase of 14% in serum total cholesterol concentrations after TPT, 9.6% after RST and 14% after IET. Serum triglyceride concentrations increased by 17% after TPT, 5.7% after RST and 8.2% after IET, but increases were not significant. High-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C), and apolipoprotein (A-I, A-II) concentrations rose significantly (P less than 0.05) after TPT. There were also insignificant increases in AI, AII and LDL-C after RST and IET, whereas HDL-C rose significantly (P less than 0.05) after IET. It is concluded that the three maximal exercises had similar metabolic effects which were independent of the exercise duration.
