A comparative study of pseudorabies virus (PRV) strains with defects in thymidine kinase and glycoprotein genes.

In the course of two experiments, an examination was made of the virulence and neuroinvasiveness for pigs of two pseudorabies virus mutants (strain 6C2TK(-), with a defect in thymidine kinase (TK) function; and strain 6C2TK(-), gI(-)/gE(-), with defects in TK and glycoproteins I and E) and of the wild-type parent strain (86/27V). At various times after intranasal inoculation, pigs were killed and samples of tonsil, lung and different levels of the trigeminal and olfactory nervous pathways were examined by methods that included viral isolation, polymerase chain reaction assay and immunohistochemistry. Both mutant viruses were of reduced virulence, as indicated by no more than moderate clinical signs and lesions, and only sporadic isolation of virus; moreover, unlike the wild-type parent strain, the mutant viruses were not reactivated from the latent state by corticosteroid treatment. In addition, migration of the mutant strains to the central nervous system (olfactory and trigeminal nervous pathways) was reduced as compared with that of the wild-type strain. Thus, mutations in the genes encoding the TK enzyme and the gI/gE complex were associated with reduced virulence, reduced replication in peripheral target tissues, and reduced migration to the olfactory and trigeminal pathways.

Histological and immunohistochemical study of the bronchial stump with flap coverage in an animal model.

OBJECTIVE: The purpose of our study was to evaluate the healing process of the bronchial stump after pneumonectomy reinforced with different pedicled flaps in an animal model. The specimens were analyzed by means of histology and immunohistochemistry. MATERIALS AND METHODS: We have considered 45 New Zealand White male rabbits that underwent a left pneumonectomy under general anesthesia. Nine animals had no bronchial coverage and represented the controls. The other 36 rabbits were divided into three groups of 12 and had bronchial coverage with either diaphragmatic, intercostal or pericardial flaps. The histological examinations were performed on the animals sacrificed 7, 14 and 30 days after surgery. Immunohistochemical analyses were done on the specimens on postoperative day 7 and 14. On postoperative day 7, the specimens were examined for expression of proliferating cell nuclear antibody (PCNA) expression. On postoperative day 14, neoangiogenesis was measured by CD31 expression. The measurements of antibody expression were done with a computer-assisted morphometric count and analyzed with the t test. RESULTS: On postoperative day 14, standard histology showed more evident neoangiogenesis in the bronchial stump specimens covered with intercostal and diaphragmatic flaps compared to pericardial flaps and controls. The immunohistochemical evaluation of PCNA by morphometric computer-assisted analysis did not show any statistically significant differences among the groups. The CD31 morphometric count revealed a higher and statistically significant antibody expression in muscular flaps compared to pericardial flaps and controls. CONCLUSIONS: Our study showed that bronchial coverage with a pedicled muscular flaps promotes the production of new vessels and gives the possibilities to optimize the healing process of a bronchial stump after pneumonectomy.

[Juvenile rheumatoid arthritis: the role of the TMJ and stomatognathic system].

Juvenile chronic arthritis (j.c.a) often affects the maxillo-facial complex over all in relation to jaw and temporo-mandibular joint (TMJ). In this work 35 j.c.a. patients (aged 6 to 16 years) have been selected and studied in order to detect functional and morphostructural involvement of stomatognathic apparatus and TMJ. The involvement of these structures is appeared significantly increased during j.c.a. also in patients without clinic symptoms referable to a stomatognathic apparatus pathology. Therefore an early diagnosis as well as a preventive orthognatodontic treatment are necessary to reduce long term effects on the stomatognathic apparatus multifunctions.

[Hohmann's operation in the treatment of humeral epitrochleitis].

7 cases of epicondylitis surgically treated by a Hohmann operation and perforations were checked. These were the only cases of epicondylitis to be operated at the G. Pini Orthopaedic Institute, Milan, over a decade. Careful evaluation of the results showed complete success in all cases. The association of the Hohmann operation and perforations is therefore to be recommended in the rare forms of epicondylitis which do not respond to nonsurgical treatment.

[Osteogenesis imperfecta and calcitonin. Considerations on 10 years of clinical experimentation].

Salmon and porcine calcitonin has been used in the drug treatment of Osteogenesis Imperfecta for ten years. The evaluation of its therapeutic efficacy is complex and inaccurate because of difficulty in clinical and pathogenetic classification of the disease and impossibility to standardize and evaluate the patients. Calcitonin therapy showed no capacity to modify the degree of osteoporosis and the incidence of pathological fracture and bone deformities, because it does not affect the underlying biochemical defect. Calcitonin may be useful only in selected patients and in addition to physiotherapeutic, orthopaedic and surgical measures.

[Radiographic evaluation of the hip in children. Proposal for standardizing findings].

Considering that the complexity of the numerous X-ray techniques proposed for study of the child hip are ill fitted to standardization of the investigation, straightforward frontal and axial projections are proposed for this purpose. These are carried out with a special technique to enable subsequent controls to be reproduced and so as to obtain, with the aid of precise measurements, a complete morphological picture of the joint in question. Possibilities of error and exposure to ionizing radiations are cut to the minimum. These ideas are all the more indispensable today in the presence of problems of selecting children by peripheral units and centres responsible for specific conditions, even though techniques of maximum simplicity and safety are used.
