Effects of vitamin supply on spontaneous and chemically induced mutagenesis in human cells.

Chromosome aberrations in donor peripheral blood lymphocyte culture were evaluated before, 14 and 30 days after treatment with a vitamin/mineral complex. Treatment with the complex had no effect on spontaneous level of aberrant cells. The number of chromosome aberrations induced by dioxidine or cadmium chloride in vitro at the G2 stage decreased on days 14 and 30 of treatment with the complex.

Damage to cell DNA in the bone marrow and testes of mice with experimental trichinosis.

Metabolites of Trichinella spiralis produced genotoxic and cytotoxic effects on somatic and generative cells of the host organism. They increased the number of single-chain breaks, alkaline-labile sites in nuclear DNA, and count of apoptotic cells in the bone marrow and testes of infected mice. These effects depended on the stage of parasite development in the host organism and became more pronounced with increasing invasion intensity.

Effect of Betula bark extract on spontaneous and induced mutagenesis in mice.

Study was performed by counting cells with chromosomal aberrations in C57Bl/6 mice. Bark dry extract was given perorally in doses of 50, 150, 450, and 1500 mg/kg. Mutagens dioxidine and cyclophosphamide were injected intraperitoneally in doses of 200 and 20 mg/kg, respectively. Bark dry extract in doses of 150 and 1500 mg/kg did not possess cytogenetic activity. Bark dry extract in doses of 50, 150, and 450 mg/kg significantly decreased the cytogenetic effect of mutagens under various regimens of treatment with the preparation (single combined administration, 5-day pretreatment, and 5-day combined administration). Our results indicate that bark dry extract possesses antimutagenic properties.

Antimutagenic activity of lipidovit.

Antimutagenic properties of Lipidovit produced from the biomass of Blakeslea trispora fungi was studied by its effect on induction of chromosome aberrations by chemical mutagens dioxidine and cyclophosphamide in mouse bone marrow cells. Antimutagenic activity of Lipidovit depended on the scheme of treatment. It was maximum during pretreatment of animals (5 day) or administration in combination with mutagens (5 days). The preparation was ineffective after single administration in combination with mutagens. Lipidovit exhibited no comutagenic properties.

Total antioxidant capacity of blood plasma from healthy donors receiving vitamin and mineral complex.

The total antioxidant capacity (TEAC assay) of blood plasma from healthy donors was studied before and after 2- and 4-week consumption of the vitamin and mineral complex containing the major antioxidant vitamins and cofactors of antioxidant enzymes. Before the treatment the donors were divided into 2 subgroups with initially high and low total antioxidant capacity. After 2-week course of vitamin and mineral complex the total antioxidant capacity considerably decreased in group 1 donors, but increased in group 2 donors. After 4-week course this index increased in donors of both subgroups and slightly surpassed the baseline level typical of individuals with high total antioxidant capacity.

Study of mutagenic activity of dioxidine by the polyorgan micronuclear method.

Antibacterial preparation dioxidine administered four times in doses of 10, 100, and 300 mg/kg increased the incidence of micronucleated cells in the bone marrow, lungs, and large intestine of mice. Bone marrow cells were most sensitive, while cells of the lungs and large intestine exhibited lower sensitivity to the cytogenetic effect of dioxidine.

In vitro genotoxic and cytotoxic effects of protein somatic products from helminths on donor blood lymphocytes.

Protein somatic products of adult helminths Hymenolepis nana and Toxocara canis and secretory-excretory somatic product of Trichinella spiralis larvae in vitro produced genotoxic and cytotoxic effects on donor blood lymphocytes, which manifested in accumulation of single-chain breaks, alkaline-labile sites in nuclear DNA, and apoptotic cells. This effect depended on the concentration of parasitic protein products during coculturing.

[Oxidative stress in the pathogenesis of anti-phospholipid syndrome].

Summarized in the paper are the data describing the significance of the oxidative stress within the pathogenesis of anti-phospholipid syndrome (APS). The mechanisms related with formation of active oxygen metabolites in human body, triggers of the oxidative stress as well as its involvement in the APS pathogenesis (i.e. in the aPL synthesis), in the endothelial malfunction development and in the development of pro-coagulation impairments within the hemostasis system leading to thrombosis are in the focus of attention. Approaches to treatment schemes of vascular disorders in APS comprising the oxidative stress correction are suggested.

Antimutagenic and antioxidant activities of crown compounds in comparison with the effects of garlic extract.

Antimutagenic activity of N-carboxyalkyl derivatives of aza- and benzoazacrown compounds was revealed and antimutagenic activity of garlic extract was confirmed. Specific genoprotective effect of crown compounds towards the effects of various mutagens was demonstrated. The antimutagenic effect of these compounds was not realized via antioxidant mechanisms, while the protective effect of garlic extract was associated with its antioxidant and reparative activities.

Effect of vitamin therapy on sensitivity of peripheral blood lymphocytes to clastogenic effect of mutagens in vitro.

Spontaneous and in vitro induced (with 0.01 and 0.1 mg/ml dioxidine and 0.1 and 1.0 U/ml bleomycin) chromosome aberrations were counted in cultured peripheral blood cells from 11 donors before and after 2-week therapy with a vitamin complex. The complex contained the major vitamins in doses not surpassing the recommended daily doses. Vitamins had no effect on spontaneous mutations, but increased cell resistance to clastogenic effects of dioxidine in a concentration of 0.1, but not 0.01 mg/ml. Cell sensitivity to bleomycin notably increased after vitamin therapy in some donors and decreased in others, the mean parameters in the group remained virtually unchanged.

[Effects of biologically active food additives with different contents of vitamins on the vitamin status in humans].

The comparative study of influence of two biologically active food additives with the different contents of vitamins is carried out: a drink "Zolotoi Shar", the dose of vitamins in which makes 50-90% from recommended daily consumption, and "Vitabalance 2000", the contents of vitamins in which at 2-17 of time exceeds need of organism. The use of both additives within 3 weeks resulted in increase of levels of vitamins C, A, E, B2, B6 and carotenoids in blood serum. However if in case of consumption of a drink an authentic level was reached only for vitamin C and beta-carotene, in a case "Vitabalance 2000" for all investigated vitamins except vitamin A. Thus, if the consumption of a drink has lowered frequency of deficiency of 3-4 vitamins, but has not allowed to liquidate it completely, in case of "Vitabalance 2000" consumption the simultaneous deficiency 3-4 vitamins. The received data allow to believe the biologically active food additives containing vitamins in amounts exceeding recommended consumption, are convenient for fast liquidation of hypovitaminoses, and the preparations containing vitamins in doses making 30-50% from need of organism, are acceptable for daily filling of insufficient consumption of vitamins with a usual diet for a long time.

Antimutagenic activity of afobazole in various regimens of treatment.

The influence of a new 2-mercaptobenzimidazole derivative afobazole on cytogenetic effects of dioxidine and cyclophosphamide was studied by counting chromosome aberrations in bone marrow cells of C57Bl/6 mice. Afobazole (1-100 mg/kg perorally) exhibited antimutagenic activity determined by its antioxidant properties. This activity depended on the dose and treatment schedule.
