The profiles of enterotoxin genes in Staphylococcus aureus from nasal carriers.

AIMS: To evaluate the occurrence of enterotoxin genes in Staphylococcus aureus recovered from nasal carriers. METHODS AND RESULTS: Eighty S. aureus strains were tested for the presence of 17 new enterotoxin genes using multiplex-PCR. Sixty-one isolates were found to carry enterotoxin genes. The majority of the enterotoxigenic isolates carried enterotoxin gene cluster (egc) genes, namely seg, sei, sem, sen and seo. The egc type containing the seu gene was found in 19 of the 47 isolates with egc-like genes. Interestingly, no seu-containing egc coexisted with sec and sel, as was the case for a considerable portion of the isolates carrying a seu-negative egc. The tst gene was detected in two isolates carrying sec and sel only and in eight isolates carrying seu, but not in the isolates containing the seu-negative egc type. CONCLUSIONS: The genes forming an egc were found to be predominant in S. aureus from nasal carriers. The coexistence of a seu-positive egc with tst in contrast to an egc lacking the seu gene apparently is not associated with the presence of tst and can reflect a difference between these gene groupings. SIGNIFICANCE AND IMPACT OF THE STUDY: The egc types carried by the analysed isolates seem to have an influence on the distribution of other genes located on staphylococcal pathogenicity islands, which may modulate the repertoire of virulence factors carried by a single S. aureus strain.

Biodegradability of organic by-products after natural organic matter oxidation with ClO2--case study.

Apart from well-known chlorites and chlorates, chlorine dioxide also generates easily biodegradable carbonyl compounds and short chain carboxylic acids during water disinfection. The main goal of the presented study was to examine the influence of natural organic matter (NOM) oxidation with chlorine dioxide, on the quantity as well as the quality of formed biodegradable by-products. In the experiments conducted at the pilot plant the sand filtered water (MWI) and ozonated/biofiltrated water (BAF) were oxidised with ClO2. The amount of BDOC formed as a result of the oxidation of both waters with ClO2 was compared. The results showed considerable differences in formation of ClO2 oxidation by-products between non-ozonated and ozonated/biofiltered waters. The disinfection of ozonated/biofiltrated water with ClO2 generated comparable amounts of aldehydes and much higher amounts of carboxylic acids than ClO2 oxidation of sand filtered water. These findings are essential for waterworks with ozonation/biofiltration units and ClO2 disinfection implemented.

Reactivity of natural organic matter fractions with chlorine dioxide and ozone.

The effects of ozone and chlorine dioxide on the aquatic natural organic matter (NOM) were studied. The natural as well as oxidised organic matter in aerated and sand filtered water were fractionated using XAD, anion-exchange, and cation-exchange resins procedure into humic acid, hydrophobic acids and neutrals, and hydrophilic acids, bases and neutrals. The main NOM components were hydrophobic, while oxidation with both ozone and chlorine dioxide increased the proportion of hydrophilic fractions. High-pressure size exclusion chromatography with UV-254 nm and UV-220 nm detection was used to determine the differences between molecular weight distribution of natural and oxidised organic matter fractions. The main purpose of this paper was to compare the reactivity of individual NOM fractions with oxidants in order to compare the productivity of biodegradable by-products after oxidation with chlorine dioxide and ozone. The quantity as well as the quality of by-products were analysed by means of ion and gas chromatography.

Formation of aldehydes upon ClO2 disinfection.

Carbonyl compounds are considered to be the most common ozonation by-products. Apart from well-known chlorites and chlorates, chlorine dioxide (ClO(2)) also generates organic by-products. The goal of this paper is an investigation into the influence of disinfection with ClO(2) on the aldehydes formation. Three types of waters were disinfected with different doses of ClO(2) at the various pH values. The results of size exclusion chromatography show that the reaction of ClO(2) with humic fraction of natural organic matter proceeds mainly on an aromatic part of the molecules. The results obtained indicate that the level of carbonyls concentration can significantly increase with the time of ClO(2) reaction with carbonyl precursors in treated water. There is no noticeable correlation between quantity of aldehydes and pH value of disinfected water. The range of "productivity" of aldehydes in water treated with chlorine dioxide is similar to the range estimated for the carbonyls formed upon ozonation.

Investigation of carbonyl compounds in bottled waters from Poland.

Poly(ethylene terephtalate) (PET) bottles are commonly used for storing mineral water. The migration of carbonyl compounds from PET bottles into mineral water was observed. Carbonation of water, sunlight and high temperature enhance the process of migration. Formaldehyde, acetaldehyde and acetone were the most important carbonyls identified in series of bottled water samples. The concentration of carbonyls can change depending on the time of storage as well as storage conditions. It was identified particularly high concentration of acetaldehyde (more than 100 microg 1(-1)) in samples of mineral water saturated with CO2 gas.

Potentiated antitumor effects of interleukin 12 and interferon alpha against B16F10 melanoma in mice.

Systemic administration of cytokines has not found broad application in cancer immunotherapy due to its toxicity and lack of effectiveness in a broad spectrum of tumors. Among the most promising cytokines used often in pre-clinical and clinical trials are interferon alpha and interleukin 12. We have shown in our study that combining IL-12 with IFN-alpha in a dose which alone does not show antitumor activity results in potentiated antitumor effects without inducing toxicity.

Low-carbon silica sorbents for solid-phase extraction.

The applicability of silica gels, modified with cyclic organosiloxanes, in solid-phase extraction (SPE) has been tested. Surface characteristics of the adsorbents prepared are determined by: elemental analysis, 29Si cross-polarisation magic angle spinning nuclear magnetic resonance spectroscopy (29Si CP MAS NMR) and diffuse reflectance infrared Fourier transform (DRIFT) spectroscopy. The sorbents with low carbon contents are used for extraction of chlorinated pesticides, polychlorinated biphenyls and nitro-compounds from water. The properties of the sorbents are compared with commercial C18 ones. It is shown that the low-carbon silica SPE sorbents can ensure satisfactory recoveries and reveal some selectivity.

Potentiation of the anti-tumour effects of Photofrin-based photodynamic therapy by localized treatment with G-CSF.

Photofrin-based photodynamic therapy (PDT) has recently been approved for palliative and curative purposes in cancer patients. It has been demonstrated that neutrophils are indispensable for its anti-tumour effectiveness. We decided to evaluate the extent of the anti-tumour effectiveness of PDT combined with administration of granulocyte colony-stimulating factor (G-CSF) as well as the influence of Photofrin and G-CSF on the myelopoiesis and functional activity of neutrophils in mice. An intensive treatment with G-CSF significantly potentiated anti-tumour effectiveness of Photofrin-based PDT resulting in a reduction of tumour growth and prolongation of the survival time of mice bearing two different tumours: colon-26 and Lewis lung carcinoma. Moreover, 33% of C-26-bearing mice were completely cured of their tumours after combined therapy and developed a specific and long-lasting immunity. The tumours treated with both agents contained more infiltrating neutrophils and apoptotic cells then tumours treated with either G-CSF or PDT only. Importantly, simultaneous administration of Photofrin and G-CSF stimulated bone marrow and spleen myelopoiesis that resulted in an increased number of neutrophils demonstrating functional characteristics of activation. Potentiated anti-tumour effects of Photofrin-based PDT combined with G-CSF observed in two murine tumour models suggest that clinical trials using this tumour therapy protocol would be worth pursuing.

[Quality of life in children and adolescent with end-stage renal disease treated with dialysis and kidney transplantation].

This study investigates the quality of life of 139 patients remaining on alternative therapies for end-stage renal disease. Data from self-report questionnaires concerning physical activity, physical and social well-being, signs of depression and general assessment of situation, and "hope for future" were compared. Results indicate that self-assessment of quality of life (physical activity, physical and social well-being) among transplant patients is the best compared to both dialysis groups. These differences sustained in spite of deterioration of general health state with time. There were no differences between dialysis groups in terms of evaluated parameters. Overall results of quality of life assessment expressed by patients treated with hemodialysis seem to slightly improve during treatment.

The quality of heart valve tissue for homograft transplantation.

BACKGROUND AND AIM OF THE STUDY: The results of heart valve replacement surgery may be improved by refining surgical techniques and/or developing new heart valve transplants. The aim of this study was to examine the effect of age on the presence of cholesterol clefts and lipid deposits in the cusp base and sinus wall of aortic homografts. METHODS: Seventy-one valves were obtained at autopsy from donors (aged 15- 40 years) with no history of illness or evidence of serious illness. Trauma was the predominant cause of death among patients. The valves were examined using an osmium-vaporization technique. RESULTS: Osmiophilic deposits were detected in the cusp base in 28 cases (42%), and in the sinus wall in 49 cases (69%). Cholesterol crystals in the cusp base were found in 16 cases (24%), and sinus wall cholesterol clefts in 26 cases (38%). The model-predicted probability of cusp base lipid deposits existing was 76% in 40-year-old donors, 36% in 30-years-olds, and 11% in 20-year-olds; the probability of cusp base cholesterol clefts existing was 45%, 21% and 8% in these age groups, respectively. The influences of immunological reactions, biochemical changes (centers of calcification) and acceleration of atherosclerotic processes are discussed. CONCLUSION: The microscopic study of heart valves demonstrated the presence of lipid deposits in subjects of an unexpectedly young age. Among our study material, 58% of valves obtained from donors aged 11-40 years were unsuitable for transplantation. Our results confirmed the need for macroscopic inspection of heart valves before their being transplanted.

Lovastatin and tumor necrosis factor-alpha exhibit potentiated antitumor effects against Ha-ras-transformed murine tumor via inhibition of tumor-induced angiogenesis.

Lovastatin, a drug commonly used in the treatment of hypercholesterolemia, has previously been reported to exert potentiated antitumor activity when combined with either tumor necrosis factor-alpha (TNF-alpha), cisplatin or doxorubicin in a melanoma model in mice. Since lovastatin interferes with the function of ras oncogene-encoded (Ras) proteins, we have investigated the antitumor activity of lovastatin and TNF-alpha using a Ha-ras-transformed murine tumor model. In in vitro studies, lovastatin inhibited the growth of cells transformed with Ha-ras oncogene (Ras-3T3 and HBL100-ras cells) more effectively than control NIH-3T3 and HBL100-neo cells. In in vivo experiments, the Ras-3T3 tumor demonstrated significantly increased sensitivity to combined treatment with both lovastatin (50 mg/kg) and TNF-alpha (1 microg/day) compared with either agent alone. Combined treatment with both agents also resulted in greater inhibition of blood-vessel formation. Ras-3T3 tumor cells produced increased amounts of vascular endothelial growth factor (VEGF) and lovastatin effectively suppressed VEGF production by these cells. Our results suggest that lovastatin increases antitumor activity of TNF-alpha against tumor cells transformed with v-Ha-ras oncogene via inhibition of tumor-induced blood-vessel formation.

The potential role of brain asymmetry in the development of adolescent idiopathic scoliosis: a hypothesis.

BACKGROUND: The size asymmetry of cerebral hemispheres may predispose to head tilt and asymmetric blocking of the zygapophysial joints, potentially leading to the development of compensatory curvatures in the lower segments of the spine. OBJECTIVE: To analyze the effects of spinal manipulation, maintained by an exercise program, on the progression of idiopathic adolescent scoliosis in 2 children aged 6 and 10. CLINICAL FEATURE: The scoliosis found was 16 and 60 degrees. INTERVENTION AND OUTCOME: For diagnosis and monitoring of therapy, we recorded qualitative parameters of shoulder asymmetry, axillary line asymmetry, and scapular angle position. Manual treatment consisted of the examinations of all sliding motion in zygapophysial joints and both sacroiliac joints and removing the limitations of the sliding motions according to the method of Karel Lewit. The treatment procedure consisted of 3 or 4 manipulations within 17 months and an exercise program. The manipulation effects were maintained by the exercise program. The exercises were done in 2 or 3 sessions weekly for a year. In both patients we observed that scoliosis decompensation was successfully stopped and the effects of the correction persisted for 10 years. CONCLUSION: Brain and head asymmetry may be only a transient state, predisposing to asymmetric blocking at the atlanto-occipital level. Removal of blocking may prevent curve progression in children who had adolescent idiopathic scoliosis. The manipulative therapy may also have a promising effect on retarding curve progression when used in skeletally immature patient.

Influence of secretin and caerulein on acid phosphatase activity in human saliva.

Experimental studies have demonstrated a structural and functional correlation between the pancreas and salivary glands. Therefore, one may suppose that caerulein plus secretin, used in functional exploration of the exocrine pancreas, could exert an influence on acid phosphatase activity (AcP) and its isoenzymes (tartrate sensitive-TsAcP, tartrate resistant-TRAP acid phosphatases) in whole saliva in humans. The current study was conducted in 14 asymptomatic volunteers (six woman and eight men, mean age 34, range 27-45). Salivary secretions were collected under basal conditions (intravenous infusion of 0.15 mol l-1 NaCl) and during intravenous infusion of secretin plus caerulein at different doses. Stimulation with secretin (50 mKE kg-1 min-1) and caerulein (2.5 ng kg-1 min-1) resulted in a marked and rapid decrease of salivary flow rate. TRAP activity decreased during the first 20 min of hormonal infusion (1.09 +/- 0.19 mU ml-1 vs. 1.93 +/- 0.18 mU ml-1, 1.16 +/- 0.12 mU ml-1 vs. 1.86 +/- 0.17 mU ml-1, p < 0.05). AcP and TsAcP activity during intravenous infusion of secretin plus caerulein remained unchanged. These results point to a wider than expected participation of the digestive system in hormonal control of biochemical processes taking place inside the oral cavity.

Interaction of M1 and M2 isozymes pyruvate kinase from human tissues with phospholipids.

The effect of pH and the presence of FBP on the interaction of skeletal muscle (PK-M1) and kidney or tumor meningioma (PK-M2) pyruvate kinase with the phospholipids liposomes were investigated by ultracentrifugation and steady-state kinetics and were compared with those results obtained using the bovine heart (PK-M1) isoenzyme which we previously studied. Pyruvate kinase specific activity increases upon the interaction with liposomes. The activation is specifically sensitive to presence of phosphatidylserine (PS) in liposomes. Liposomes made of phosphatidylcholine + phosphatidylserine mixture are good adsorptive systems for both human and bovine of M-type isozymes at low ionic strength. Interaction of PK-M1 with PS liposomes results in the change of Vmax and K(m) values for PEP without marked effect on Hill coefficients. Addition of PS liposomes to PK-M2 induces hyperbolic saturation curves for PEP.

G-CSF prevents the suppression of bone marrow hematopoiesis induced by IL-12 and augments its antitumor activity in a melanoma model in mice.

BACKGROUND: IL-12 has been successfully used in experimental tumor therapy. However, administration of this cytokine induces dose-dependent suppression of hematopoiesis that could potentially limit its use in clinical trials. We decided to examine whether the myelosuppressive activity of IL-12 could be corrected by the administration of G-CSF. MATERIALS AND METHODS: In the initial experiments the influence of IL-12 and/or G-CSF on bone marrow and spleen GM-CFC was evaluated. To examine whether C-CSF could influence the antitumor activity of IL-12 the combination therapy with these agents was carried out starting on day seven following inoculation of melanoma MmB16 cells into the footpads of B6D2F1 mice. To obtain insight into the mechanism of the observed augmented antitumor activity of the combination therapy with IL-12 and G-CSF, the influence of these cytokines on macrophage activity (cytotoxicity and nitric oxide release) was analyzed. RESULTS: In accord with our expectations, the application of G-CSF partially prevented the suppression of bone marrow myelopoiesis in IL-12 treated mice. Unexpectedly, G-CSF also showed potentiation of antitumor effects of IL-12 in this melanoma model. The augmented antitumor activity of combined IL-12/G-CSF immunotherapy could result from the enhanced stimulation of macrophage NO production and cytotoxicity. CONCLUSION: The simultaneous administration of IL-12 and G-CSF partially prevented suppression of bone marrow myelopoiesis in IL-12-treated mice. Moreover, treatment with these cytokines also results in potentiated antitumor effects in a murine melanoma model.

Granulocyte-macrophage colony-stimulating factor potentiates antitumor activity of interleukin-12 in melanoma model in mice.

To study the antitumor activity of the combination immunotherapy with interleukin-12 (IL-12) and granulocyte-macrophage colony-stimulating factor (GM-CSF), a murine MmB 16 melanoma tumor model was used. Seven days after inoculation of MmB 16 melanoma cells into the footpad of the right hind limb, mice were treated with IL-12 and/or GM-CSF administered intratumorally for 7 consecutive days. IL-12 used both at a high (1 microg) and at a low (0.01 microg) dose per day produced retardation of tumor growth, although neither treatment resulted in any significant prolongation of the survival of tumor-bearing mice. GM-CSF did not by itself exert antitumor activity in this model; however, it potentiated antitumor effects of IL-12. In particular, survival of tumor-bearing mice treated with IL-12 (0.01 microg per day) and GM-CSF was significantly prolonged compared with that in mice treated with either IL-12 or GM-CSF alone.
