Parietal cell autoantibodies: localization of their binding sites on isolated gastric glands.

The localization of antigens reacting with parietal cell antibodies found in the sera of some patients with pernicious anaemia was studied by the indirect immunoperoxidase method on viable gastric glands isolated from the mouse or human fundus by collagenase digestion. We observed that only the luminal surface of parietal cell bound antibodies, whereas their lateral and basal surfaces were not stained. From this data it is concluded that parietal cell antibodies are unlikely to be cytotoxic in vivo.

Long-term management of insulin-treated diabetic patients with continuous subcutaneous insulin infusion.

Ten ambulatory diabetics were submitted to continuous subcutaneous insulin infusion (CSII) for periods of 6-18 months. With the exception of a patient who demonstrated a subcutaneous abscess, local tolerance was good. One patient suffered a severe hypoglycemic attack during the first days of CSII. Among the other subjects, hypoglycemic reactions were rare. However, blood glucose values under 50 mg/dl were recorded as frequently as during the control period. There was a non significant increment in insulin requirements and weight increased in almost all patients. In comparison with prepump period, we observed at the end of the study a significant decrease of mean blood glucose (140 vs 203 mg %), of urinary glucose output (8 vs 21 g/24 hr) and of HbA1 levels (7.6 vs 9.6%). As a rule, the improvement of diabetes control was noted throughout the study period. Nevertheless, normoglycemia was rarely reached. Diabetes stability improved as evidenced by a decrease of the standard deviation of monthly blood glucose values and by a reduction of M and MAGE indexes. In spite of the better metabolic control, we did not observe an improvement of renal function while lipid values were slightly modified. Motor nerve conduction velocities increased in 6 patients and ocular fluoroangiographic records demonstrated a reduction of microaneurysms and/or microhemorrhagic dots. As a group, the patients were satisfied with CSII and 8 of them wished to continue indefinitely with this treatment.

[Haemoglobin AIc in patients on venesection therapy for haemochromatosis (author's transl)].

Haemoglobin AIc values measured in diabetic patients with idiopathic haemochromatosis tended to be lower than in diabetics without haemochromatosis. The observed difference was analysed with respect to different parameters related to diabetes or haemochromatosis and their treatment (sex, age, duration of diabetes, insulin-dependency, frequency and volume of blood withdrawn) which could possibly explain these low values. Among the twenty-eight subjects with haemochromatosis, thirty-two non-diabetics, twenty-six insulin-dependent diabetics, and twenty-four non-insulin-treated diabetics, we observed that the more frequently venesection therapy was performed, the lower the levels of haemoglobin HbAIc, HbAIa+b tended to be. Thus, the decreased HbAIc rates observed in haemochromatosis patients may be ascribed to the venesection therapy, which induces an increased turnover of red cells, and consequently a decrease of the time available for their glycosylation.

Quantitative determination of 6-Oxo-PGF1 alpha in biological fluids by gas chromatography mass spectrometry.

A selected ion monitoring method for the determination of 6-oxo-PGF1 alpha, the stable end-product of prostacyclin, in biological fluids has been developed. In this method, biosynthetically prepared [2H6]-6-oxo-PGF1 alpha is used as internal standard. The method involves extraction, thin-layer chromatography purification and derivatization into the methyl ester, methoxime, trimethylsilyl ether derivatives by carrying out the methoximation first. Quantitative gas chromatographic mass spectrometric analysis is performed in the electron impact mode by monitoring the [M - (TMSOH + CH3O)]+ fragment ions. The use of this method in the measurement of 6-oxo-PGF1 alpha in serous fluids and in incubation media of serous tissues is described.

Separation of epoxy-embedded cell cultures from plastic flasks for electron microscopy.

Monolayers of cells grown in ordinary plastic flasks are fixed and embedded "in situ" into Epon. When polymerized at 40 C for 4 days instead of the usual 60 C, the Epon sheet containing the cells is easily detached from the bottom of the plastic container. The Epon sheet is observed by light microscopy as a histological preparation. Ultrathin sectioning of preselected areas can then be carried out in a horizontal plane.
