Histochemical determinations of copper, zinc, and iron in pigmented nevi and melanoma.

Histochemical determinations of copper, zinc, and iron in intradermal pigmented nevi and melanomas revealed the presence of copper and iron in melanoma but not in nevi. Zinc was not detected in either melanomas or nevi. However, melanin was removed from the tissues prior to staining; therefore, it is possible that zinc was also removed by the procedure. Although the function of copper and iron in the melanoma cell is not known, they may be components of abnormal enzymes.

Copper

Histochemical determination of copper, zinc, and iron in some benign and malignant tissues.

Histochemical tests for copper, iron, and zinc revealed their presence in the stratum germinativum of normal skin and skin from which an early melanoma and basal cell and squamous cell carcinomas originated. However, only copper was seen in invasive cells of basal and squamous cell carcinomas originating from skin. The normal oral mucous membrane was free of copper, iron, and zinc, but cells of invasive squamous carcinoma originating from the oral mucous membrane contained copper. The fluorescent brightening agent, applied as a counterstain, aided in the location of the specimen.

Basal Cell Carcinoma

Fluorescent brightening agents for biological staining.

Excitatory and emission wavelengths and quantum yields were determined for 16 fluorescent brightening agents. Optimal microscopic excitatory and barrier filters, determined by these results, were used to examine tissues with Phorwhite BKL and Leucophor PAF, the two agents showing the highest quantum yields. All slides exhibited a high degree of fluorescence and did not fade after several hours of exposure to light.

Fluorescence

Spectral properties and relative quantum yields of new fluorescent stains.

Excitation and emission spectra were obtained for thirty-two new fluorescent stains that resist fading when exposed to light. Quantum yields were determined for the fourteen stains that showed the greatest fluorescent intensity. Optimal microscopic excitatory and barrier filters, determined by the experimental results, were used to examine tissues stained with some of these dyes. Cellular detail was evident and there was no fading after several hours of examination.

Fluorescent Dyes

Dental survey of 620 Soviet immigrants.

Six hundred twenty recently arrived Soviet immigrants were examined at the Northern Dispensary. There was a need for extensive dental treatment among these persons and much of their previous treatment did not meet currently accepted American standards.
