[Chlamydial laryngitis: clinical picture and amixine treatment].

Chlamydia trachomatis DNA was detected by PCR in 12 patients with continuously recurrent catarrhal pharyngolaryngitis. These included 9 males and 3 females whose age ranged 16 to 72 years. All the patients were found to have degrees II-III alpha- and g-INF deficiencies. The interferon inducer amixine in combination with sumamed was used in the treatment of patients with chlamydial laryngitis. After treatment, there was a significant improvement in two thirds of patients. The pain syndrome was relieved, hoarseness diminished, and the laryngoscopic and microlaryngostroboscopic picture and interferon status became normal. Control studies showed no Ch. trachomatis in this group of patients. Despite positive changes in the course of the disease, the control study again detected Ch. trachomatis in 4 patients, which required repeated treatment courses.

[New principles and technology in the management of medical research].

New principles of management, planning, and finance of medical science with the aim to increase its efficacy are discussed. The transfer from financing the scientific administration to financing research is taken into account. The technology of expert examination is analysed as this is a basis for adopting decisions in the management of medical science.

[Isolation and characteristics of a continuous human splenic cell culture].

A continuous cell line of human spleen has been obtained. The methods of seeding large numbers of cells and prolonged cultivation of the culture played an important role in isolation of this cell line; both methods have contributed to the creation of conditions for gradual reconstruction of the cellular metabolism in the absence of which the cells wound not be able to get adapted to contiuous cultivation. As the authors failed to obtain a contiuous cell line from a single cell colony the method of "shaking" was applied. The culture was obtained only in case of a 1% PHA solution was added to the nutrient medium.

[Mutual stimulation of oncornavirus and herpetic infections in experiments in vivo and in vitro].

The evidence of mutual stimulation of herpes simplex virus (HSV) and oncornaviruses (OV) reproduction was obtained. HSV titers in mice pre-infected with Rauscher leukosis virus (RLV) were 2.25-3.0 lg LD50 higher than in mice of the same batch but without RLV infection. The continuous HEp-2 culture chronically infected with oncornaviruses was found to be more sensitive to the cytopathic effect of HSV strains, types 1 and 2, and in the culture fluid HSV accumulated to a greater titer than in primarily trypsinized culture of chick embryo fibroblasts. Electron microscope examinations of the continuous J-96 cell culture chronically infected with OV and HSV simultaneously revealed an 8.3-fold increase in the number of oncornavirus particles as compared to the control culture, infected with OV alone.

[Comparative study of the effectiveness of the purification and concentration of herpes simplex virus types 1 and 2].

The results of comparative studies on concentration and purification of herpes simplex virus type 1 and type 2 (HSV-1 and HSV-2) by ficol density gradient centrifugation are presented. A two-phase distribution of extracellular HSV was established in phycol density gradient centrifugation: in zones with density of 1.110-1.114 and 1.088-1.085 g/ml. The effectiveness of purification of HSV preparations recovered from the corresponding gradient zones was determined by electron microscopy and quantitation of the contaminating cellular (radioactive) proteins in virus purification from a mixture of the culture fluid from infected cultures and the culture fluid from uninfected labeled human embryo skin-muscle tissue cultures (HESM) and a mixture of unlabeled extracellular HSV and a homogenate of labeled uninfected HESM cultures. In HSV purification from the virus-containing culture fluid the amount of cellular proteins was shown to decrease 500-fold in 150-fold virus concentration. In purification of extracellular HSV from the mixture with cell homogenate the amount of cellular proteins decreased 70- and 100-fold for HSV-2 and HSV-1, respectively. The infectious virus yield in phycol gradient centrifugation of a precipitate obtained by the addition of polyethylene glycol-6000 to the culture fluid for HSV-1 was 34.7% (in titrations in HESM cultures) and 38.4% (by intracerebral inoculation of mice weighing 5-6 g), and for HSV-2 20.2% and 26.3%, respectively.
