Substrate-dependent autoaggregation of Pseudomonas putida CP1 during the degradation of mono-chlorophenols and phenol.

A bacterium, CP1, identified as Pseudomonas putida strain, was investigated for its ability to grow on and degrade mono-chlorophenols and phenols as sole carbon sources in aerobic shaking batch culture. The organism degraded up to 1.56 mM 2- and 3-chlorophenol, 2.34 mM 4-chlorophenol and 8.5 mM phenol using an ortho-cleavage pathway. P. putida CP1, acclimated to degrade 2-chlorophenol, was capable of 3-chlorocatechol degradation, while P. putida, acclimated to 4-chlorophenol degradation, degraded 4-chlorocatechol. Growth of P. putida CP1 on higher concentrations of the mono-chlorophenols, >or=1.56 mM 4-chlorophenol and >or=0.78 mM 2- and 3-chlorophenol, resulted in decreases in cell biomass despite metabolism of the substrates, and the formation of large aggregates of cells in the culture medium. Increases in cell biomass with no clumping of the cells resulted from growth of P. putida CP1 on phenol or on lower concentrations of mono-chlorophenol. Bacterial adherence to hydrocarbons (BATH) assays showed cells grown on the higher concentrations of mono-chlorophenol to be more hydrophobic than those grown on phenol and lower concentrations of mono-chlorophenol. The results suggested that increased hydrophobicity and autoaggregation of P. putida CP1 were a response to toxicity of the added substrates.

Variability in measurement of internal carotid artery stenosis by arch angiography and duplex ultrasonography--time for a reappraisal?

OBJECTIVES: to determine the inter- and intra-observer variability of ICA stenosis measurement using duplex, ECST and NASCET methods. DESIGN: a retrospective review of arch angiograms and carotid duplex scans in 50 patients. MATERIALS AND METHODS: carotid stenoses were calculated by three independent observers according to NASCET and ECST methods. Variation between observers for NASCET and ECST was determined. For each observer, the variation between NASCET and ECST was determined. The variation between duplex and both NASCET and ECST was determined. RESULTS: inter-observer agreement on the degree of ICA stenosis was clinically and statistically good for NASCET but was poorer for ECST. For each observer, comparison between NASCET and ECST showed 95% limits of agreement of around 50 percentage points. Comparison of duplex with NASCET and ECST showed similar 95% limits of agreement. CONCLUSIONS: arch angiography allows reproducible measurement of carotid stenosis by the NASCET method between different observers. For the ECST method, reproducibility is not so good. Variations in results between NASCET and ECST and between angiography and duplex are significant. In view of the similar results of the NASCET and ECST trials, this suggests that degree of stenosis may only be a surrogate marker for outcome following carotid endarterectomy.

Should the food intake of patients admitted to acute hospital services be routinely supplemented? A randomized placebo controlled trial.

BACKGROUND: Many patients admitted to acute hospital services are underweight or harbour vitamin deficiencies. OBJECTIVES: To determine the effect on patient throughput of a policy of routine vitamin supplementation, and of early routine sipfeed supplementation in 'thin' patients (5-10% weight loss or body mass index 18-22). DESIGN: Factorial randomized placebo controlled trial of oral multivitamins from the first day of admission, and, after nutritional screening, of a nutritionally complete sipfeed from the second day in 'thin' patients. SETTING: Acute medical, surgical and orthopaedic hospital services of a London teaching hospital. PARTICIPANTS: 1561 patients admitted as emergencies were included in the vitamin study of which 549 were included in the sipfeed study. MAIN OUTCOME MEASURE: Length of hospital stay (LOS). RESULTS: Offering multivitamins to acute admissions resulted in a mean change (reduction) in LOS of -0.4 days 95% CI (-2-1.2days). The results suggest greater reductions for those discharged after 10 days: mean change=-2.3 days 95% CI (-5.7 to 1.2). Sipfeed supplementation was associated with an increased mean length of stay 2.8 days 95% CI (-0.8-6.3). 18% of acute admissions were classified undernourished on the basis of BMI, MUAC or percent weight loss combined. CONCLUSIONS: No benefit was observed for sipfeed intervention although a small benefit of less than one day is not excluded. Vitamin supplementation may have slight but economically important benefit.

Gene therapy for myocardial protection: transfection of donor hearts with heat shock protein 70 gene protects cardiac function against ischemia-reperfusion injury.

BACKGROUND: Heat shock protein 70 (HSP70) gene transfection has been shown to enhance myocardial tolerance after normothermic ischemia-reperfusion. We investigated the effect of HSP70 gene transfection on mechanical and endothelial function in a protocol mimicking clinical heart preservation. METHODS AND RESULTS: Rat hearts were infused ex vivo with Hemagglutinating Virus of Japan-liposome complex containing HSP70 gene (HSP, n=8) or no gene (CON, n=8), and heterotopically transplanted into recipient rats. Four days after surgery, transfected hearts were perfused on a Langendorff apparatus for 45 minutes, arrested with St Thomas' No. 1 cardioplegia for 4 hours at 4 degrees C, and reperfused for 1 hour. Mechanical and endothelial function was studied before and after ischemia. Creatine kinase was measured in reperfusion effluent. Hearts underwent Western blotting and immunohistochemistry to confirm HSP70 overexpression. Postischemic recovery of mechanical function (% preischemic+/-SEM) was greater in HSP versus CON: Left ventricular developed pressure recovery was 76.7+/-3.9% versus 60. 5+/-3.1% (P:<0.05); dP/dtmax recovery was 79.4+/-4.9% versus 56. 2+/-3.2% (P:<0.05); dP/dtmin recovery was 74.8+/-4.6% versus 57. 3+/-3.6% (P:<0.05). Creatine kinase release was attenuated in HSP versus CON: 0.22+/-0.02 versus 0.32+/-0.04 IU/min/g wet wt. (P:<0. 05). Recovery of coronary flow was greater in HSP versus CON: 76. 5+/-3.8% versus 59.2+/-3.2% (P:<0.05). Recovery of coronary response to 5-hydroxytryptamine (5 x 10(-)(5) mol/L) was 55.6+/-4.7% versus 23. 9+/-3.2% (P:<0.05); recovery of coronary response to glyceryltrinitrate (15 mg/L) was not different between HSP and CON: 87.4+/-6.9% versus 84.3+/-5.8% (NS). CONCLUSIONS: In a clinically relevant donor heart preservation protocol, HSP70 gene transfection protects both mechanical and endothelial function.

MRI measurements correctly predict the relative effects of tumor oxygenating agents on hypoxic fraction in rodent BA1112 tumors.

PURPOSE: We evaluate whether magnetic resonance imaging (MRI) with blood oxygenation level-dependent (BOLD) contrast correctly predicts the relative effects of tumor-oxygenating agents on hypoxic fraction in BA1112 rhabdomyosarcomas in WAG/Rij rats. METHODS AND MATERIALS: The response of ten tumors to carbogen (95% O(2)/5% CO(2)), a perfluorocarbon emulsion (PFC), and the combination of PFC + carbogen was studied with high spectral and spatial resolution MR imaging of the water resonance at 4.7 Tesla. Decreases in MR signal linewidth indicate increases in tumor blood oxygen levels. RESULTS: Average MR signal linewidth was decreased 2.0% by carbogen, 2.5% by PFC + air, and 4.9% by PFC + carbogen. PFC + carbogen caused a larger linewidth decrease than either treatment alone (p < 0.04 by ANOVA). Maps of pixels responding to treatment indicate that combining PFC with carbogen significantly enlarges the area of the tumor in which oxygen levels are increased (p < 0.01 by ANOVA). CONCLUSION: MRI predicts that PFC + carbogen will increase radiosensitivity more than either treatment alone; this agrees with the known effects of these treatments on hypoxic fraction. Utilizing MRI to choose the treatment that maximizes the size and extent of increases in tumor oxygenation could reduce hypoxic fraction.

Relationship between endothelial cell markers and arterial stenosis in peripheral and carotid artery disease.

Damage to the endothelium is an important component of atherosclerosis and can be quantified by measuring plasma markers, such as von Willebrand factor, thrombomodulin, intercellular adhesion molecule-1, and E-selectin. We hypothesized that increased levels of these markers would be related to objectively defined disease severity among patients with peripheral atherosclerosis or carotid atherosclerosis. To test this, we measured the markers by using ELISA in the plasma of 45 patients with intermittent claudication alone and in 53 patients presenting with transient ischemic attack. Disease severity in the former was by ankle-brachial pressure index and in the latter by ultrasound defined % stenosis. Any symptomatic dual disease or history or present coronary atherosclerosis warranted exclusion. Data were correlated according to Spearman's method. The only significant correlation was between von Willebrand factor and ankle-brachial pressure index (r = -0.39, p = 0.008). Our data suggest that von Willebrand factor is the most sensitive marker of peripheral atherosclerosis and that none of the plasma markers seems to be a useful marker of the degree of carotid artery stenosis.

Cdc37 promotes the stability of protein kinases Cdc28 and Cak1.

In the budding yeast Saccharomyces cerevisiae, Cdc37 is required for the productive formation of Cdc28-cyclin complexes. The cdc37-1 mutant arrests at Start with low levels of Cdc28 protein, which is predominantly unphosphorylated at Thr169, fails to bind cyclin, and has little protein kinase activity. We show here that Cdc28 and not cyclin is specifically defective in the cdc37-1 mutant and that Cdc37 likely does not act as an assembly factor for Cdc28-cyclin complex formation. We have also found that the levels and activity of the protein kinase Cak1 are significantly reduced in the cdc37-1 mutant. Pulse-chase analysis indicates that Cdc28 and Cak1 proteins are both destabilized when Cdc37 function is absent during but not after translation. In addition, Cdc37 promotes the production of Cak1, but not that of Cdc28, when coexpressed in insect cells. We conclude that budding yeast Cdc37, like its higher eukaryotic homologs, promotes the physical integrity of multiple protein kinases, perhaps by virtue of a cotranslational role in protein folding.

Degradation of mono-chlorophenols by a mixed microbial community via a meta- cleavage pathway.

A mixed microbial community, specially designed to degrade a wide range of substituted aromatic compounds, was examined for its ability to degrade mono-chlorophenols as sole carbon source in aerobic batch cultures. The mixed culture degraded 2-, 3-, and 4-chlorophenol (1.56 mM) via a meta- cleavage pathway. During the degradation of 2- and 3-chlorophenol by the mixed culture, 3-chlorocatechol production was observed. Further metabolism was toxic to cells as it led to inactivation of the catechol 2,3-dioxygenase enzyme upon meta- cleavage of 3-chlorocatechol resulting in incomplete degradation. Inactivation of the meta- cleavage enzyme led to an accumulation of brown coloured polymers, which interfered with the measurement of cell growth using optical density. Degradation of 4-chlorophenol by the mixed culture led to an accumulation of 5-chloro-2-hydroxymuconic semialdehyde, the meta- cleavage product of 4-chlorocatechol. The accumulation of this compound did not interfere with the measurement of cell growth using optical density. 5-chloro-2-hydroxymuconic semialdehyde was further metabolized by the mixed culture with a stoichiometric release of chloride, indicating complete degradation of 4-chlorophenol by the mixed culture via a meta- cleavage pathway.

Do some men with acne vulgaris have raised levels of LH?

OBJECTIVE: Acne vulgaris is androgen related and in some cases is associated with excess androgen production, which in men would be mainly testicular or adrenal in origin. Ordinarily, testosterone synthesis in the testis is controlled by serum LH. In order to ascertain whether acne vulgaris in men might be consequent on abnormalities at the hypothalmic/pituitary level we have compared serum LH levels in men with and without acne. DESIGN AND PATIENTS: 111 men with acne vulgaris were compared with 51 acne-free men. MEASUREMENTS: Serum levels of LH, FSH and oestradiol were measured by automated ELISA. Dehydroepiandrosterone sulphate (DHEA-S), 17 alpha-hydroxyprogesterone, 11-deoxycortisol, androstenedione and testosterone were measured by radioimmunoassay, and SHBG by immunoradiometric assay. RESULTS: The controls showed a fall in serum LH with age: the rate of fall was less in the acne patients (difference between the slopes 0.073 U/year (95% confidence interval (CI), 0.016 to 0.130)). The age distributions in the acne and control groups differed. In order to compare means, standardizations were used to assess the difference in the means in populations of men with age distributions similar to the cases and controls. After standardization to the age distribution of the controls, the acne patients had higher means of serum LH, difference between the means 0.80 U/I (95% CI, 0.31 to 1.30), androstenedione, 1.09 nmol/l (95% CI, 0.15 to 2.03) and testosterone levels, 2.74 nmol/l (95% CI, 0.53 to 4.95) and a lower mean BMI, -1.30 kg/m2 (95% CI, -2.40 to -0.19). After standardization to the age distribution of the acne patients the means of LH, androstenedione and testosterone were still higher in the acne patients but not necessarily significantly so. CONCLUSION: LH fell more slowly with age in the men with acne as compared to the controls. In our heterogeneous group of patients with acne vulgaris the findings suggest that a sub-group of men with acne have raised serum LH levels.

Recessive x-linked ichthyosis: role of cholesterol-sulfate accumulation in the barrier abnormality.

Cholesterol sulfate is a multifunctional sterol metabolite, produced in large amounts in squamous keratinizing epithelia. Because patients with recessive x-linked ichthyosis display not only a 10-fold increase in cholesterol sulfate, but also a 50% reduction in cholesterol, we assessed here whether cholesterol sulfate accumulation and/or cholesterol deficiency produce abnormal barrier function in recessive x-linked ichthyosis. Patients with recessive x-linked ichthyosis display both an abnormal barrier under basal conditions, and a delay in barrier recovery after acute perturbation, which correlate with minor abnormalities in membrane structure and extensive lamellar-phase separation. Moreover, both the functional and the structural abnormalities were corrected by topical cholesterol. Yet, topical cholesterol sulfate produced both a barrier abnormality in intact skin and extracellular abnormalities in isolated stratum corneum, effects largely reversed by coapplications of cholesterol. Together, these results suggest that cholesterol sulfate accumulation rather than cholesterol deficiency is responsible for the barrier abnormality. Despite the apparent importance of cholesterol sulfate-to-cholesterol processing for normal barrier homeostasis, neither steroid sulfatase activity nor mRNA levels are upregulated following acute perturbations. These results demonstrate both a potential role for cholesterol sulfate-to-cholesterol processing in normal permeability barrier homeostasis, and that basal levels of steroid sulfatase are sufficient to accommodate acute insults to the permeability barrier.

Carotid disease in acute stroke.

BACKGROUND: the Oxfordshire Community Stroke Project (OCSP) devised a simple clinical classification for acute stroke which predicted mortality, functional recovery and patterns of recurrent stroke. We aimed to determine whether this could predict the presence of carotid disease and be used to select which patients with acute stroke should be referred for carotid imaging with a view to subsequent carotid endarterectomy. METHODS: we assessed patients with acute stroke admitted to seven hospitals over a 10-month period. Patients were classified according to the OCSP system and their carotid arteries investigated using portable continuous wave Doppler. Those with abnormal portable assessments had colour duplex Doppler imaging. RESULTS: of 305 patients with proven or probable cerebral infarction, severe (70-99%) ipsilateral carotid stenosis was found in 16 (16%) of the 101 with partial anterior circulation infarct (PACI), four (4%) of the 100 with total anterior circulation infarct (TACI), none of the 80 with lacunar infarct (LACI) and one (4%) of the 24 with posterior circulation infarct (POCI). Complete ipsilateral carotid occlusion was found in 25 (25%) of the TACI group, 11 (11%) of the PACI group, three (4%) of the LACI group and none of the POCI group. Severe carotid stenosis or occlusion was more common in the ipsilateral than the contralateral carotid artery for the TACI and PACI groups (chi2 P< 0.05), but there was no difference between ipsilateral and contralateral carotid disease in the LACI and POCI groups. If the OCSP classification is used to detect patients with 70-99% carotid stenosis, then the sensitivity is 76% and specificity is 70%. CONCLUSION: these findings suggest that ipsilateral carotid disease is an important cause of stroke for those with anterior circulation infarcts but not for those with LACI or POCI. Subjects with PACI should be referred for early carotid imaging to identify those with severe disease who may be suitable for elective carotid surgery.

Cak1 is required for Kin28 phosphorylation and activation in vivo.

Complete activation of most cyclin-dependent protein kinases (CDKs) requires phosphorylation by the CDK-activating kinase (CAK). In the budding yeast, Saccharomyces cerevisiae, the major CAK is a 44-kDa protein kinase known as Cak1. Cak1 is required for the phosphorylation and activation of Cdc28, a major CDK involved in cell cycle control. We addressed the possibility that Cak1 is also required for the activation of other yeast CDKs, such as Kin28, Pho85, and Srb10. We generated three new temperature-sensitive cak1 mutant strains, which arrested at the restrictive temperature with nonuniform budding morphology. All three cak1 mutants displayed significant synthetic interactions with loss-of-function mutations in CDC28 and KIN28. Loss of Cak1 function reduced the phosphorylation and activity of both Cdc28 and Kin28 but did not affect the activity of Pho85 or Srb10. In the presence of the Kin28 regulatory subunits Ccl1 and Tfb3, Kin28 was phosphorylated and activated when coexpressed with Cak1 in insect cells. We conclude that Cak1 is required for the activating phosphorylation of Kin28 as well as that of Cdc28.

Cellular localisation of the ataxia-telangiectasia (ATM) gene product and discrimination between mutated and normal forms.

The recently cloned gene (ATM) mutated in the human genetic disorder ataxia-telangiectasia (A-T) is involved in DNA damage response at different cell cycle checkpoints and also appears to have a wider role in signal transduction. Antibodies prepared against peptides from the predicted protein sequence detected a approximately 350 kDa protein corresponding to the open reading frame, which was absent in 13/23 A-T homozygotes. Subcellular fractionation, immunoelectronmicroscopy and immunofluorescence showed that the ATM protein is present in the nucleus and cytoplasmic vesicles. This distribution did not change after irradiation. We also provide evidence that ATM protein binds to p53 and this association is defective in A-T cells compatible with the defective p53 response in these cells. These results provide further support for a role for the ATM protein as a sensor of DNA damage and in a more general role in cell signalling, compatible with the broader phenotype of the syndrome.

Pilot study of carotid surgery for acute stroke.

BACKGROUND: Carotid endarterectomy is usually delayed for 2 months following an acute stroke, but the stroke may progress or a further stroke may occur. A randomized pilot study of urgent carotid surgery for acute stroke was undertaken to assess the feasibility of a definitive multicentre trial. METHODS: Seven Manchester hospitals referred patients with acute stroke (less than 7 days previously), defined according to the Oxfordshire Community Stroke Project classification The carotid arteries were examined by portable continuous-wave Doppler ultrasonography. Patients with more than 50 per cent carotid stenosis on portable Doppler ultrasonography underwent colour duplex Doppler imaging. Patients with more than 70-99 per cent carotid stenosis were randomized to urgent carotid surgery or best medical care. RESULTS: A total of 593 patients were assessed over 2 years. Of these, 414 had acute stroke, of whom 380 had probable cerebral infarction. Complete ipsilateral internal carotid artery occlusion was most common in patients with total anterior circulation infarcts (33 (28 per cent) of 117), whereas severe ipsilateral (70-99 per cent) stenosis was most common in partial anterior circulation infarcts (25 (20 per cent) of 128). Only 16 patients (one total and 15 partial anterior circulation infarcts) were fit enough for surgery and consented to randomization. CONCLUSION: A definitive trial of urgent carotid surgery would need to screen large numbers of patients but could focus on patients with partial anterior circulation infarcts.

Cloning and characterization of tomato leaf senescence-related cDNAs.

Senescence-related cDNA clones designated SENU1, 4, 5 (senescence up-regulated) and SEND32, 33, 34, 35 and 36 (senescence down-regulated) isolated from a tomato leaf cDNA library [9] were characterized. Southern analysis showed that SEND32 is encoded by a single-copy gene while SEND33, 34, 35, 36 and SENU1 and SENU5 are members of small gene families. DNA and protein database searches revealed that SEND32, SEND35, SENU1 and SENU5 are novel cDNAs of unknown function. SEND33 encodes ferredoxin, SEND34 encodes a photosystem II 10 kDa polypeptide and SEND36 encodes catalase. The SENU4 sequence is identical to the P6 tomato protein previously reported to be pathogenesis-related [46]. The mRNA levels of SENU1, 4 and 5 increased during leaf senescence and SENU1 and SENU5 were also expressed at high levels during leaf development and in other plant organs. The SENU4 mRNA was associated more specifically with leaf senescence, although low expression was also detected in green fruit. The mRNAs for all SEND clones decreased during tomato leaf development and senescence and all except SEND32 were expressed at low levels in other plant organs. The accumulation of mRNA homologous to SENU4 and the decrease in abundance of SEND32 provide good molecular markers for leaf senescence.

Intracellular cytokine expression in whole blood preparations from normals and patients with atopic dermatitis.

In recent years, the importance of characterizing the role of cytokines in a wide range of clinical conditions has resulted in development of new methods to assess cytokine expression in clinical samples. The use of anti-cytokine MoAbs and flow cytometry to detect cytokines intracellularly at the single-cell level has the potential to quantify cytokine production in different diseases. For this technique to be useful in a clinical setting, rapid throughput of clinical samples and a cheap, reliable assay would be required, therefore the development of the above technique using unseparated whole blood samples would be advantageous. Using this technique, only one study to date (Maino et al., 1996) has used unseparated whole blood as the source of cells for detecting intracellular cytokines. In clinical practice, whole blood may be optimal, since this most closely approximates conditions in vivo: as no purification of blood mononuclear cells is required, very little blood is needed to detect a number of cytokines simultaneously in various lymphocyte subpopulations, and the assay can be applied to samples from infants and children. In this study we describe an intracellular cytokine assay using unseparated whole blood from normals. In activated CD8- T cells, IL-2 and interferon-gamma (IFN-gamma) were optimally induced after 10 h stimulation with phorbol 12-myristate acetate (PMA)/ionomycin, and in CD8+ T cells IL-2 was optimally induced after 10 h and IFN-gamma after 6 h. The levels of IL-2 and IFN-gamma in CD8+ and CD8- T cells in four healthy individuals were consistent on four occasions over a 3-month period. In a large group of 34 normal subjects, there was considerable heterogeneity in CD3/IL-2+ (range 9.7-41.3) and CD3/IFN-gamma+ cells (10.1-44), expressed as a percentage of total lymphocytes. In patients with atopic dermatitis (n = 5) there was a significantly decreased percentage of CD3+/CD8+ peripheral blood T cells expressing IFN-gamma and an increased percentage of CD3+/CD8- T cells expressing IL-4 compared with non-atopic dermatitis controls (n = 5). Possible applications of this technique are discussed.

The effect of temperature and adrenaline on the relative importance of the sarcoplasmic reticulum in contributing Ca2+ to force development in isolated ventricular trabeculae from rainbow trout

The sarcoplasmic reticulum (SR) is central to intracellular Ca2+ regulation during excitationcontraction (E-C) coupling in mammalian cardiac tissue. The importance of the SR to E-C coupling in lower vertebrates is less certain. This uncertainty can be attributed, in part, to the temperature-dependency of the SR Ca2+-release channel and to interspecific differences in the ryanodine-sensitivity of ectotherm cardiac muscle. Furthermore, the relative importance of the SR in contributing intracellular Ca2+ to force development may be influenced by adrenergic stimulation, which increases trans-sarcolemmal (extracellular) Ca2+ influx. The objective of this study was to assess the relative importance of SR (intracellular) and sarcolemmal (SL; extracellular) Ca2+ fluxes during the isometric contraction of isolated ventricular trabeculae from rainbow trout Oncorhynchus mykiss. To approximate in vivo Ca2+ availability to the muscle better, a tonic level (10 nmol l-1) of adrenaline was used in all control experiments, and SL Ca2+ influx was stimulated with high levels (10 µmol l-1) of adrenaline. Ryanodine, a noted blocker of SR Ca2+ release in mammals, was used to assess SR involvement. To examine the role of temperature on the relative Ca2+ contribution from each source, experiments were performed at two temperatures (12 and 22 °C), using ventricular trabeculae from fish acclimated to both 12 and 22 °C. Under all test conditions studied, SL Ca2+ influx was the primary source of activator Ca2+, as assessed by the change in isometric force after ryanodine application. Even so, the SR contribution of activator Ca2+ was significantly greater at a test temperature of 22 °C than at 12 °C. We attribute this observation to the temperature-dependent nature of the SR Ca2+-release channel. At 22 °C and under control conditions, ryanodine reduced peak tension at all pacing frequencies (by approximately 50 % at 0.2 Hz, approximately 25 % at 1.2 Hz and approximately 20 % at 2.0 Hz), regardless of acclimation temperature. Therefore, the SR is a significant, but secondary, contributor of activator Ca2+ for tension development at warm temperatures. The magnitude of SR Ca2+ contribution was inversely related to pacing frequency, but remained significant at physiological pacing frequencies. This was a novel finding. The degree of ryanodine-sensitivity in the present study was greater than that reported previously for the rainbow trout. We attribute this difference to the use of tonic adrenergic stimulation in the present study. In contrast to the experiments at the warmer test temperature, at 12 °C and under control conditions, ryanodine significantly reduced peak tension only at low frequencies (by approximately 25 % at 0.2 Hz), regardless of acclimation temperature. These findings suggest that at cold temperatures, and at physiologically relevant pacing frequencies, the SR may not be important in supplying Ca2+ to the contractile elements of the trout heart. At both test temperatures and regardless of acclimation temperature, stimulation with 10 µmol l-1 adrenaline caused positive inotropy of sufficient magnitude to ameliorate the negative inotropic effect of ryanodine completely, with the exception of high pacing frequencies (>1.2 Hz) at 22 °C, where adrenergic stimulation did not fully compensate for the effects of ryanodine. This exception is discussed in relation to the reduced adrenergic sensitivity of the trout myocardium at warm temperatures. The adrenergically mediated compensation for the loss of the SR Ca2+ supply is a novel finding for fish hearts. Therefore, while our study clearly demonstrates that the relative importance of SR Ca2+ release is subject to temperature and frequency, adrenaline-mediated increases in SL Ca2+ influx decrease the importance of the SR in contributing Ca2+ to E-C coupling in trout ventricular myofilaments.
