[Structure of carbohydrate chains of molecular forms of alpha1-acidic glycoprotein from ascitic fluid from patients with stomach cancer].

The immunochemically pure alpha 1-acid glycoprotein (aAGP) from ascitic fluid of patients with stomach cancer was separated by chromatography on Con A-Sepharose into Con A-nonbound (aAGP-1, 43, 5 kDa, 70%) and Con A-bound (aAGP-2, 41.5 kDa, 24% and aAGP-3, 40.0 kDa, 5%) forms, differing in the monosaccharide composition. Comparative study of structures of their N-carbohydrate chains with the aid of the HPLC of fluorescence-labelled oligosaccharides showed that the molecular forms differ by the ratio of the di-, tri-, and tetraantennary carbohydrate N-chains of a complex type. The molecular forms of aAGP differ from nAGP by amounts of Le(x)-fragments and agalacto-oligosaccharides.

[Effect of alpha-acidic glycoprotein on oxyproline metabolism in experimental thermal trauma].

alpha 1-Acid glycoprotein, isolated from human blood plasma, was shown to accelerate healing of burn wounds in rats and to increase daily urine excretion. The glycoprotein restricted an elevation of the peptide-bound hydroxyproline in blood plasma, decreased the ratio between peptide-bound and free hydroxyproline within all the periods of experiment during 3 weeks as well as stimulated hydroxyproline excretion with urine at the step of active reparation of burn wounds. The glycoprotein appears to protect connective tissue metabolism in burns.

[Effect of alpha1-acid glycoprotein on rheologic blood parameters in experimental thermal burns].

Experiments were conducted on albino noninbred mice and rats to study the effect of alpha 1-acid glycoprotein (alpha 1-AG) on burn healing. Injection of alpha 1-AG in the first 24 hours after the burn causes rapid epithelialization and increases contracture of the burn wounds. At the same time it increases the deformity of the red cells, reduces their hemolysis due to a mechanical effect, and reduces spontaneous aggregation of blood platelets. No authentic effect of alpha 1-AG on the aggregation of red cells was revealed. It is suggested that alpha 1-AG is a natural defence factor of the organism which prevents the development of microvascular thrombosis and limits in this way the size of the inflammatory and necrotic processes.

[Immunotropic activity of human alpha1-glucoprotein].

Experiments were conducted to study the effect of alpha-acid glycoprotein on a fatal infection caused by Pseudomonas pyocyanea, growth of melanoma B-16, and transplantation of a skin graft from C57BL/6 mice to CBA mice. Injection of the agent significantly increased the anti-infection resistance in mice, suppressed growth of melanoma-16, and prolonged the survival of the skin grafts, which was evidence of marked glycoprotein immunomodulating activity.

[The effect of alpha-1-acidic glycoprotein on neutrophil chemiluminescence and lipid peroxidation in experimental thermal trauma].

Induced chemoluminescence of neutrophils was distinctly inhibited both in intact mice and the animals with thermal injury after administration of acid alpha 1-glycoprotein. At the same time, the glycoprotein decreased the rate of thermal burn-induced activation of lipid peroxidation in rat blood, skin and liver tissue. Antioxidative and therapeutic effects of acid alpha 1-glycoprotein in burns appear to be related to regulation of neutrophil activity.
