[The enzymatic activity of ammonia metabolism in the liver during the parenteral nitrogen feeding of animals with experimental liver failure].

It was established that liver insufficiency the activity of hepatic glutamate dehydrogenase is markedly reduced that is, apparently, the main cause of hyperammonemia that accompanies hepatic encephalopathy. Here occurs a significant reduction of the processes of ammonia detoxication with formation of glutamine in the liver as evidenced by changes of the activity of glutamine synthetase. Administration parenterally of a preparation of nitrous feeding increases essentially ammonia detoxicating function of muscular tissue which supplements the detoxicating function of the liver.

[Dynamics of asparaginase and adenosine deaminase activity in the liver with intraperitoneal administration of aminosol and amikin preparations of parenteral nitrogen nutrition].

In experiments with mongrel male rats the asparaginase and adenosine deaminase activities in the liver tissue and adenosine deaminase in blood serum were determined under different conditions of parenteral nutrition. The intraperitoneal administration of the preparations of parenteral nitrogen nutrition-aminosol and amikin (0.25 g of conditioned protein per 100 g of body weight) against a background of protein deficiency and exhaustion is shown to cause no changes as compared to the control of these enzymes activity in the liver tissue and blood serum. The asparaginase activity in the liver increases noticeably with the dose of aminosol and amikin up to 0.5 g of conditioned protein per 100 g of body weight and the adenosine deaminase activity undergo no essential changes. A statistically significant decrease in the adenosine deaminase activity is observed only under administration of aminosol against a background of protein deficiency. Under oral feeding of rats with amikin in the composition of protein-free (0.5 g of conditioned protein per 100 g of body weight), as distinct from its parenteral administration, the asparaginase activity in the liver is considerably lower. The adenosine deaminase activity in the liver and blood serum is not practically changed. A part of the nitrogen excreted from the organism with urea and ammonia under protein deficiency is supposed to be a product of deamination of endogenic purine and pyrimidine derivatives.

[The possibility of intensifying endogenous nitrogen reutilization in protein starvation by the parental administration of small doses of methionine and threonine].

Experiments conducted on albino Wistar male rats showed that endogenous nitrogen reutilization can be intensified by parenteral administration of regulatory amino acids methionine and threonine and their mixture in doses of 100-200 mg/kg in short-term protein deprivation. The data obtained may be used in clinical practice to reduce nitrogen excretion from the patient's organism with the patient deprived of food in the early postoperative period.

[Direct phenol-hypochlorite method for determining glutaminase activity].

A simple and sensitive method is developed to determine the glutaminase activity. Conditions are selected for detecting enzymatically formed ammonia in the presence of such a high-labile substrate as glutamine. The method is simple in performance, readily reproducible and high-sensitive.

[Aspartate and alanine aminotransferase activity in the liver and blood serum of protein-deficient rats receiving parenteral nitrogen nutrition].

The activity of aspartate and alanine aminotranspherase in the liver and blood serum was determined under parenteral nitrogen nutrition against a background of protein deficiency and protein inanition. The activity of these enzymes in the rat liver under conditions of protein deficiency and inanition is practically the same. The intraperitoneal administration of aminosol and amikin against a background of 10- and 20-day protein deficiency increases the activity of the enzymes under study in the liver. With protein deficiency and inanition the aspartate aminotranspherase activity in the liver under conditions of amikin per os feeding in the composition of protein-free semisynthetic food is considerably lower than under parenteral administration of this preparation respective dose. Under given condition of the experiment the alanine aminotranspherase activity in the liver is, vice versa, higher under per os feeding than under parenteral administration of amikin. It is also shown that administration of the parenteral nitrogen nutrition preparations containing only free amino acids (amikin) or a mixture of free amino acids and low-molecular peptides (aminosol) has a unidirected effect on reamination reactions adaptability in the liver.

[Asparaginase and adenosine deaminase activities in rat liver under different conditions of protein and protein-free diet].

The activities of asparaginase and adenosine desaminase were determined in the liver tissue and that of adenosine desaminase--in blood serum of rats under different conditions of protein and protein-free diet. The asparaginase activity is shown to decrease significantly in the liver tissue under conditions of protein-free diet and increases under long complete protein and complete caloric diet as well as under complete fasting. The adenosine desaminase activity in the liver tissue lowers significantly as compared to the control under conditions of both the 20-day complete protein diet and protein diet and increases to some extent as compared to its level in the animals under conditions of protein deficiency and complete fasting. In blood serum the adenosine desaminase activity decreases under the 20-day complete protein diet and when transferring rats from the 10-day protein-free diet to the 10-day complete protein diet. It is supposed that the unidirectional change in the adenosine desaminase activity in the liver tissue under different conditions of feeding is associated with the diversity of ways stimulating the production of adenosine in the decomposition processes.

[Effect of protein and protein-free nutrition on the adaptability of transamination reactions in the liver].

It has been shown in experiments on rats that aspartate aminotransferase (AsAT) and alanine aminotransferase (A1AT) playing an important role in amino acid metabolism by the liver are easily adaptable to nutrition conditions. The activity of the enzymes increase in the liver of rats kept on diet containing 24% of casein and on complete fasting but decreases in the course of administering isocaloric protein-free diet. Replacement of protein-free diet by protein diet after 10 days leads to a drastic increase in the AsAT and A1AT activity.
