Results of miconazole therapy in twenty-eight patients with paracoccidioidomycosis (South American blastomycosis).

Results are presented of treatment with miconazole, orally and intravenously, in patients with paracoccidioidomycosis. Twenty-eight male patients aged from 34 to 66 years and exhibiting various clinical forms of the disease were studied. Twenty-five came from endemic areas in north east Argentina (Chaco, Formosa, Misiones, Corrientes and northern Santa Fe) and the remaining three from Paraguay. Twenty patients were engaged in agricultural work or at woodmills. single or multiple lesions were observed in 24 cases. Thirteen were suffering from infection of the larynx and in two of them a tracheotomy was necessary. Twenty-three showed pulmonary lesions on X-rays. Twelve had ganglionic lesions, eight had cutaneous lesions and one patient had osteoarthritis of the knee. One patient had hepatomegaly which was unrelated to chronic alcoholism. Fourteen patients had received previous treatments such as sulphonamides and amphotericin B (7 cases); sulphonamides (3), sulphonamides and the combination sulfamethoxazole + trimethoprim (3), and one patient had received all three medications. All patients had relapsed before starting miconazole therapy. Diagnosis was established by the presence of P. brasiliensis in all cases, recovered either from cutaneous or mucosal biopsy samples or from the sputum. Complement fixation tests were positive in all patients at the onset of the treatment and the immunodiffusion reactions showed precipitation bands in 27/28 patients. Skin tests with P. brasiliensis antigens proved to be positive in 18 cases and negative in 10. The erythrocyte sedimentation rate was markedly accelerated in 22 patients (greater than 20 mm in the first hour).(ABSTRACT TRUNCATED AT 250 WORDS)

Freeze-drying of a purified human diploid cell rabies vaccine.

A rabies vaccine prepared in human diploid cell strains was purified by continuous-flow sucrose density gradient centrifugation. The peak fractions containing 35-40% sucrose were diluted with a stabilizer consisting of degraded gelatine in a Tris-EDTA-buffer, and then inactivated with beta-propiolactone and freeze-dried to a residual moisture of approximately 2.5%. This vaccine was stable as proved by accelerated stability tests.

Biochemical identification of a lipoprotein with maltose-binding activity in the thermoacidophilic Gram-positive bacterium Alicyclobacillus acidocaldarius.

Growth of the thermoacidophilic Gram-positive bacterium Alicyclobacillus acidocaldarius strain ATCC 27009 on maltose resulted in the increased production of a protein with apparent molecular mass of 40 kDa. By metabolic labelling with 14C-palmitic acid, the 40-kDa protein was identified as a lipoprotein. The protein exhibited maltose-binding activity at pH 3.5, as demonstrated by chromatography on cross-linked amylose. Partial amino acid sequence analysis revealed that the 40-kDa protein corresponds to the product of an open reading frame downstream from the amylase gene (amy) that displays similarity to enterobacterial maltose-binding proteins.

Noise and the PSTH response to current transients: I. General theory and application to the integrate-and-fire neuron.

An analytical model is proposed that can predict the shape of the poststimulus time histogram (PSTH) response to a current pulse of a neuron subjected to uncorrelated background input. The model is based on an explicit description of noise in the form of an escape rate and corresponding hazard function. Two forms of the model are presented. The full model is nonlinear and can be integrated numerically, while the linearized version can be solved analytically. In the linearized version, the PSTH response to a current input is proportional to a filtered version of the input pulse. The bandwidth of the filter is determined by the amount of noise. In the limit of high noise, the response is similar to the time course of the potential induced by the input pulse, while for low noise it is proportional to its derivative. For low noise, a second peak occurs after one mean interval. The full nonlinear model predicts an asymmetry between excitatory and inhibitory current inputs. We compare our results with simulations of the integrate-and-fire model with stochastic background input. We predict that changes in PSTH shape due to noise should be observable in many types of neurons in both subthreshold and suprathreshold regimes.

Noise and the PSTH response to current transients: II. Integrate-and-fire model with slow recovery and application to motoneuron data.

A generalized version of the integrate-and-fire model is presented that qualitatively reproduces firing rates and membrane trajectories of motoneurons. The description is based on the spike-response model and includes three different time constants: the passive membrane time constant, a recovery time of the input conductance after each spike, and a time constant of the spike afterpotential. The effect of stochastic background input on the peristimulus time histogram (PSTH) response to spike input is calculated analytically. Model results are compared with the experimental data of Poliakov et al. (1996). The linearized theory shows that the PSTH response to an input spike is proportional to a filtered version of the postsynaptic potential generated by the input spike. The shape of the filter depends on the background activity. The full nonlinear theory is in close agreement with simulated PSTH data.

Role of T-region borders in Agrobacterium host range.

The limited host range AB3 strain of Agrobacterium tumefaciens induces tumors by transferring two T-regions, TA and TB. TA is a deleted version of the well-known biotype I octopine TL-region that lacks the iaa and ipt genes, but carries an intact oncogene, gene 6b, and typical left and right border sequences. TB carries two iaa genes that together code for the synthesis of indoleacetic acid. Gene 6b and the iaa gene act synergistically when transferred in a coinoculation experiment. The TA-region of the limited host range isolate Ag57 is related to the TA-region of AB3, but differs from it at several positions. The most significant difference is the absence of the right border region. In spite of this, Ag57 and the exconjugant strain C58C9(pTiAg57) induce normal tumors on Nicotiana rustica and Vitis vinifera. Various experiments indicate that gene 6b of the Ag57 TA-region is active and transferred in spite of the absence of the right border. On N. tabacum, C58C9(pTiAg57) is nononcogenic but becomes oncogenic when the pTiAg57 TA-region is restored by the right TA border sequence of pTiAB3. Thus, the right TA border sequence of the biotype III limited host range strains is required for tumor induction on some hosts, but not on others.
