Splayleg: a spontaneous limb defect in rabbits. Genetics, gross anatomy, and microscopy.

A limb defect in rabbits known as splayleg arose in a closed breeding colony of Dutch rabbits. Since the causes of this postural deformity are not fully understood, an effort was made to clarify the mode of inheritance and the etiology of the disease. Pedigree analysis and genetic mating tests showed that, in contrast to published reports, the disease is not determined by a single recessive gene with complete expressivity. The underlying genetic system is composed of one recessive gene with reduced expressivity or of more genes with the probable involvement of environmental factors. Femurs of abnormal limbs displayed endotorsion of the shaft and anteversion of the neck as the main features. Endotorsion of the femur was associated with exotorsion of the tibia. Dislocation of the hip was not observed. There was no abnormnal laxity of the joint capsule or the ligamentaum teres. Only minor differences in the acetabulum between control and splayleg rabbits were observed. Enzyme histochemistry and electron microscopy of the gluteus medius, adductor longus- and magnus, sartorius, gracilis, and semitendinosus muscles and the femoral, peroneal, and tibial nerves did not reveal any qualitative differences between splayleg and control rabbits. A study on muscle fiber diameter showed that fibers of splayleg animals were in general smaller, particularly in the semitendinosus. Atypical mitochondria of muscles were observed in all rabbits, but more frequently in splayleg animals. A hypothesis is presented in which imbalanced development of the neural, muscular and skeletal systems of the limb is the primary defect. This results in torsion of the femur. It depends on the degree of femoral torsion and the capacity of the young rabbit for compensation by exorotation of the limb in the hip whether a persistent splayleg posture will develop.

Cysts in rabbit foetal brains. Malformation or artefacts?

Reports have appeared in the literature on brain cysts in rabbit foetuses. This paper reports on investigations carried out to assess whether "cystic dilatation" is a malformation or an artefact. The results show that "cystic dilatation" arises by splitting of the pia-arachnoid membrane leading to a space between the skull and the neural tissue which is lined by the pial layer on its interior aspect and the arachnoid on its exterior aspect. The evidence presented indicates that "cystic dilatation" is a fixation-induced artefact. In addition, the presence of several artificial tissue clefts and spaces in preserved rabbit foetal brains is reported.

Mutagenicity studies with o-tolidine and 4,4'-tetramethyldiaminodiphenylmethane.

This paper confirms the mutagenicity of 2 carcinogenic chemicals, o-tolidine and 4,4'-tetramethyldiaminodiphenylmethane (TDDM). o-Tolidine and TDDM were both tested in the Salmonella/mammalian-microsome test and in the sister-chromatid exchange (SCE) test with rabbit lymphocytes in vitro. The number of revertants was increased in the presence of S9 mix by o-tolidine in the Salmonella typhimurium strains TA98 and TA1538, and by TDDM in strains TA98 and TA100. Both compounds showed weak SCE-inducing activity in cultured rabbit lymphocytes in the absence, but not in the presence, of an exogenous metabolic system.

Sister-chromatid exchanges induced in vitro by cyclophosphamide without exogenous metabolic activation in lymphocytes from three mammalian species.

The induction of sister-chromatid exchanges (SCEs) by cyclophosphamide (CP) in human, rat and rabbit lymphocytes was studied in vitro without exogenous metabolic activation. In contrast with published reports, CP induced SCEs in lymphocytes from all three species, indicating that cultured lymphocytes possess the metabolic capacity to convert this indirect-acting mutagen.

Early detection of asymptomatic hereditary chemodectoma with radionuclide scintiangiography. A possibility for family screening and surveillance.

A large family with a history of hereditary chemodectoma has been successfully screened by means of radionuclide scintiangiography. This procedure could restrict selective and more invasive roentgenangiography only to those cases in which flow patterns are abnormal. Three asymptomatic cases of chemodectoma were revealed by this method. Early therapeutic measures may now be feasible. We discuss the possibilities for surveillance of these families without causing discomfort to the group involved.

The role of the pituitary-ovarian axis in the termination of early pregnancy in rabbits treated with ethinyl-oestradiol.

The pathogenesis of embryonic death following maternal treatment with ethinyloestradiol was investigated. Pregnant rabbits received daily oral doses of 0.4 mg/kg body weight of ethinyloestradiol starting on day 6 of pregnancy. Dams were autopsied after 3, 6, or 9 days of treatment. Uterine swellings, ovaries, and the pituitary gland were examined using histochemical techniques. The results showed that in the treated animals there were morphological signs indicating decreased secretory activity of gonadotrophin and prolactin producing cells of the pituitary gland, luteolysis, and embryonic death within the first few days of treatment. The present study indicated that termination of early pregnancy in rabbits is most probably due to the interference of ethinyloestradiol with the pituitary-ovarian axis which in turn results in luteolysis and impairment of luteal steroid synthesis.

Just audible pitch and loudness modulations with cochlear impairment.

Just noticeable combined modulations in amplitude and frequency were determined for an observer (DP) with a bowl-shaped sensorineural hearing loss of about 40 dB between 1 and 2 kHz. In all stimulus conditions, interactions were found between the detection of amplitude modulations and frequency modulations. These interactions appear to correlate quantitatively with the effect of frequency on loudness (as given by the slope of the equal-loudness contour). When this effect of frequency on loudness if taken into account, the data support the hypothesis of independent detection of pitch and loudness modulations. The experimental data are also discussed in relation to the single-mechanism hypothesis for the detection of changes in amplitude and frequency (Zwicker, Maiwald). This model could not provide a satisfactory explanation of interactions between amplitude and frequency.

Screening of a family for chemodectoma.

59 relatives with a family history of hereditary chemodectoma have been successfully screened by means of radionuclide scintiangiography. The findings showed a close correlation with selective roentgenangiography. 3 asymptomatic patients with chemodectoma were revealed by these methods without causing discomfort to the group involved.

S--s--U-- phenotype in South African Negroes.

A Negro woman of the Xhosa tribe of Port Elizabeth in the Eastern Cape was found to be S--s--U--with anti-U in her serum. She had two S--s--U-- children, and her husband, father and other children all had single doses of S or s antigen. Three furhter S--s--U-- Negroes were found in a random sample of 1,000 Negro antenatal patients at Port Elizabeth.

Reversible harmless interruption of testicular blood supply in the ram.

An effective method of interrupting testicular blood flow temporarily and repeatedly in the ram has been developed. Blockade of flow has been achieved mechanically by an inflatable occluder placed around the testicular artery at the level of the spermatic cord. The effect of the blockade on total testicular blood supply was investigated using Doppler flowmetry and a percutaneous Xenon-133 injection method. With both approaches, the blood flow changes after inflation or deflation of the occluders could be estimated satisfactorily. A substantial decrease of testicular blood flow was achieved in eight of the 10 testes with inflated occluders. However, there were indications that in the remaining two testes blockade of the arterial flow was not complete. After deflation of the occluders, blood flow was restored rapidly and completely in all testes. Macro- and microscopic examinations revealed no long-term damage to the testis after blood flow interruptions lasting 30 or 60 minutes.

Blood damage, platelet and clotting activation during application of radiofrequency or cryoablation catheters: a comparative in vitro study.

During catheter ablation of cardiac tissue, the insulting stimulus may damage and activate the blood. Specifically activation of platelets and the clotting system is a potential risk by their formation of thrombi. In this study the effect of two different techniques, a radiofrequency and a cryo-application procedure, on the activation of platelets and clotting in an in vitro blood circulation model was investigated. The radio-frequency procedure induced significantly more blood cell damage, platelet activation and clotting than did the cryo-application procedure. Macroparticles were circulating in blood after the radiofrequency procedure. In the cryo procedure, blood damage was limited to the frozen blood spherule around the tip of the catheter. It is concluded that at least in those circumstances where blood is directly contacting the tip of the radiofrequency or cryo-application catheter, the latter is more safe with regard to thrombus formation.

Effect of oral calcium and vitamin D on glucocorticoid-induced osteopenia.

Bone-loss in glucocorticoid-induced osteopenia is most evident in trabecular bone such as the vertebrae. Bone mineral content (BMC) measurements of the spine were prospectively studied, comparing the preventive effect of calcium (500 mg a day) and calcium plus Vitamin D (4000 IU on alternate days) during long-term corticosteroid treatment. A small increase in BMC was noted in both groups, but no differences between preventive calcium and calcium plus Vitamin D became evident. It is suggested that patients who are receiving prednisone should be advised to use supplemental calcium.
