[Evaluation of phagocytic activity of peripheral blood neutrophils in patients after kidney transplantation treated with cyclosporin and prednisone or with azathioprine with prednisone].

In 32 renal transplant patients neutrophil phagocytic activity was evaluated using living bacterial cells of Staphylococcus aureus Oxford 209 P strain. The total group of patients was divided into two subgroups according to the kind of the applied immunosuppressive treatment 1) cyclosporin--12 patients treated with cyclosporin A along with prednisone and 2) azathioprine--20 patients treated with azathioprine along with prednisone. The index of phagocytizing cells i.e. the percentage of neutrophils phagocytizing the bacterial cells was assessed in the investigation of phagocytosis. In the renal transplant patients as compared with the control group, statistically significant increase in the phagocytic of peripheral blood neutrophils in relation to the used bacterial cells of Staphylococcus aureus Oxford 209 P strain was found. Statistically significant differences in the investigated range were also present between the compared subgroups. In the cyclosporin subgroup statistically significant increase was revealed in the percentage of phagocytizing neutrophils.

[Lysozyme activity in undiluted and diluted blood of patients treated with cyclosporin A with prednisone or azathioprine with prednisone after kidney transplantation].

It is a general opinion that the renal transplant patients may show the decrease of immunity which manifests by their increased susceptibility to bacterial, viral and fungal infections. One of the important non-specific defensive factors of the organism participating in the protection against infections is lysozyme. It is the purpose of the present paper to estimate the lysozyme activity in the un- and diluted serum in the renal transplant patients and to determine the possible mechanism of the observed deviations. 32 renal transplant patients (24 men and 8 women) aged of 16 up to 50 were investigated, being in the period of 1 until 4 years after transplantation. In the investigation the kind of immunosuppressive treatment used was considered which served the purpose of dividing the total group of patients into two subgroups: one treated with cyclosporine (12 patients) and the other treated with azathioprine (20 patients). The control group consisted of 20 healthy persons, matching by age, sex and living place the group investigated. In both groups investigated and control, were determined: lysozyme activity in the serum of the undiluted and diluted blood, total leukocyte number, the absolute number of peripheral blood neutrophils as well as their adherence to fibres was evaluated. In the renal transplant patients compared with the control group and in the separated subgroups, no statistically significant differences were found concerning leukocyte count, absolute neutrophil count and adherence of those cells.(ABSTRACT TRUNCATED AT 250 WORDS)

[Cytochemical properties of peripheral blood neutrophils in patients with hyperthyroidism].

In the cytoenzymatic investigations of peripheral blood neutrophils in patients with hyperthyroidism there was found the increase of acid phosphatase activity, beta-glucuronidase, leucine aminopeptidase, and catalase moreover there was found the decrease of the activity of alkaline phosphatase. After a two-week treatment with thiamazole (methimazole++) 50 mg in 24-hour dose there was observed the decrease of acid phosphatase activity in neutrophils. During incubation of plasma containing leucocytes, from healthy persons, with L-thyroxine there was observed the increase of the activity for acid phosphatase and beta-glucuronidase. In patients with hyperthyroidism there appear many changes of enzymic equipment of neutrophils which are concerned with lysosomal and connected with cell membrane enzymes. The results of cytochemical investigations after application of thiamazole and no difference, with exception of catalase, between patients with Graves-Basedow disease and with toxic goitre and the results of investigations in vitro with L-thyroxin point out, that there is the possibility of connection between the observed changes in the range of enzymic equipment of neutrophils and the hormonal state of the investigated group of patients.

[Bactericidal activity of plasma and leukocytes in patients after kidney transplantation].

In renal transplant patients the plasma and leukocytes bactericidal activity was estimated towards the vivid bacterial cells of the standard strain of Staphylococcus aureus Oxford 209P. Considering the kind of the immunosuppression used, the patients were divided into two subgroups. The first subgroup comprised the patients treated with cyclosporin A and prednisone (12 persons) and the second one--the patients treated with azathioprine and with prednisone (19 persons). The control group consisted of 20 healthy persons. In the renal transplant patients significantly more bacterial cells survived the incubation with plasma and peripheral blood leukocytes and the reduction of bactericidal activity of the leukocytes alone was also found. The reduction of bactericidal activity of the peripheral blood leukocytes observed in the patients of the group tested may have resulted from the inefficient microbicidal systems of those cells. The kind of the immunosuppressive treatment did not seem to affect the bactericidal activity of the leukocytes and plasma examined in the renal transplant patients peripheral blood. It may be supposed that the increased susceptibility of those patients to bacterial, viral and mycotic infections is due in part to the reduced bactericidal capability of the peripheral blood leukocytes.

Neutrophil function in chemical plant workers employed in the production of dust pesticides.

In two divisions of a chemical plant producing dust pesticides, employees exposed to dust, containing 28%-65% SiO2, were examined. In the first division (group I, 38 males and 35 females), the average air dust concentration was 5.0 mg/m3, and the active substances were: chlorinated hydrocarbons, captan, carbamates and dodine. In the other division (group II, 26 males and 33 females), the air dust concentration was 4.8-5.2 mg/m3, and the active substances included: carbamates, triazine compounds, cupric oxychloride, captan, lindane, carboxine. Spirographic investigations showed signs of pulmonary emphysema (RV/TLC) in 65.8% males and 60% females of group I and 38.5% males and 40.6% females of group II. In leucocyte concentrate smears, the cytochemical reactions to beta-glucuronidase, acid phosphatase and myeloperoxidase, as well as the nitroblue tetrazolinum (NBT)-dye reduction of neutrophils were performed. The random migration and chemotaxis of isolated neutrophils, washed or incubated in 10% autologous serum, their phagocytic activity and tube adherence test were also investigated. Compared to the controls, the reaction to beta-glucuronidase as well as the NBT reduction were increased, whereas the acid phosphatase and myeloperoxidase reactions were lowered. Impairment of neutrophil chemotaxis stimulated with zymozan-activated serum was observed in all groups of workers; random migration was enhanced in workers of group I and lowered in male workers of group II. Higher phagocytosis of latex particles occurred in workers of group I and in males of group II, while tube adhesion was impaired in group I and enhanced in males of group II.

[Spontaneous migration and adherence of leukocytes and the NBT test of peripheral blood neutrophils in patients with iron deficiency anemia].

Literature reports suggest that iron-deficiency may contribute to the development or intensification of the condition termed generally "immunodeficiency". In the available literature no reports were found on a precise evaluation of the functions of peripheral blood neutrophils in iron deficiency anaemia. The purpose of the presented study was: 1) estimation of the adherence of neutrophils from peripheral blood to fibres, 2) evaluation of the indices of spontaneous and latex-stimulated NBT reduction by the neutrophils, and 3) the test of spontaneous migration of peripheral blood leucocytes. The study of 20 patients with iron-deficiency anaemia, with counts of total leucocytes and absolute neutrophil count in peripheral blood showed increased adherence of the neutrophils and reduced spontaneous migration of leucocytes in the 3-hour test, but no changes in the test of spontaneous and latex-stimulated NBT reduction. In the discussion an interpretation of the observed abnormalities in the studied biological functions of the neutrophils in patients with iron-deficiency anaemia is tentatively put forward, stressing that in this anaemia despite absence of changes in the counts of peripheral blood neutrophils, spontaneous migration of peripheral blood neutrophils in the 3-hour test is reduced, perhaps as a result of increased adherence of neutrophils.

[Evaluation of the phagocytic activity of peripheral blood neutrophils in patients with hyperthyroidism].

In 80 women with hyperthyroidism (40 with diagnosed Graves disease and 40 with hyperactive nodular goiter) the following tests related to the function of peripheral blood neutrophils have been carried out: 1. nitrotetrazolium blue (NRT) reduction test. 2. evaluation of phagocyting activity by using latex particles and living bacteria cells, and 3. determination of immunoglobulin concentration and the C3 component of the complement in blood serum. The following features were found in the patients with hyperthyroidism: 1. the elevated values of the index of spontaneous NBT reduction which return to normal following the treatment with propranolol or metizol lasting 14 days, 2. a decrease in the phagocyting activity of neutrophils occurring with stimulation of phagocytosis by both the latex particles and bacteria cells. 3. the return to normal values of the index of neutrophils phagocyting the latex particles following two-week treatment with propranolol or metizol. It was concluded that in patients with hyperthyroidism the changes in NRT reduction and phagocyting activity of peripheral blood neutrophils return to normal following the two-week treatment of these patients with both propranolol and metizol.

[Peripheral blood neutrophils in automobile tire factory workers in the light of cytochemical studies].

In 73 workers producing rubber mixtures and tubes the number and percentage composition of white peripheral blood cells as well as the NBT reduction test and cytochemical reactions to lactic (LDH) and succinic (SDH) dehydrogenases in neutrophils were estimated. The amount and percentage composition only slightly differed from controls. The NBT reduction index and the reactivity of LDH and SDH tests in neutrophils were elevated.
