[Liver mitochondrial phospholipase activation in hyperthyroid rabbits].

Mitochondria obtained from hyperthyroid rabbit liver contained an approximately 4-fold greater amount of free fatty acids (FFA) than preparations from the control animals. The temperature dependence of FFA accumulation (29--38 degrees C) showed that the activation energy of mitochondrial lipid hydrolysis for control and experimental animals was 7 kcal/mole and 29 kcal/mole, respectively. The mitochondria were kept at 3 degrees C for 48 h. During this period there was an accumulation of fatty acids. The rate of the process in the mitochondria of hyperthyroid animals was several times than that in the control ones. This indicates the activation of mitochondrial phospholipase by the liver in hyperthyrosis. The increased mitochondrial phospholipase activity seen in hyperthyrosis is assumed to be determined by changes in the physical properties of lipid membranes.

[Accumulation of lipid peroxidation products and suppression of the retinal electrical activity of vitamin E deficient rats exposed to high intensity light].

Induction of alimentary vitamin E deficiency in rats is accompanied by accumulation of lipid peroxidation products in the retina in vivo and by reduction of the magnitude of electroretinogram waves. Photodamage to the retina caused by the action of light of high intensity (10 000 lx for 3.5 h) is more pronounced in vitamin E deficient animals than in control rats fed standard laboratory chow. The exposure of rats to the light of high intensity results in accumulation of lipid peroxides, the magnitude of which is far greater in vitamin E deficient animals than in controls. The photodamage process is reversible and by the end of the 14th day after exposure to light the content of lipid peroxidation products in the retina and its electric activity are close to the values found in controls.

[Asymmetry of aging of the large hemispheres of the rat brain].

The dynamics of the accumulation of free-radical oxidation products in the brain hemispheres of rats in rate ontogenesis was studied. Multiphase changes in the concentration of products in the right and left hemispheres in different age groups were registered. Significant differences in the contents of hydroperoxides, Schiff's bases of phospholipids and lipofuscin in brain hemispheres revealed, which correlates with different rate of ageing of hemispheres.

[Synchronization of changes in the level of direct current potential and concentration of lipid peroxidation products in the developing rat brain].

The dynamics of the level of direct current potentials and the content of lipid hydroperoxides, conjugated dienes, Schiff bases of phospholipids and lipofuscin in the brain of rats during ontogenesis was studied. It is shown that changes in neurophysiological and biochemical characteristics have an M-shaped character. The synchronization of age-dependent changes in the content of lipid peroxidation products and the dynamics of direct current potentials was revealed.

[Systemic organization of functional interhemisphere asymmetry. Mirror asymmetry].

The systemic organization of hemispherical asymmetry at the behavioral, neurophysiological, and biochemical levels was studied. An interconnection and hierarchic coordination of the parameters of different levels were revealed, which are confirmed by multiple correlations between them. An interrelation between the processes of peroxidation of brain membrane lipids, the level of direct current potential, and the behavior of animals was shown. A significant difference in correlations between right-hander and left-hander animals was found.

[Dynamics of functional interhemisphere asymmetry and sexual dimorphism in rat ontogenesis].

The dynamics of functional interhemispherical asymmetry in the ontogenesis of rats, males and females separately, was studied. The asymmetry was determined by the neurophysiological criterion, the interhemispherical gradient of the distribution of direct current potential of the brain. The sexual dimorphism upon age-related changes in the interhemispherical asymmetry was shown. It is assumed that this asymmetry is involved in the dynamics of neuroendocrine interactions determined by sex.

[Morpho-biochemical study of the liver and serum albumins during tetrachloromethane intoxication and administration of antihypoxants, antioxidants, and actoprotectors].

Organism intoxication with tetrachloromethane induces a complex of transformations in the liver and serum albumins system manifested in the liver as necroses in the central lobe, dilatation of sinus capillaries and blood stasis in them, increased content of total water, and changed magneto-relaxation properties indicating decreased structurization of water. The system of serum albumins shows decreased total concentration of albumins in the serum, decreased constant of 1-(phenylamino)-8-sulfonaphthalene probe binding, increased mean number of probe binding per albumin molecule, and increased accessibility of probe in the protein-probe complex for the quencher indicating changed type of interaction between the protein and fluorescent probe. Introduction of a antihypoxant, antioxidant, or actoprotector has a positive effect on the condition of the liver and serum albumins system: The studied indices are nearly completely normalized.

[Method for assessing involution ophthalmoendotoxicosis by fluorescent analysis of lacrimal fluid].

A method for evaluating the binding capacity of aqueous humor albumin as endoophthalmotoxicosis indicator is proposed, consisting in evaluation of this parameter in the lacrimal fluid by fluorescent probe K-35. A correlation between the studied parameter in aqueous humor, lacrimal fluid, and blood serum has been detected. Albumin binding capacity in the above biological fluids of patients with involution ophthalmopathy was low.

[Nitrogen oxide in the pathogenesis of glaucoma and cataract].

The content of nitrite (final metabolite of nitroradical) was measured in aqueous humor of patients with glaucoma and cataract. The level of NO formation is increased at the initial stage of glaucoma and decreases with the disease progress. A direct correlation between the level of nitrite in aqueous humor and coefficient of easiness of its discharge was detected. Cataract is associated with a high level of nitrite in intraocular fluid. The results are discussed from the viewpoint of ocular hydro--and hemodynamic regulation and NO involvement in the free radical oxidation reactions, playing an important role in the pathogenesis of both diseases.

[A computer method for assessing the average rate of movement of Paramecium caudatum by a cross-sectional light beam].

The possibility of using the motility of Paramecium caudatum for computer biotesting of toxic substances was studied. The method is based on measuring the number and duration of impulses that arise when protozoa chaotically moving in a layer 0.5 mm thick cross a red light beam (1 mm in diameter). It was found that, in the presence of Cu2+ ions and menadione bisulfite, a compound having prooxidant properties, the number of impulses decreased with a concurrent increase in the duration of impulses. The high correlation between the number of impulses and the reciprocal of impulse duration indicates that the motility of paramecium by the action of Cu2+ ions and menadione slow down, and even an immobilization of some of them occurs. The mean swimming velocity of protozoa was reduced by half within 20 min by the action of Cu2+ at a concentration of 90 microM (6 mg/l of Cu2+ ions) and within 80 min by the action of menadione bisulfite at a concentration of 50 microM. It was shown that the slowing down of motility, which indicates the presence of a toxicant in the medium, is a faster physiological reaction than cell death.

[Distribution of small hydrophobic molecules in membranes. I. Artificial lipid membranes].

A hydrophobic uncharged fluorescent probe of 4-dimethylaminochalcone (DMC) interacted with synthetic phospholipid membranes. Comparison of absorption spectra and fluorescence of DMC in the membranes and organic solvents shows that in the membranes the DMC molecules are located not in the hydrocarbon layer but in the polar regions near the surface. The probe is distributed regularly along the surface forming no dimers and clusters. Polar groups which surround the probe in the membrane are less mobile than the molecules of organic solvents at the same temperature. The evaluation shows that the relaxation time of polar groups in the probe environment is longer than 0.15-10(-9) sec. The DMC molecules may be located in different sites of the membrane surface, which seem to differ from one another in the mobility of polar groups.

[Study of phenothiazine binding to membranes by the fluorescent probe method].

Binding of chlorpromazine, etaperazine and triftazine with model phospholipid membranes was investigated by using a 3-methoxybenzanthrone (MBA) fluorescent probe. All these preparations interacted with the membranes, this being registered by the MBA fluorescence extinction. The phenothiazine molecules bound with the membrane are localized in its superficial layer and extinguish the MBA fluorescence in different ways. The chlorpromazine binding was heterogenous and there are, apparently, 2 different ways of its molecules combination with the membranes. The effective extinction of MBA by phenothiazine increased in the following order: chlorpromazine, etaperazine, triftazine.

[Comparative study of the hemolytic action of group D vitamins].

Hemolytic effects of the vitamins of the D group (ergocalciferol - D2, cholecalciferol - D3, and 1 alpha-hydroxycholecalciferol - 1 alpha-OH-D3) were compared using rat erythrocytes. The latent period of hemolysis, caused by 1 alpha-OH-D3 was approximately 3-fold shorter as compared with the hemolysis caused by D2 or D3. Apparent values for energy of activation of hemolysis, estimated at 30-45 degrees, constituted for D2 - 23.2 kcal/mol, D3 - 25.3 kcal/mol, 1 alpha-OH-D3 - 13.1 kcal/mol. Despite the higher hemolytic activity, the rate of 1 alpha-ON-D3 autooxidation was distinctly lower than that of D3.

[Photo damage to the eye in exposure to the radiation from a Nd:YAG Q-switching laser: the physicochemical structural changes to the crystalline lens and the vitreous body].

Structural changes in the lens and vitreous body exposed to short-pulse Nd:YAG Q-switching laser were under study. The laser was focussed in the lens nucleus or vitreous center plane. A pulse energy was 7.1-9.3 mJ, with a total of 75-100 pulses. Cataract development was induced via the formation of cavities with the guidance spot focal plane localized in the lens nucleus plane. When the focus was in the vitreous body and the laser operated in a similar energy mode, great numbers of small cavities rapidly formed, this evidencing a shock wave propagation. Specific and structural conformational changes in the lens and vitreous protein molecules were detected by nitrate quenching of the triptophane amino acid residue fluorescence. Laser exposure was found to reduce triptophanile availability for nitrates, this evidencing protein complexes aggregation (collapse); besides, laser exposure essentially increased the amino acid residue quenching constants, which fact pointed to a decreased density of the vitreous collagen and lens crystalline negative charges (increased hydratation). These findings permit a conclusion that the shifts connected with injury to the vitreous body, with macular edema, or with detachment of the retina after exposure to Nd:YAG laser may be due to collapse of the vitreous gel liquified components.

[Effect of the structure of liposomal membrane surface layer on the binding of phenothiazines].

By using a fluorescent 3-metoxybenzatrone probe the binding of the phenothiazine series drugs (chlorpromazine, triftazine, aethaperazine) with phospholipid vesicles--liposomes, was investigated. Cholesterol is shown not to affect the binding of these drugs with liposomes. The surface charge of liposomes influences only the combination with the chlorpromazine membranes. With a higher positive charge of the membranes the binding of chlorpromazine diminishes in force.
