[The mechanisms of the curative action of systemic enzyme therapy].

Employment of the authors' highly sensitive biochemical method allowed the fact of resorption of proteolytic enzymes after their peroral ingestion to be confirmed, with a question having been raised as to further fate of proteinases and mechanisms of curative action of combined enzyme preparations. The analysis of relevant data from published literature and the results of the author's own studies permitted identifying the most important moments of realization of the systemic enzyme therapy effects, which are as follows: 1) activation of receptors on the surface of enterocytes; 2) formation of activated alpha 2-macroglobulin (alpha 2-M) under the action of proteinases from the intestines, and realization of catalytic activity of those enzymes involved in this restrictor of proteinases; 3) modulation of cell activity by complexes alpha 2-M-proteinase; 4) activation of the system of mononuclear phagocytes; 5) influence on the balance of bodily endogenous proteolytic systems.

Autofibrin glue compound and its utilization during reconstructive operations on the ear.

A new autofibrin glue compound (AFGC) is suggested. The experiment has demonstrated that the inclusion of antibiotics and lysozyme does not influence its adhesive qualities or sterilization with gamma radiation. It has been revealed that the dose to sterilise the compound was 75 Gy. AFGC was used during tympanoplasties in 55 patients for fixation of ossiculoplasties, fascia autotransplant and skin of the external auditory meatus. Morphological and functional results of tympanoplasties turned out to be better than those of the control group in which the glue was not used.

[Use of Soviet fibrin adhesive in liver surgery in experimental animals].

The results of a comparative experiment for studying the native glue composition on a fibrin base, which possesses the hemostatic and adhesive properties and has the influence on the processes of regeneration, and modern synthetic medical glues are presented. The essential advantages of a fibrin-based glue were revealed.

[The effect of acute alcoholic intoxication on the body's immunologic and biochemical indices].

Acute experimental intoxication in rats produced essential changes of the absorptive capacity of neutrophilic blood granulocytes, proteolytic activity, level of proteinase inhibitors and content of blood plasma fibrinogen. Female rats showed a distinct reduction of the activity of natural killer cells. Alcohol had no essential effect on the production of hemolytic antibodies as well as reagins in the respiratory organs.

[Contact activation of prekallikrein and plasminogen in plasma].

Activation of the Hageman factor-prekallikrein system in the whole human blood plasma is studied as affected by organic silica (aerosils) with anionic and cationic properties. Positive- and negative-charged aerosils are shown to possess the same ability to activate prekallikrein. Activity of prekallikrein was manifested in hydrolysis of the chromogenic substrate--Benz-Pro-Phen-Arg-paranitroanilide . HCl, kininogen and protamine sulphate formed by kallikrein. The data permit supposing that optimal activation of the Hageman factor requires the polar (but not ionic) groups with hydrophilic properties on activating surfaces. Plasminogen under contact activation, in contrast to prekallikrein is activated only in the diluted plasma (pH 4.8), and not completely. Possible mechanisms of the contact activation and interaction of the Hageman factor, prekallikrein and high-molecular kininogen in this process are discussed.

[Effect of blood plasma inhibitors on activity of serine and cysteine proteinases].

Data on study of action plasma inhibitors on activity of pancreatic proteolytic enzymes (trypsin, chymotrypsin) and plant proteinases (papain, bromelain), included in composition of enzyme mixes, used for orally application are submitted. It is established, that serine proteases are more sensitive to inactivation of plasma inhibitors, than cysteine enzymes. Main inhibitor of the papain and bromelain is alpha-2-macroglobulin in complex with which they preserve significant part of initial activity. A high-sensitivity method of determination of activity enzyme combinations, enabling to detect nanograms of them in presence of plasma inhibitors is offered. It can be used for study pharmacokinetic and optimization of enzyme mixes application in clinical practice.
