[Morphobiochemical study of the liver in systemic endotoxinemia].

Dog liver in systemic endotoxemia induced by intravenous injection of E. coli lipopolysaccharide in 2 mg/kg dosage has been studied. Morpho- and pathogenetic characteristics of developing intoxication have been specified histologically and biochemically.

[Structural-metabolic characteristics of acute hepatic failure].

Basing on early autopsy data, the study was made of structural and metabolic characteristics of the liver in marked intoxication and blood loss. Histologic, histoenzymatic and biochemical methods were used to evaluate morpho- and pathogenesis of acute liver failure arising in the above conditions.

[The histophotometric characteristics of the hepatocytes in acute blood loss and pulmonary artery thromboembolism (based on the data from early autopsies)].

A comparative morphological study of the liver in acute blood loss and massive thromboembolism of the pulmonary artery was carried on basis of early autopsies. Histoenzymatic and correlation analysis was used to study the characteristic features of the metabolism of hepatocytes in conditions of acute blood loss and massive pulmonary artery thromboembolism.

[Hepatic insufficiency in patients with peritonitis].

An examination of 14 patients with general forms of peritonitis of different genesis was performed, morphological alterations in the liver tissue and changes of activity of intracellular enzymes of hepatocytes were estimated by groups of living and dead patients. A number of biochemical parameters of the patients' blood plasma were analyzed in parallel. It was established that the development of peritonitis was followed by deep morphological and metabolic disorders.

[Changes in hepatocyte dehydrogenase activity in different zones of liver acini during the revascularization of ischemic extremities].

After ischemia (3, 6, 9 and 12 h) of the hind extremities in dogs with a subsequent revascularization for 2 h, enzymatic activity of hepatocytes changes. After ischemia for 3 h the enzymatic activity increases. Restoration of the blood flow at later stages of the experiment results in a progressive decrease of dehydrogenase and diaphorase activities. To a greater extend the changes of the enzymatic activity are observed in perivenular hepatocytes (in the 3d zone of the hepatic acinus).

[Morphometric analysis of the toxic effect of ethanol on the structure of chromatin in interphase nuclei of rat hepatocytes].

A method of automated morphometric analysis of hepatic cell interphase nuclear chromatin (CHIN) structure was elaborated using scanning microspectrophotometer SMP-05 ("Opton", FRG). Liver sections of rats were used to investigate hepatic cell CHIN structure. The animals were administered ethanol intraperitoneally once per day within a month period in doses 4, 6, 8, 10 g/kg (1/3-4/5 LD50). Morphometric indexes indicating dose-dependent changes in hepatic cell CHIN structure are demonstrated. Statistical analysis of morphometric data has led to the conclusion that hepatic cell CHIN structural modification depends on two factors: ethanol damaging effect and hepatic cell regeneration.

[Changes in the dehydrogenase activity of cardiomyocytes during acute experimental arterial occlusion of extremities].

The results of quantitative histoenzymologic investigations of succinate dehydrogenase, lactate dehydrogenase and NAD-diaphorase in cardiomyocytes of dogs with acute experimental arterial occlusion in ischemic and postischemic periods are reviewed. An increased activity of dehydrogenases in the early periods (3,6 h) of ischemia and during recirculation was established, with its noticeable reduction at later terms (9,12 h). Medical correction of postischemic disorders was shown to improve cardiomyocyte metabolism.

[Cytophotometric analysis of the chromatin structural conformity in interphase nuclei detected in UV light and by gallocyanine staining].

Geometric and optical parameters of chromatin of hepatocyte nuclei have been examined before (UV, lambda = 265 nm) and after gallocyanine staining. Quantitative parameters of the chromatin structure in the same nuclei measured in situ by a scanning microscope-photometer (step size 0.125 micron) before and after staining were equal. Tinctorial properties of chromatin granules (condensed part of the nuclear material) and its diffuse part were different. It is suggested that the difference between granules and the nongranular part of chromatin is not only of optical but also of chemical nature.

[Quantitative changes in liver dehydrogenase activity during temporary ischemia of the extremities and in the postischemic period].

Experimental temporary occlusion of the dog hind limbs was produced. Histoenzymological changes in the liver induced by the temporary ischemia of the limbs and by ischemia with a 2-hour revascularization were subjected to comparative histophotometric examination. The data obtained indicate an appreciable rise in the enzymatic activity of hepatocytes at the early stages of experiments and increasing reduction in metabolic processes at the later stages. A substantial decrease was noted in the activity of the liver enzymes under study, with that decrease being especially pronounced at the later times in experiments with limb revascularization, which is accounted for by a far greater release to the bloodstream of metabolic products from the ischemized limbs.

[Quantitative informational estimation of the degree of liver damage on a model of chronic alcoholic intoxication].

In the rat liver at a chronic alcoholic intoxication produced with various doses of ethanol, a double phased picture of informational characteristics has been revealed in the cellular nuclei under a linear increase of the dose. An analysis of corresponding distribution of the hepatocytic nuclei has been performed according to their size and informational characteristics. On this base, the limiting dose of ethanol is estimated for development of the hepatic alcoholic lesion.
