[Determination of HIV RNA concentration for evaluation of the efficacy of combined therapy of HIV infection].

Plasma HIV RNA (viral load) and count of CD4+ T cells were evaluated in 23 patients with HIV infection treated with invirase (1800 mg/day), zidovudine (600 mg/day), and zalcitabine (2.25 mg/day) for 6 months in order to evaluate the efficiency of antiretroviral therapy. Viral load was measured by AMPLICOR HIV-1 Monitor test. The reproducibility of HIV RNA measurements was in line with reported data (CV 15-41%), allowing highly accurate (15%) evaluation of RNA in a standard control sample provided by National Institute for Biological Standards and Control, Great Britain. Plasma HIV RNA concentration decreased to an undetectable level (below 400 RNA copies/ml plasma) after 6 months of treatment in 52.2% patients. In 17.4% the therapy failed, and in 30.4% it resulted in a reduction of viral load to > 1 lg, although HIV RNA was still detected in the plasma after 6-month therapy. The count of CD4+ T cells increased by 9.5%. Changes in the viral load outstripped changes in CD4+ cells. Viral load was in high correlation with the count of CD4+ lymphocytes: -0.53, p = 0.01 before treatment and -0.61, p = 0.002 after 6-month treatment.

[The etiological structure and characteristics of liver involvement in patients with HIV infection].

Hepatic lesions were analyzed in 33 patients with HIV-infection. The patients were divided into two groups by the disease stage: early (stage IIB, IIIA, n = 12) and late (stage IIIB and IIIC, n = 21). Markers of hepatitis A, B and C were found in 42.4% of patients. Patients of group 1 had acute and chronic viral hepatitides (75%), hepatic alcoholic damage. Patients of group 2 developed combined hepatic lesions resultant from generalized bacterial, fungal and parasitic infections (66.7%), chronic hepatitides and viral cirrhoses (33.3%), alcohol abuse (33.3%). Elevated levels of the enzymes (AsAT, AlAT, LDG) at early stages of HIV-infection were brought about by hepatic involvement while at late stages by polyorganic abnormalities.

[Immunomorphologic and ultrastructural features of the thymus in myasthenia].

Morphologic, ultrastructural changes in the epithelial component of the thymus in 41 patients with myasthenia are analyzed. Studies by histochemical, ultrastructural, and immunofluorescent methods revealed that the severity of the clinical course of myasthenia was related to the degree of changes in the epithelial component of the thymus, in its specific structures, Hassal's bodies. Effect of thymic epithelial component on lymphoid immunogenesis-regulating component was detected.

[Interaction of insulin with receptors in insulin-independent diabetes mellitus].

Parameters of insulin binding with its specific receptors were determined in small concentrations of peripheral blood mononuclear cells. Examination of 22 patients has shown that the higher the level of insulinemia and the degree of obesity in the insulin-independent form of diabetes mellitus, the lower insulin binding. A decrease in the binding results from the reduction of the number of binding sites and partially from the affinity of type I receptors for insulin. Causes of a decrease in the number of receptors in patients with diabetes mellitus who are not suffering from obesity are discussed.

[Rosette-forming ability of virus-treated human leukocytes (V-rosettes)].

Rosette-formation with auto- and allogeneic red blood cells was applied to detection of human leucocyte subpopulations interacting with Sendai virus (V-rosettes). It was shown that the majority of V-rosette-forming cells appeared to be monocytes. T lymphocytes did not take part in V-rosette-formation since selective elimination of T cells from the mononuclear cells population did not lead to reduction of but increased the number of V-rosettes. Enrichment of cell suspension with B lymphocytes was followed by a rise in the number of V-rosettes thereby allowing the attribution of B lymphocytes along with monocytes to the cell population interacting with virus. The results suggest that ability of virus-exposed immunocompetent cells to react with their own red blood cells may lie at the basis of the development of autoimmune hemolytic anemia and other autoimmune diseases.

[Origin of immunoglobulins bound to the surface membranes of malignant cells in non-lymphoid forms of acute human leukemia].

Anti-immunoglobulin antibody binding to the surface membrane of blast cells from patients with acute myelomono- and monoblastic leukemia has been examined. Surface receptors for IgG Fc-fragment have been demonstrated to be not responsible for anti-immunoglobulin antibody binding and blast cells not capable of intracellular synthesis of immunoglobulins. It has been shown that antigen-antibody complexes appear in the medium after a short-term cultivation of blast cells and that immunoglobulins appear in the eluates after treating the blast cells with an acid buffer. Analysis of the results indicates that immunoglobulins detectable on the surface of the blast cells in the cited forms of leukemia are antibodies to the antigens of these cells.

[Determination of human leukocyte interferon by a microfluorimetric immunologic method].

The possibility of quantitative determination of human leucocyte interferon using FITC-labeled antiinterferon antibodies was studied. A highly specific antiinterferon immunoglobulin was obtained as a result of longterm immunization of a donkey with human leucocyte interferon followed by fractionation and immuno-absorbtion of immune plasma. This immunoglobulin was labeled with FITC and used for human leucocyte interferon assay in direct and indirect reactions of fluorescence immunoinhibition. The titres of different human leucocyte interferon preparations in this immunoassay were comparable with the titres of the same preparations detected by interferon inhibition of viral cytopathic effect.

[Production of monospecific immunoglobulin against human leukocyte interferon].

Donkeys were regularly, at seven-day intervals, immunized subcutaneously with human leukocyte interferon (IF) with the activity of 1.6 X 103 U/10 ml. IF-neutralizing antibodies in the titre of 1:128-1:256 were detected in the animals' sera after 38-40 injections. As a result of continuing injections the titre of these antibodies increased considerably. The antibodies to the components of the system in which the IF was obtained were revealed in parallel. Donkey antiinterferon plasma was prepared by plasmapheresis and antiinterferon immunoglobulin (AIFIG) was released from the plasma by centrifugation using ammonium sulphate at 50% saturation. Contamination antibodies were removed from AIFIG by affin chromatography on combined immunosorbent.
