A contemporary review of the factors involved in complete dentures. Part III: support.

Dentists must base their technique on an understanding of the biologic aspects of the relationship between the denture base and supporting tissues. Those tissues must be able to tolerate functional stresses without promoting patient discomfort and should be recorded in such a manner that these areas provide complete denture support. Anatomic regions that satisfy the requirements for providing primary support should make positive contact with the denture base under functional loading. Those that are less resistant to long-term changes or are unable to tolerate stress should be relieved of excessive contact with the denture base. Selection of those regions that should provide primary and secondary support depends on the anatomic variations unique to each patient.

On the operon structure of the nitrogenase genes of Rhizobium leguminosarum and Azotobacter vinelandii.

The transcription of the nitrogenase genes in Rhizobium leguminosarum was studied by analysing total cellular RNA from bacteroids for the presence of nitrogenase messenger RNA. The RNA was separated by agarose gel electrophoresis and blotted onto nitrocellulose filters. Messenger RNA for nitrogenase was detected by hybridization with probes derived from plasmid pSA30, a recombinant plasmid carrying the nitrogenase genes of Klebsiella pneumoniae. In the same way nitrogenase mRNA was detected in RNA isolated from Azotobacter vinelandii and from Klebsiella pneumoniae, but only if both were cultured under conditions of derepression of the nitrogenase genes. The size of the RNA hybridizing with the probes indicates that in all three organisms the genes for the subunits of the two components of nitrogenase constitute a single operon.

Rotational path removable partial denture design.

A detailed description of a rotational path of insertion design for removable partial dentures has been presented. By minimizing the use of conventional clasps, this technique offers some advantages. Rotational path designs may minimize adverse periodontal response to a removable partial denture by reducing plaque accumulation and may be applied in esthetically demanding situations. The design concept involves the use of rigid retentive components that gain access to undercut areas through a rotational path of insertion. These rigid components satisfy the basic requirements of conventional direct retained design.

Expression of large plasmids in the endosymbiotic form of Rhizobium leguminosarum.

Isolated plasmid DNA from Rhizobium leguminosarum was hybridised with cellular RNA from broth-cultured bacteria and endosymbiotic bacteroids. From these hybridisation, experiments it is concluded that plasmid genes are strongly expressed in bacteroids and only weakly or not at all in bacteria. From the hybridisation of plasmid DNA with the cloned structural nif genes of Klebsiella pneumoniae it is shown that at least part of the nif genes are located on a plasmid.

Trigeminal neuralgia--a new treatment concept.

A concept for the treatment of trigeminal neuralgia is presented. On the basis of distinctive pain patterns, localized areas of pathosis within the jawbones are detected and obilterated. To date, we have treated eight patients with idiopathic trigeminal neuralgia. All patients have experienced total or near total abatement of pain.

On the isolation of TI-plasmid from Agrobacterium tumefaciens.

An efficient lysis method for Agrobacterium cells was developed, which allows a reproducible isolation of the tumor inducing (TI)-plasmid. The lysis method is based on the sensitivity of this bacterium to incubation with lysozyme, n-dodecylamine,EDTA, followed by Sarkosyl, after growth in the presence of carbenicillin. We also present a procedure for the isolation of the TI-plasmid on a large scale, that might be used for the mass isolation of other large plasmids which like the TI-plasmid, can not be cleared with earlier described procedures. The purity of the plasmid preparations was determined with DNA renaturation kinetics, which method has the advantage that the plasmid need not to be in the supercoiled or open circular form.
