Effects of coagulation temperature on measurements of complement function in serum samples from patients with systemic lupus erythematosus.

Blood samples from 15 patients with systemic lupus erythematosus (SLE) and 15 healthy blood donors were allowed to coagulate for one hour at room temperature, followed by one hour at 4 or 37 degrees C. The complement activity of the serum samples was assessed by three different functional assays. Serum samples from patients with SLE obtained by coagulation at 37 degrees C had a lower complement activity than serum samples from blood coagulated at 4 degrees C when the capacity of the serum samples to solubilise precipitable immune complexes and to support the attachment of complement factors to solid phase immune complexes was determined. Haemolytic complement activity was not affected by the coagulation temperature. The content of C1q binding immune complexes in paired serum samples obtained after coagulation at 4 and 37 degrees C was similar and the size distribution of the immune complexes, determined by high performance gel permeation chromatography, was also similar. This study shows that the results of functional complement assays, applied to serum samples from patients with SLE cannot be compared unless the conditions for blood coagulation and serum handling are defined and are the same. The data also indicate that assays measuring complement mediated solubilisation of immune complexes and the fixation of complement factors to solid phase immune complexes are more sensitive indicators of complement activity than the haemolytic assay.

A case of biotin-responsive 3-methylcrotonylglycin- and 3-hydroxyisovaleric aciduria.

During selective screening for organic acidurias, a 10-week-old girl with muscular hypotonia and recurrent fits was shown to be excreting 3-methylcrotonylglycin and 3-hydroxyisovaleric acid. Besides these metabolites of leucine the presence of small but pathological amounts of propionic and methylcitric acids were demonstrable in her urine, pointing to a defect in the metabolism of biotin. On treatment with biotin (2 x 5 mg/day) the convulsions stopped at once, her clinical condition improved gradually, and the abnormal metabolites disappeared from the urine. Within 6 weeks the child was discharged in a good general condition without apparent signs of neurological damage.

[Ultrahard longitudinal pendulum irradiation of tumors in the region of the visceral cranium (author's transl)].

An analysis of 71 patients with malignant, mostly extended tumors in the region of the visceral cranium is made, and the irradiation technique as well as the results of a longitudinal pendulum irradiation with high-energy ultrahard X-rays are reported. The advantages in comparison with rays of another quality and with other techniques of irradiation consist in a very small radiation exposure of the skin and in a homogeneous distribution of the dose in the target volume as well as a sparing of the contralateral eye. The adjustment is simple and easy to reproduce, the acute side-effects are small and reversible, the cosmetic result is very good.

[Results of female breast diagnostics (author's transl)].

A retrospective 2-years' study conducted in 3,563 patients showed that triple diagnostics involving clinic, mammography and puncture cytology occupies a central position in the diagnostics of the female breast, whereas plate thermography represents an additional method to clarify unclear clinical or mammographic findings. 88.9% of tumours were detected by means of a careful clinical examination. Over and above this, mammography enables identification of the occult carcinoma, which is extirpated in a controlled manner by means of preparation radiography. Puncture and cytology enable a nonsurgical clarification and treatment of cysts and are, therefore, an important control indicator of mastopathically changed mammae.

[Comparison between conventional X-ray diagnostics and computed tomography as techniques for irradiation planning in renal carcinoma and carcinoma of the bladder (author's transl)].

In order to obtain quantitative results by means of a comparison between irradiation planning with X-ray diagnostics and planning with cross-sectional computed tomography in abdominal tumors, therapeutic programs have been established twice for the same patients, each time using one of the two techniques. Fifteen patients with renal carcinoma and 24 patients with cancer of the urinary bladder have been examined.

[Radiotherapeutic aspects of testicular tumours: results and problems (author's transl)].

The five-year survival rate of patients with seminoma (61 cases) of all stages was 81% (seven-year survival rate 67%). Within this group the T1-2N0M0 stage had a five-year survival rate of 100%. Patients with tumours other than seminoma (52 cases) of all stages had a five-year survival rate of 54% (seven-year survival rate 47%). These results and those reported by others are the basis for not advocating routinely total radiation of the lymphatic pathways in the early tumour stages.

[The quantitative composition of natural and technologically changed aromas of plants. IV. Enzymic and thermal reaction products formed during the processing of tomatoes (author's transl)].

After the qualitative and quantitative investigation of the flavor compounds of tomatoes, the flavor compositions of tomato juices treated by different technological methods were determined. 80 aroma components were identified by gas chromatography and mass spectrometry. The quantitative comparisons showed that during the processing of juices unsaturated fatty acids acted as precursors for a series of C6-compounds, especially 1-hexanol, and cis-3-hexen-1-ol and that on the other hand some aroma compounds derived from carotenoid like structures, e.g. 6-methyl-5-hepten-2-one, geranyl- and farnesylacetone and beta-ionone were formed. As reaction products of thermal treatments among other increased amounts of acetoin, furfural, 5-methylfurfural and 6-methyl-5-hepten-2-one were established. The distribution of the aroma compounds dissolved in the serum or bound on the pulp was determined quantitatively by employing gaschromatographical methods.

[The quantitative composition of natural and technologically changed aromas of plants. II. Aroma compounds in oranges and their changes during juice processing (author's transl)].

After enzyme inhibition with methanol the original volatile compounds of the fruits were investigated in order to compare the quantitative composition of the aroma substances in orange fruits and juices. Aroma compounds of fresh, flash pasteurized and pasteurized juices were analysed. After aroma enrichment by liquid-liquid extraction, prefractionation of the flavor compounds by column chromatography on silicagel 56 flavor substances in the fruits and in the different juices were identified and determined by gas chromatography and mass spectrometry. In comparison to their content in the fruits only 10% of the terpenic hydrocarbons and about 30-66% of the esters were found on average in the fruits. Among the secondary flavor substances, which were not present or present only in traces in the fruits, but showed in juices concentrations up to 0,2 ppm, were 4-terpinenol, carveol and tr-2,8-menthadien-1-ol. After a thermic treatment also 3-hydroxy-2-butanone and 3-methyl-2-buten-1-ol were identified.

Acetoin

[Biosynthesis of aroma compounds by microorganisms II. Formation of sulphur containing flavour substances from methionine by Saccharomyces cerevisiae (author's transl)].

Using methionine as single nitrogen source 3-(methylthio)-1-propanol, 3-(methylthio)-propanal, 3-(methylthio)-propylacetate and 2-methylthiophan-3-one were formed in anaerobic model fermentations by Saccharomyces cerebisiae. The sulphur compounds were enriched by liquid-liquid extraction and detected by a selective flame photometric detector (FPD). The identities of the components were confirmed by mass spectrometry. It could be shown by quantitative determiniations that methionine was metabolized with 56--61% to 3-(methylthio)=1-propanol, when it was given to the medium in concentrations from 10 to 100 mg/1. 3-(methylthio)-propylacetuate, 3-(methylthio)-propanal, and 2-methylthiophan-3-one only could be detected in traces.

1-Propanol

[GLC-mass-spectrometrical investigation of the volatile components of wines vii. Aroma compounds of tokaj aszu wines b) organic acids (author's transl)].

An inventory of the organic acids detectable by liquid-liquid extraction has been made in different Tokaj wines. After the separation of the acidic compounds in the aroma extracts using Na2CO3 and after the derivatization with diazomethane 65 methylester have been separated by gaschromatography and identified by mass spectrometry. Using a special fractionation method organic acids, amino acids, carbohydrates and phenolic compounds have been isolated in different groups. After the derivatization of the components 50 organic acids have been quantitatively determined by gaschromatography. The compositions of the acids in the wines differ considerably. The quantitative differences are caused by the vintages, the method of production and the kind of storage. The possible biosynthetic pathways of the various acids are discussed.

[GLC-mass-spectrometrical investigation of the volatile components of wines. VI. Aroma compounds of Tokaj aszu wines. a) Neutral compounds (author's transl)].

The aroma compounds of Tokaj aszu-wines (1963, 5-butt. and 1973, 6-butt.) have been enriched by liquid-liquid extraction (pentane-methylenchloride 2 + 1) and compared with the aroma substances of a magyar Furmint wine (1973). After the separation of the acids and after the fractionation of the aroma extracts on silicagel 119 aroma components have been identified in the neutral fractions by means of the combination gaschromatography--mass spectrometry. Concerning the volatile constituents relatively high concentrations of furfural, 5-methyl-furfural, acetoin, different acetals, the ethyl esters of keto-, hydroxy- and dicarbonic acids and the lack of terpenic compounds are characteristic of the aszu wines. Quantitative differences depending on the vintages and the conditions of storage have been determined; particularly the concentrations of the acetales depend on the age of the wines.

After separation by column chromatography on silicagel and preparative gas chromatography 14 sesquiterpene hydrocarbons have been identified in aroma extracts from grapes by gas chromatography--mass spectrometry: alpha-copaene, beta-ylangene, beta-bourbonene, beta-caryophyllene, alpha-guaiene, alpha-humulene, alpha- and gamma-muurolene, germacren D, beta-selinene, alpha-farnesene, gamma- and delta-cadinene and calamenene.

[The biosynthesis of aroma compounds by microorganisms. I. Formation of N-acetyl-amines by saccharomyces cerevisiae].

Only primary amines were transformed to secondary acetamides by Saccharomyces cerevisiae in anaerobic model fermentations. Secondary amines and L-amino acids don't form N-acetyl-amines. Maximal rates of acetylation were obtained with 2- and 3-methyl-butylamine as acceptors of the postulated aminacetylase-reaction. The rate of acetylation of 2-phenylethylamine amounted to 12%; n-heptyl- and n-octylamine were transformed only in traces. At the example of 2-methyl-butylamine the kinetics of the reaction and the influence of the pH-value, of different N-sources and different concentrations of 2,4-dinitrophenol on the rate of acetylation have been examined. The identification and the determination of the microbiologically formed volatile compounds have been realised by means of the gaschromatography, concerning the qualitative experiments under mass-spectrometrical control.

Acetamides

[GLC-mass-spectrometrical investigation of volatile constituents of wine. IV. Identification of secondary amides in wines (author's transl)w].

Aroma extracts obtained by liquid-liquid extraction of wines from various sorts have been fractioned employing column-chromatography and using silicagel as adsorbent. In the polar methanol-elutions the following secondary amindes have been gaschromatographically separated and identified by means of a mass-spectrometer: N-ethyl-acetamide, N-isobutyl-acetamide, N-(2-methylbutyl)-acetamide, N-(3-methylbutyl)-acetamide, N-(3-[methylthio]-propyl)-acetamide, and N-(2-phenylethyl)-acetamide. Moreover the sulphur containing compound has been identified by means of an attached flame photometric detector. The different possibilities of the formation of the secondary amides during the fermentation process are discussed.
