Hyperuricaemia and perinatal outcome in pregnancy induced hypertension.

The effect of hyperuricaemia on perinatal outcome was evaluated in 40 primigravidae with pregnancy induced hypertension and 20 normotensive women in the 3rd trimester of pregnancy. Serum uric acid level was a better indicator than blood pressure as an index of foetal prognosis. Even severe hypertension without hyperuricaemia was associated with better prognosis for the foetus. Conversely when hypertension was mild and hyperuricaemia was severe, the prognosis for the foetus was poor. A rise in serum uric acid level > or = 5.5 mg% is associated with increased perinatal morbidity and mortality. Women in pregnancy induced hypertension group with serum uric acid level > or = 5.5 mg% had a higher incidence of intra-uterine growth retardation, low birthweight, and stillbirth making perinatal mortality rate of 200/1000 total births. The results of this study indicate that serum uric acid level > or = 5.5 mg% can reflect the perinatal outcome.

Alterations in rat intestinal sucrase and alkaline phosphatase activities in alloxan induced experimental diabetes.

Rat intestines revealed a significant loss of proteins after seven days of alloxan induced diabetes. The data suggested the presence of two forms of alkaline phosphatase (ALP) in normal rat intestines. Along with the loss of proteins from the intestines during diabetes, a form of ALP which appears to be loosely bound to the intestine is also flushed out. Total brush border membrane (BBM) proteins are relatively preserved from such leaching effect of alloxan induced diabetes. Thus, sucrase and another form of ALP which appears to be strongly bound to the BBM are flushed out at a slower rate as compared to the other intestinal proteins and loosely bound soluble ALP. BBM preparations from diabetic rat intestines showed lower ratios for BBM/intestinal homogenate sucrase or ALP activity/mg proteins as compared to the normal control rats. Such ratios, therefore, misdepict the purity as low for the BBM from diabetic rats which is merely because of the decreased contents of proteins in the intestinal homogenate during alloxan-induced acute experimental diabetes.

Lipid composition of intestinal brush border membrane in alloxan induced acute experimental diabetes.

Several alterations were observed in the rat intestinal brush border membrane (BBM) lipid composition after 7 days old alloxane-induced diabetes as compared to the control animals. There was no change in the total protein contents but a significant increase in the total lipid contents was observed. Glycolipids constituting the major lipid components showed a two-fold increase. No significant difference was observed in the total phospholipid contents. A significant decline in the free cholesterol (CH) level, free fatty acids, triglycerides and sialic acid contents was observed in membranes from diabetic rats. Esterified CH, monoglycerides+diglycerides, phosphatidyl serine+phosphatidyl inositol and phosphatidyl choline levels remained unaffected. A significant increase in sphingomyelin with a parallel decrease in phosphatidyl ethanolamine was observed in BBM preparations from diabetic rats. The observed changes in intestinal BBM might be responsible for altered functions of the diabetic intestines.

Production of antibodies to dextran using liposome as adjuvant.

The kinetics and nature of the immune response to dextran-BSA entrapped into liposomes was studied in rabbit. The antibody titer in the secondary immunization was found to be higher as compared to that in the primary response. In the primary response antibodies were of both IgG and IgM type but following secondary immunization the IgG level was increased. The conjugate entrapped into liposomes could establish an immunological memory for dextran. In order to eliminate the carrier effect of BSA, dextran was either entrapped or coupled to liposome and the immune response was studied in mice. The results revealed that dextran entrapped into liposomes or coupled to liposome surface induced IgM type immune response and antibody titer did not increase on secondary immunization.

Polyene antibiotics in the membrane environment.

The cytotoxic activity of the polyene antibiotics mainly depends on the appearance of the drug species which arises from drug-sterol complexation. The unsaturation and intact macrolide ring of the polyenes are the requirements for the biological activity. All the polyene antibiotics can form the complex with the sterol having 3 beta-OH group, and planar ring and a hydrophobic side chain. Aromatic polyene antibiotics with positively charged head group have been considered as most potential antifungal agents.

Recent outbreak of chloramphenicol resistant typhoid fever in West Bengal.

Forty patients admitted with typhoid fever from 1-1-1990 to 31-3-1990 were studied. The clinical features differed from the classical clinical profile. Thirty cases (75%) did not respond to chloramphenicol. Gentamicin, nitrofurantoin, nalidixic acid, kanamycin, cephalexin and norfloxacin were tried according to the sensitivity pattern. But response was variable. Ciprofloxacin was used in 18 cases which failed to respond to one of the above drugs. The result was quite satisfactory with minimal side-effects.

Serum potassium levels following suxamethonium administration in septic peritonitis patients.

Serum potassium and sodium changes following suxamethonium (1.5 mg/kg) administration were studied in 25 patients with septic peritonitis and 25 with no signs of peritonitis. A highly significant rise (P less than 0.001) in serum potassium (0.1-2.5 mEq/l) above the pre-induction levels was observed following suxamethonium administration in patients with septic peritonitis with maximum rise at 5 min following suxamethonium. This rise in potassium was significantly higher (P less than 0.001) in peritonitis patients at 3,5, and 10 min interval. A positive correlation was found between rise of potassium and duration of illness (P less than 0.01). No statistically significant changes were observed in serum sodium levels in both groups.

Synthesis of benzyl O-(2-O-methyl-beta-D-galactopyranosyl)-(1----3)-2- acetamido-2-deoxy-beta-D-glucopyranoside [benzyl 2'-O-methyllacto-N-bioside I], and its higher saccharide containing an O-(2-O-methyl-beta-D- galactopyranosyl)-(1----3)-2-acetamido-2-deoxy-beta-D-glucopyranosyl group as a potential substrate for (1---4)-alpha-L-fucosyltransferase.

Treatment of benzyl O-beta-D-galactopyranosyl-(1----3)-2-acetamido-2- deoxy-4,6-O-isopropylidene-beta-D-glucopyranoside with tert-butylchlorodiphenylsilane afforded the 6'-O-tert-butyldiphenylsilyl ether, which was converted into the 3',4'-O-isopropylidene derivative. Methylation and subsequent removal of protecting groups afforded benzyl O-(2-O-methyl-beta-D-galactopyranosyl)- (1----3)-2-acetamido-2-deoxy-beta-D-glucopyranoside (7). The trisaccharide methyl O-(2-O-methyl-beta-D-galactopyranosyl)-(1----3)-O-(2- acetamido-2-deoxy-beta-D-glucopyranosyl)-(1----3)-beta-D-galactopyranosi de (17) and the tetrasaccharide O-(2-O-methyl-beta-D-galactopyranosyl)-(1----3)-O-(2-acetamido-2-deoxy-b eta-D- glucopyranosyl)-(1----3)-O-beta-D-galactopyranosyl-(1----4)-D-glucopyran ose (32), both containing the 2'-O-methyllacto-N-biose I unit at the nonreducing end, were synthesized, and the structures of 7, 17, and 32 were confirmed by 13C-n.m.r. spectroscopy.
