Serum bile acids in man during vitamin C supplementation and restriction.

A daily dosage of 5 g ascorbate was given to 14 persons during one month. Various routine biochemical parameters were studied and the concentrations of individual bile acids in serum were determined. A significant increase in chenodeoxycholic acid concentration was found on interruption of vitamin C supplementation, whereas no other changes in bile acid concentrations were significant. One person, used to a large ascorbate intake (1 g/day), was deprived of ascorbate. On resuming the high ascorbate intake, serum bile acid concentrations showed an increasing trend. Urinary oxalate excretion and concentrations were impressively increased during vitamin C supplementation but no effects on kidney function were observed.

On the absorption of ascorbic acid in man.

The daily intake of ascorbic acid with the diet was strongly restricted in seven healthy non-smoking males and 90 to 180 mg of ascorbic acid were orally supplemented. After an equilibration period of three weeks about 5 muCi of (1-14C) ascorbic acid was orally administered with carrier (30 mg or 60 mg). The daily intake was then increased to 4 X 1000 mg and the cumulative excretion of radioactivity in the urine measured over a period of 10 days. The average absorption was estimated to amount to 80-90% of the dose.

Hepatic 7alpha-hydroxylation of cholesterol in ascorbate-deficient and ascorbate-supplemented guinea pigs.

Cholesterol 7alpha-hydroxylase activity was assayed in liver microsomes from guinea pigs supplemented with ascorbate and from guinea pigs in a state of ascorbate deficiency. A mass fragmentographic method was used by which the 7alpha-hydroxylation of endogenous cholesterol could be measured. The 7alpha-hydroxylation was markedly reduced in the ascorbate-deficient animals as compared to animals treated with ascorbate. Addition of ascorbate to the incubations did not increase this activity. 11- and 12-Hydroxylation of laurate as well as 25- and 26-hydroxylation of 5beta-cholestane-3alpha, 7alpha-diol were not significantly affected by the ascorbate status of animals. In the presence of excess NADPH-cytochrome P-450 reductase and a phospholipid, partially purified cytochrome P-450 from the microsomal fraction of liver of an ascorbate-deficient guinea pig had a much lower capacity to 7alpha-hydroxylate [4-14C]cholesterol than a corresponding system containing cytochrome P-450 from liver of an ascorbate-supplemented guinea pig. It is suggested that ascorbate affects the synthesis or breakdown of the 7alpha-hydroxylating system, in particular the cytochrome P-450 component.

Determination of phosphate in serum and urine by a single step malachite-green method.

The highly sensitive malachite-green method for the determination of phosphate requires a protective colloid in order to avoid precipitation of the formed dye salt. A polyvinylalcohol proved to be suitable and permit the determination of phosphate in urine and in serum without prior precipitation of the proteins. The present method is suitable for a one-step procedure which allows the analysis of several hundred samples per man-hour and is applicable to microquantities of serum or urine.

Fatty acid patterns of serum triglycerides and subcutaneous adipose tissues after ileal bypass in obesity.

A group of 9 obese subjects with a mean body weight of 140 kg was treated for their obesity with a jejuno-ileal intestinal shunt. During the first post-operative year their mean body weight decreased to 100 kg. Fatty acid patterns of triglycerides inserum and subcutaneous adipose tissue were studied at intervals during one year after surgery. Myristic and palmitic acids' contents of serum triglycerides were found to decrease during this period of time, whereas the fatty acid pattern of triglycerides in adipose tissue was practically unchanged. Triglycerides, cholesterol, albumin, and lactate dehydrogenase in serum decreased during the first year after operation, whereas aminotransferases showed a transient elevation. It is put forward that these findings are consistent with starvation rather than liver dysfunction.

Amino acid pattern in plasma before and after jejuno-ileal shunt operation for obesity.

The amino acid pattern in plasma was studied in a reference group (n=26) and in three groups of massive obese subjects (n=9, 8, and 9 respectively) before and at intervals after jejuno-ileostomy. The obese subjects had preoperatively an amino acid pattern significantly different from that in the reference group. The concentrations of lysine, tyrosine, 1/2 cystine, and glutamic acids were higher, and aspargin, glutamine, serine, and glycine were lower than in the reference group. During the post-operative period the amino acid pattern changed significantly; thus serine, glycine, and taurine increased and valine, lysine, leucine, tryptophan, thyrosine, 1/2 cystine, and citrulline decreased. The amino acid pattern in the obese group with the longest post-operative observation time and a stable body weight differed significantly from that in the reference group only with regard to a low valine concentration and high concentration of taurine and glutamatic acid.
