Ovine bone marrow extracellular matrix and soluble protein extraction: fetuin amino terminus microheterogeneity.

In this article, the authors describe a new method using phosphate-buffered saline for the initial extraction of extracellular matrix (ECM) and soluble proteins from hematologic tissues. Direct comparisons between this method and previously reported methods demonstrate superior total protein yields with the novel technique in a fraction of the time for these ovine hematologic tissues: bone marrow, marrow aspirate, spleen, liver, and blood. Analysis by one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis and silver staining demonstrate comparable protein patterns with this method and a previously reported method. This method also successfully extracts the ECM glycoproteins fibronectin and laminin as well as the proteoglycan, chondroitin sulfate from marrow. These findings are demonstrated by Western and dot blotting. Bone marrow ECM and soluble proteins extracted by the novel method stimulate myeloid progenitor growth in a methylcellulose assay. Using an assay for elastase inhibitory capacity, the authors demonstrate that alpha 1-proteinase inhibitor, the principal inhibitor of neutrophil elastase, is present and that its activity is preserved in bone marrow samples extracted with this team's method. In contrast, very low total protein yields are obtained with the method used previously to recover hemonectin, a rabbit bone marrow ECM granulocytic cytoadhesion molecule. This team's novel procedure, which extracts ECM and soluble proteins from small samples of tissue in a rapid, efficient, and reproducible manner, greatly enhances the analysis of these proteins from tissue culture, animal, and human clinical samples. In addition, with purification by chromatofocusing chromatography and molecular sieving gel electrophoresis, N-terminal amino acid sequencing could be performed on a developmentally regulated marrow protein with biochemical properties similar to those of hemonectin and the plasma protein fetuin. The authors propose that this novel technique be used for the initial extraction of ECM and soluble proteins from hematologic tissues and that subsequent, definitive recovery of insoluble proteins be accomplished using previously reported, though less efficient, methods.

The development of Lewis Henry Morgan's evolutionism.

Lewis Henry Morgan never used the term evolution in any of his major sociological works. If he may be labeled an evolutionist, the specificity of his views must be taken into account. The main "evolutionist" issue that concerned him was that of the unity or diversity of the human species. This was an urgent political and theological issue in the America of his day, and it impinged also on research on the American Indians. Morgan's first major study was designed to demonstrate the unity of origin of the American aborigines and their "Asian" origins. His methods were derived from the tradition of Indo-European philology. It was virtually as an afterthought that he added a social evolutionary component to what he conceived of as an exercise in philology. Later he came under the direct influence of the English evolutionists, and this was crucial for the conception of Ancient Society.

The structure of dream sequences.

The present study examines the relationships between dreams reported by a subject in the course of two nights in a sleep laboratory. The method used is derived from Lévi-Strauss's structural analysis of myth. The dreams are shown to constitute a set of systematic transformations of a single structure. The structure itself is formed by a set of oppositions, which in part represent a dilemma or conflict facing the dreamer. In the course of the night's dreams a series of transformations of the problem are generated and scanned. These transformations are governed by regular quasi-logical procedures.

The dream of Irma's injection: a structural analysis.

Lévi-Strauss developed a radically new, structural approach to the analysis of myth that may be adapted for use in the analysis of dreams. The method provides an alternative to psychoanalytic dream interpretation, focusing particularly on the internal dialectic of the dream--the movement from the initial premise to the resolution. This emphasis on the internal coherence and structured development of the dream contrasts strongly with the Freudian piecemeal decoding of dreams. The method is exemplified by analysis of Freud's classic Irma dream; it reveals a structure and message that have hitherto remained obscure. The interpretation connects the dream to Freud's theoretical ideas rather than to his supposed psychosexual fixations.
