Detection of sequence variation in PCR-amplified fragments of omp2 gene from three species of the family Chlamydiaceae using agarose gel electrophoresis containing bisbenzimide-PEG.

A simple technique providing a means for rapid genetic differentiation of chlamydial strains is described. The technique is based on a single-step sequence-specific separation of PCR-amplified DNA fragments by electrophoresis in an agarose gel containing a DNA ligand - bisbenzimide-PEG. A hypervariable region at the 5' end of the omp2 gene of Chlamydiaceae species encoding the 60-kDa cysteine-rich outer membrane protein was selected as a target for PCR. The appropriate fragments were amplified from strains of Chlamydia trachomatis, Chlamydophila pneumoniae, and Chlamydophila psittaci, and the PCR products originating from different species were electrophoretically separated in the presence of the DNA ligand. We therefore demonstrated that PCR with a single pair of primers followed by simple agarose gel electrophoresis with bisbenzimide-PEG can be applied to the differentiation of three members of the family Chlamydiaceae which are commonly recognized as human pathogens.

[The effect of the heart rhythm on the respiratory rhythm in anesthetized cats].

In 27 anesthetized cats, histograms of the ECG respiratory cycle phases distribution revealed that in 89% of cases the histograms were significantly non-uniform. The heart rate (HR) was relatively constant: high probability of respiratory phases switching was conditioned by the HR influences on respiratory rhythm. The intensity of such influence was about 10% on the average. The influence of cardiac rhythm on respiratory rhythm was caused by relatively weak signals. The closer the relation of HR to respiratory rate (f) to whole figures, the higher the probability of cardiac rhythm influence on respiratory phases switching.

[The effect of the heart rhythm on a shift in the respiratory phases in cats].

The probabilities of inspiratory on- and off-switches during the ECG's R-R interval were studied in 27 anesthetized cats. The tops of the high-probability peaks were located in the middle or in the second half of the R-R interval for the inspiratory off-switches. The effect of heart rate on the respiratory rhythm seems to be transmitted through the inspiratory off-switch mechanism. The high-probability peaks of inspiratory on-switches are subsequent to the inspiratory off-switch peaks.

[Features of the temporal structure of the respiratory cycle in the Tourette syndrome as a possible reflection of intensified dopaminergic processes].

The effect of dopamine blockers (chlorpromazine and haloperidol) and the dopaminomimetic piribedil on the structure of the breathing cycle was studied under conditions of the clinical employment of the drugs. It was found that piribedil increased the inspiration fraction (IF) in the respiratory cycle whereas neuroleptics diminished it. Twenty-six children with Tourette's syndrome (TS) and 24 with a temporal form of epilepsy were examined. Patients with TS showed two specific features of respiration regulation: a greater value of IF and a greater variability of respiratory parameters. Taking into account the pharmacological data, an increase in IF is considered as physiological evidence of dopaminergic hyperfunction in TS whereas a greater variability of respiratory parameters may reflect the paroxysmal activity of deep cerebral structures.

[Pharmacological analysis of the regulation of the phase duration of the respiratory cycle].

The oxotremorine, arecoline, physostigmine, armine, hyoscine and amphetamine affected the inspiratory (Ti) and expiratory (Te) duration as well as the Ti/(Ti + Te) ratio in 20 anesthetized cats. M-cholinopositive agents decreased the Ti/(Ti + Te) ratio due to shortening of inspiratory duration and simultaneous lengthening of expiratory one. Changes in the blood pressure and Ti/(Ti + Te) patterns appeared to be similar. The difference between our data and the data obtained by means of iontophoretic application of the agents to identified respiratory neurons might be accounted for by the fact that the routine technique used to identify respiratory neurons was unreliable.
