Human infection with Streptococcus zooepidemicus (Lancefield group C): three case reports.

Three unrelated severe infections with Streptococcus zooepidemicus occurred in England in 1985. The first patient developed septic arthritis, which has not been recorded before with this organism. The second died with septicaemia, pneumonia and post-streptococcal glomerulonephritis, the only record so far of nephritis following sporadic S. zooepidemicus infection and of nephritis and systemic sepsis in the same patient. The third patient experienced septicaemia during pregnancy but recovered without complications. A likely animal source of infection was found in only one case.

Quantitation of Salmonella O-antibodies in human sera by enzyme-linked immunosorbent assay (ELISA).

The enzyme-linked immunosorbent assay (ELISA) has been applied to the detection of antibodies against Salmonella O-antigens in human sera. Phenol-water extracted lipopolysaccharides (LPS) from serogroups A (O-antigens 2, 12), B (4, 5, 12) and D (9, 12) were used as antigens. When compared to the tube agglutination method according to Widal employing sera from patients with verified or suspected typhoid--or paratyphoid fever and from healthy controls it was found that ELISA (i) correlated significantly with the Widal reaction, (ii) was up to 100-fold more sensitive, and (iii) showed a greater reproducibility.

Effect of synthetic disaccharide-protein conjugate as an immunogen in Salmonella infection in mice.

The synthetic disaccharide 3-O-alpha-tyvelopyranosyl-alpha-D-mannopyranoside, identical to the O antigen 9 determinant in Salmonella, covalently linked to bovine serum albumin and suspended in Freund complete adjuvant, was used as an immunogen in mice. Antibodies in high titer were formed against the O9 determinant as estimated by a bactericidal assay, whereas only low titers of hemagglutinating antibodies were found. A sensitive in vivo assay demonstrated that in immunized mice, after challenge with two strains of equal virulence but with different O antigens (O9, 12 and O4, 5, 12, respectively), the growth of the strain with the O9 determinant was suppressed in comparison to that with the O4 determinant.
