Antioxidant defenses in a B16 melanoma line resistant to doxorubicin: an in vivo study.

A B16 melanoma line was repeatedly transplanted subcutaneously in C57BL/6 mice. On day 4 after every transplant, the animals were treated with doxorubicin (DXR), 10 mg/kg i.p. The aim of the work was to develop an in-vivo model of resistance to the antiblastic in order to analyze some possible mechanistic aspects of the process in the course of time. After 16 transplants and treatments the melanoma completely lost its sensitivity to the antiproliferative effects of maximal tolerated doses of DXR and showed over-expression of P-glycoprotein. Compared to the parental line, the in vitro resistance index was 4.6. After 27 transplants and treatments the melanoma did not increase its in vitro resistance to DXR further, and this resistance was completely reversed by verapamil. The behavior of the antioxidant defenses (superoxide dismutase, catalase, glutathione peroxidase, glutathione transferase, glutathione reductase and glutathione) was evaluated after 4, 16 and 27 transplants and treatments with DXR. At no stage did the treated melanoma show any variation in the antioxidant enzymes. Compared to the parental counterpart its glutathione levels were elevated after four treatments (+80%), when, however, the line was still sensitive to the in vivo effects of DXR, and after 16 treatments (+30%). Instead, no variation of the glutathione content was seen after 27 treatments with DXR. These results seem to exclude the possibility that the antioxidant defenses play a major role in the resistance of this B16 melanoma line to DXR. On the other hand, the low but, however, 'clinically' significant resistance of the tumor to the antiblastic seems mainly related to the mechanisms linked to the P-glycoprotein over-expression.

Xanthogranulomatous cholecystitis. Histopathological study and classification.

Xanthogranulomatous cholecystitis (XC) is a chronic inflammatory lesion of the gallbladder histologically characterized by the presence of varying amounts of foamy histiocytes in the inflammatory infiltrate. In this study a review of 63 cases selected from 1207 surgically removed gallbladder is presented; the percentage found (5.2%) is slightly higher than that of previous reports showing that XC is less uncommon than generally believed. A detailed microscopic study is performed: the authors observed according to the histological features particularly the different patterns of distribution of the inflammatory infiltrate and postulate the existence of three subtypes of XC: multinodular, focal and diffuse XC. Finally, the main etiopathogenetic hypotheses are briefly discussed.

[Correlations between morphometric characteristics and the presence of lymph node metastasis in stage T1 and T2 breast carcinoma].

In this study the relationship between cyto-morphometric characteristics of breast cancer and lymph node status was investigated. The study was conducted on 20 cases of T1-T2 breast cancer with node metastasis and 20 cases of T1-T2 breast cancer without node metastasis. About 100 nuclei for each patient were analyzed by means of a computerized image analyzer, and mean nuclear diameter, mean nuclear area, mean form factor and number of cells in 8 classes of diameter were studied. Mean mitotic index was also calculated. No statistic difference was found for all these variables, between the two groups. In conclusion, cyto-morphometric parameters although often represent an index of aggressiveness, they are not related to lymph node status. This one is, in fact, only an indicator of tumor's ability to spread, and tumor's diffusion can occur through different ways from the lymphatic one.

Pleomorphic adenoma and adenoid-cystic carcinoma of the salivary glands: comparative immunohistochemical patterns.

A series of 20 cases of pleomorphic adenoma and 19 cases of adenoid-cystic carcinoma of the salivary glands, and one case in the mammary location, were investigated regarding immunohistochemical reactivity for Tissue Polypeptide Antigen (TPA), Pre-Keratins, Vimentin, S-100 Protein, and their arrangement pattern of fibronectin. As a whole, the results support the hypothesis of morpho-structural and mainly, onto-histogenetic similarities between these tumours, but they also underline the need for great care in outlining their morpho-functional features, in relation to their different prognoses.

Correlation of an estrogen receptor-related phosphoprotein with histopathological features in breast cancer.

A series of 65 cases of different histological types of breast carcinoma was investigated for the immunohistochemical location of the estrogen receptor-related, 29 kD phosphoprotein using the ER-D5 monoclonal antibody. The ER-D5 response is heterogeneous in relation to some therapeutic limitations and is correlated with histopathological features of the tumors and survival. The main parameters for evaluation of breast cancers are reviewed, both those that are statistically correlated and those that are not apparently always correlated but are known to have considerable biological meaning, such as the ER-status of tumors.

Monocytoid B-cell lymphoma of the prostate.

A case of a primary malignant non-Hodgkin's lymphoma of the prostate, with the histological and immunohistochemical features of monocytoid B-cell lymphoma, is presented. The tumor histology was identical to that described in the forms of node-based monocytoid B-cell lymphoma being composed of a dense, monomorphous lymphoid infiltrate with ovoid nuclei and rather abundant, pale cytoplasm. Phenotypic analysis revealed high expression of B markers 4KB5 and L26, and negativity for T-associated antigens. This unusual localization broadens the spectrum of extra-nodal sites of monocytoid B-cell lymphoma.

Xanthogranulomatous salpingitis. Case report and review of the literature.

A xanthogranulomatous salpingitis, occurring in a fifty-two year old patient and appearing itself as a hydrosalpinx, is reported. The histological picture was characterized by the presence of several foamy histiocytes admixed with other inflammatory cells. Considerations about epidemiology, etiopathogenetic mechanisms and problems of differential diagnosis with other diseases are discussed.

[Role of prostate-specific antigen in prostatic carcinoma. Validity of an integrated methodologic approach and morpho-biological correlations].

A series of 72 cases of prostatic carcinoma, together with another of 20 cases of benign prostatic hyperplasia, serum PSA levels of which were available, are investigated, concerning their immunohistochemical responsiveness both to PSA and PAP. Particularly, parameters of 54 cases are statistically correlated, a significant correlation standing out between tumoral "grading" and PSA and PAP immunohistochemical patterns. Moreover, the two markers used show some useful complementary features. Concerning the serum PSA levels, they correlate more significantly with "staging" than with "grading" of the tumors. Then, the biological characterization of prostatic carcinomas, at least in the Authors' experience, should be improved any further if both the serum PSA levels and the PSA and/or PAP immunohistochemical patterns of tumors may be available together.

Computerized interactive morphometry as a potentially useful tool for the study of human placenta.

Two hundred human term placentas underwent morphometric study by means of semiautomated computerized method. Outer villous perimeter per cent parenchymal area and mean chord length were the investigated parameters. The mean value of outer villous perimeter, mean chord length and per cent area were respectively 46.9 mu (X 1000 mu 2) with a standard deviation of 4.6, 57.7 mu (standard deviation 9.3) and 66.1% (standard deviation 7.4). The relative distribution of these various parameters can constitute a valid basis for a comparison in cases of placental insufficiency.

Perineural infiltration by cutaneous squamous cell carcinomas of the head and neck.

In this paper six cases of cutaneous squamous cell carcinoma of the head and neck region with histologically-documented perineural tumoral infiltration are presented. Most patients showed recurrent disease during follow-up and have been treated with surgery and radiotherapy with or without systemic chemotherapy. The prognostic implications of perineural neoplastic infiltration as well as of therapeutic options are discussed.

Mental nerve invasion by basal cell carcinoma of the chin: a case report.

A case of perineural infiltration from recurrent cutaneous basal cell carcinoma of the chin is reported. Clinical presentation, surgical treatment, and pathological evidence are discussed. This report stresses the importance of a correct follow-up and an accurate clinical staging in order to identifying clinical signs highly suspicious for neoplastic perineural infiltration.
