The role of lysosomes in the pathogenesis of unicameral bone cysts.

Unicameral bone cyst fluid possesses N-acetyl-beta-D-glucosaminidase, beta-glucuronidase, PZ-peptidase, cathepsin D, acid phosphatase, N-acetyl-beta-D galactosaminidase, and beta-galactosidase activities. The activities of lysosomal enzymes in the cyst fluid are, as a rule, higher than in the serum, whereas the total protein content is lower. The content of collagen degradation products in the cyst fluid is higher compared to the serum. In bone cavity wall tissues, the collagen content is decreased. Adenosine 3':5'-cyclic phosphate and cyclic guanosine 3,5'-monophosphate accumulate in the cyst cavity. However, in some cases, there is no correlation among the activities of lysosomal enzymes in the cyst fluid, blood serum, and cyst wall tissues. The ratios of lysosomal enzyme activities in the cyst fluid differ from those in the cyst wall tissues, cultured skin fibroblasts, and blood polymorphonuclear leucocytes. The lack of coincidence of enzymatic spectra of the cyst fluid, wall tissues, and serum is suggestive of the diversity of ways of lysosomal enzyme enter the cyst cavity, i.e., blood, cyst fluid cells, and cyst cavity walls. The cysts with different locations (i.e., active and latent cysts) have similar lysosomal lytic potentials. The presence in the cyst cavity of extracellular lysosomal enzymes and collagen degradation products testifies to the permanent corrosion of the cyst cavity walls from the inside as well as to the increase in the osmotic pressure of the cyst fluid. Lysosome destruction should be regarded as an important pathogenetic factor that requires surgical or pharmacologic correction or both in the course of bone cyst management.

[The role of antioxidants in the action of thyroxine on myocardial functional properties in rats].

In conditions of excessive hormones in the blood, alpha-tocopherol increases the cardiomyocytes antioxidant defence, partially prevents the enhancement of the heart tissue sensitivity to induced peroxide oxidation of lipids, and considerably diminishes disorders in the heart contractile function. Under the same conditions, reduced glutathione no only does not exert its defensive action, but even decreases the heart resistance against hypoxia.

[Relation of the outcome of disk epiphysectomy in dysplastic scoliosis and the biochemical composition of the vertebral body].

Pronounced changes in the content of collagen, non-collagen proteins and the activity of a number of lysosomic enzymes have been established in the bone tissue of the vertebral bodies obtained on diskepiphysectomy in the patients with dysplastic scoliosis. Numerous correlation links between the metabolic parameters and the spinal deformation a year after diskepiphysectomy have been determined.

[The function of enzymes for the antioxidative protection of the bone marrow cells in rats during irradiation, bone fracture and combined radiation injury].

The activity of superoxide dismutase, glutathione peroxidase, glutathione reductase and glutathione: dehydroascorbate oxidoreductase is lower in the bone marrow than in the liver. The changes in the cellular antioxidative enzymatic system during leg bones fracture and X-ray irradiation are more pronounced in the rat bone marrow, as compared to the liver. The data obtained are in keeping with selective bone-marrow radiosensitivity and with the effect of exogenous superoxide dismutase and glutathione as radioprotectors.

[Thiol-dependent generation of superoxide radical induced by menadione and vicasol].

It was demonstrated that in the presence of blood serum menadione and vicasol reduce cytochrome c in two ways, i. e., via O2-. -dependent and O2-. -independent routes. T ability of the serum to generate O2-. in the presence of menadione is retained after dialysis. Albumin can also act as an electron donor during O2-. generation mediated by menadione; however, this ability of the serum does not correlate with the presence of albumin in it. N-Ethylmaleimide and Cu2+ inhibit the ability of albumin and blood serum to induce menadione-mediated generation of O2-. It is concluded that in the presence of thiol-containing proteins and O2-. vitamin K3 forms an oxidative system generating O2-.

[Effect of glutathione on choleragenic diarrhea and disorders of the antioxidant system of rat intestinal and liver cells].

The effects of reduced glutathione on the development of choleragenic diarrhea and the activity of glutathione transferase (GT), glutathione peroxidase (GP-GTB and GP-H2O2), superoxide dismutase (SOD), glutathione reductase (GR) in the small intestine and liver of rats with experimentally ligated jejunal loop have been studied. Diarrhea syndrome was found to decrease markedly after glutathione administration in a dose of 1 g/kg bw. GR activity in the jejunum and liver of rats treated with toxin and the following glutathione administration rose by 210 and 186%, respectively, and then reached the control level. Glutathione transferase activity in the jejunum increased by 150% (P less than 0.05), remaining, however, lower than the control values. The activity of other enzymes tested was unchanged. Polyfunctional cellular activity of glutathione suggests that antidiarrhea effect should be considered as an element of pathogenetic therapy.

[Inhibition of superoxide-dependent processes by aminoglycoside antibiotics].

The ability of various antibiotics to inhibit superoxide anion(O-2)-mediated formation of adrenochrome from adrenaline and recovery of cytochrome c by xanthine oxidase was studied. In the adrenaline system (pH 10.2), aminoglycosides might be arranged, according to the inhibitory effect, in the following order: monomycin greater than gentamicin greater than kanamycin greater than lincomycin greater than streptomycin. In the xanthine oxidase system (pH 7.8), that order was the following: monomycin greater than gentamicin greater than lincomycin greater than greater than kanamycin. It was suggested that the antibiotic inhibition of the O-2-dependent processes at the essential sites of metabolism and/or the antibiotic involvement into the process of free radical oxidation initiated by O-2 in the cells might be one of the mechanisms of the drug action and toxicity with respect to the host.

[Superoxide dismutase inhibition as a prerequisite for disordered myocardial function under oxygen loading].

Changes in the contractility of rabbit myocardium following administration of diethyl dithiocarbamate were studied to determine the role of superoxide dismutase (SOD) in cardiac support function. It was observed that in healthy rabbits, a 50% decrease in the left ventricle SOD level induced by the inhibitor was not followed by any considerable disturbances in myocardial contractility as determined without additional stimulation and load. HBO sessions caused appreciable disorders in heart contractility which could be partly prevented by SOD administration. In rabbits with adrenaline-induced heart lesions, depression of myocardial contractility induced by the inhibitor alone or in combination with intensive oxygenation was also observed.

[Gentamycin inhibition of adrenaline auto-oxidation].

The effect of gentamicin sulfate on the rate of free radical oxidation of adrenalin was studied. Inhibition of the oxidation process at the final concentrations of gentamicin of 0.1-0.2 mg/ml was observed. The mechanism of gentamicin antiradical activity and its role in the cells are discussed.

[Effect of combined use of hyperbaric oxygenation and alpha-tocopherol on the contractile function and various components of the antioxidant system of the heart after adrenaline lesioning].

A session of hyperbaric oxygenation (HBO) at 2.5 at. abs. conducted for one hour in rabbits with adrenalin heart affection (AHA) immediately after adrenalin administration prevented a decrease in the cardiac contractile function usually developing within two hours after AHA. On the third day of the follow up, rabbits with AHA receiving daily HBO displayed a decrease in the pump and contractile functions as well as in superoxide dismutase (SOD) activity and the antioxidant activity (AOA) of the left ventricle lipids. Combination of HBO sessions with the intra-muscular administration of alpha-tocopherol (50 mg/kg) made the decrease in SOD activity less pronounced and prevented both the fall in lipid AOA and the depression of the heart contractile and pump functions on the third day following AHA. It is suggested that the combined use of HBO and substances increasing the potency of the cell antioxidation system may become one of the principles of reducing HBO side effects and of expanding the potentialities of HBO in treating ischemic and adrenergic heart impairments.

[Effect of a combined radiation lesion on the enzyme activity of the glutathione redox system of the rat liver].

A study was made of activity of glutathione peroxidase, glutathione reductase and glutathione-dehydroascorbate oxidoreductase in the rat liver under x-ray treatment in doses of 206 or 155 mC/kg coupled with mechanical injury (closed fracture of the leg bones). Radiation in a dose of 206 mC/kg gave rise to the following changes in activity of glutathione redox-system enzymes: reduced activity of glutathione peroxidase on the 3d day, and increased activity of glutathione reductase, and glutathione-dehydroascorbate oxidoreductase on the 7th day after combined radiation injury. Radiation in a dose of 155 mC/kg was accompanied by increased activity of glutathione-dehydroascorbate oxidoreductase on the 7th day and reduced glutathione reductase activity on the 30th day after the injury infliction.
