[Anthrax toxin dynamics in the body].

Anthrax toxin introduced subcutaneously into white rats Fisher-344 caused specific intoxication accompanied by characteristic pathomorphological changes and, when studied by the immunoperoxidase method in histological sections, could be detected in the blood stream (in plasma) and in the cytoplasm of macrophages (in the lungs and the spleen). The content of the preparation introduced into the animals in the blood stream is directly related to its toxicity, being probably indicative of the relative functional failure of the macrophagal system.

[Pathogenic properties of freshly isolated strains of the pertussis microbe].

The serovar composition, toxicity, virulence and lymphocytosis stimulating activity (LSA) of B. pertussis strains circulating in the 1980-ies were studied in comparison with the strains circulating in previous years. The study revealed changes in the toxic properties of B. pertussis: their decrease in the years of the intensive immunization of children against whooping cough and rise at the period when the number of immunized children was reduced. The toxic properties and LSA in most B. pertussis strains were less pronounced in the 1980-ies than in the 1960-ies. The serovar composition of the circulating strains remained stable for 15 years with the prevalence of serovar 1.0.3.

[Effect of anthrax toxin on the chemiluminescence of human leukocytes].

The effect of substrate containing crude anthrax toxin on the phagocytosing leukocyte chemiluminescence has been studied. Preliminary toxin incubation with leukocytes for 60 min blocks cell chemiluminescence in the linear ratio effect concentration in the protein component of the toxin; the minimum concentration of the toxin protein component inhibiting the phagocytosing leukocyte luminescence is 3-5 micrograms per 5 x 10(5) cells. The substrate pure mixture of the oedema factor and protective antigen inhibits the chemiluminescence more intensively than crude toxin does. Inhibiting chemiluminescence activity of the anthrax toxin is directly proportional to its adenylate cyclase activity.

[Experimental characteristics of anthrax intoxication].

In experimental anthrax intoxication, a highly important stage of its pathogenesis consists in microcirculatory disturbances with the phenomena of blood sludge, accompanied by the increased permeability of blood vessels not only for plasma, but also for red blood cells. These disturbances result in perivascular hemorrhages, hemorrhagic infiltrations, edema and cavitary transudates. Pulmonary edema and, as a consequence, the accumulation of fluid in pulmonary alveoli and the respiratory tract are of particular importance and, probably, can be considered the basic cause of the ensuing acute and fatal asphyxia. Such vascular and pulmonary insufficiency is accompanied by a decrease in the content of macroergic compounds (and in particular ATP), the characteristic deformation of red blood cells and disturbances in their oxygen transport function, which is linked with the decreased content of 2,3-diphosphoglycerate.

[The use of gel chromatography for the isolation of biologically active toxin complex from Bacillus anthrax].

The multistep fractioning of the protein components from cultural filtrate of B. anthracis grown on casaminoacids containing medium was done. The scheme for preliminary purification of a toxin complex of B. anthracis against low and high molecular mass contaminants has been elaborated and includes ultrafiltration, gel chromatography in ultragel AcA-202 and TSK-gel toyopal HW-55. Biological activities of the complex,toxicity for laboratory animals and adenylate cyclase activity characteristic of B. anthracis toxin, are shown to remain intact in course of preliminary purification. Molecular masses of protein subunits from the fraction containing anthracis toxin activity reach 85-90 kD, 0.3-0.5 mg of toxin complex protein is yielded from 1 l of B. anthracis cultural filtrate.

[Polyacrylamide gel disc electrophoretic study of laboratory and freshly isolated strains of Bordetella pertussis and Bordetella parapertussis].

Differences in the antigenic complexes of laboratory and newly isolated B. pertussis and B. parapertussis strains have established by means of disc electrophoresis in polyacrylamide gel. B. pertussis strains have been found to contain 2 or 3 antigens more. Laboratory and newly isolated B. pertussis strains have been found to differ in antigenic complexes, the antigens in these complexes having different electrophoretic mobility. The antigens detected in newly isolated strains have greater electrophoretic mobility in comparison with that of the antigens detected in laboratory strains. No such differences have been revealed in laboratory and newly isolated B. parapertussis strains. Disc electrophoresis in polyacrylamide gel allows to detect 2 or 3 more antigens than immunoelectrophoresis in agar gel.

[Intensity of the epidemic process of whooping cough under conditions of mass immunization over a period of many years].

On the basis of analysis of whooping cough incidence in 1959 to 1975 in Moscow the authors present characteristics of the epidemic process under conditions of planned many-year immunization of children against this infection. Whooping cough morbidity proved to undergo significant changes-from sharp falls the first 8 years of immunization to a relative stabilization with a gradual reduction the last 5 years. Marked seasonal and periodic elevations persist against the background of reduction of morbidity. Preschool- and schoolchildren are equally involved in the epidemic process; the most frequently involved are nonvaccinated children aged 1 year and 4 to 10 years, which lost postvaccinal immunity. Further reduction of the severity of whooping cough and increase in the number of unimanifested forms of the infection was noted. This was apparently connected not only with immunization, but also with the reduction of the virulence of H. pertusis detected in studying the cultures isolated in Moscow from 1967 to 1974. The intensity indices of the epidemic process in whooping cough pointed to the necessity of using a more effective vaccine for the protection of children from whooping cough, despite the noted reduction of morbidity, and diminished severity of the course of the disease, and of the causative agent virulence.
