Use of lattice square designs in bioassays.

The use of Lattice Square designs for estimating the relative potencies in a multiple bioassay is illustrated in this paper. The analysis is presented with and without recovery of inter-row and column information.

Analysis of multivariate parallel-line bioassay with composite responses and composite doses, using canonical correlations.

The use of a composite response to estimate the relative potency in a multivariate parallel-line bioassay is described by Finney. Occasionally a multivariate bioassay has multiple drugs also. Such a situation arises, for example, when a treatment contains several drugs. A procedure for combining multiple responses and combining multiple drugs is proposed in this paper, in order to obtain point and interval estimates of the relative potency. A composite response and a composite dose level for this purpose are obtained by using canonical analysis.

Moderate protection of renal function and reduction of fibrosis by colchicine in a model of anti-GBM disease in the rabbit.

A rabbit model of renal glomerulosclerosis induced by anti-glomerular basement membrane antibody was used to determine whether colchicine would protect renal function and reduce fibrosis. Initial studies established the time course of renal function changes and fibrosis. Colchicine at a dose of 0.02 to 0.04 mg/kg per day injected ip was begun at day 4 when injury had been initiated, and the experiment was ended at day 21 when fibrotic changes were established. Colchicine significantly reduced the rise in serum creatinine (serum creatinine = 2.7 +/- 0.3 mg% in vehicle-treated animals versus 1.8 +/- 0.1 mg% in colchicine-treated animals) and interstitial fibrosis (fibrosis score = 2.6 +/- 0.2 in vehicle-treated versus 1.5 +/- 0.2 in colchicine-treated animals). Colchicine treatment did not significantly affect weight, anti-guinea pig immunoglobulin level, % fibrocellular crescents formed, hydroxyproline per gram (dry weight) in tissue, or urine protein: creatine ratio. Regression analysis was performed to examine the interrelationships between variables for all animals and the effect of colchicine on pairs of variables. No clear-cut site of colchicine action could be identified. These data show that colchicine, in doses that could be used in humans, protected renal function by about 25% and reduced interstitial fibrosis in a model of severe crescentic nephritis.

A semi-Markovian model for predator-prey interactions.

In the study of population dynamics, the predator-prey system is recognized as a vitally important aspect in natural population control. On aspect of predator-prey interactions is studied in this paper. The attack cycle of a predator is assumed to consist of four different activities, namely, search, pursuit, handle and eat, and digestion. A semi-Markovian model is proposed to obtain the number of prey devoured by a predator during the activity of a day. The advantages of a new semi-Markovian model to the queueing model developed by Curry and DeMichele (1977) is demonstrated, as the results from the semi-Markovian model are closer to Holling's (1966) experimental data than their results, which were based entirely on Poisson assumptions.
