Inhibition of neutrophil superoxide secretion by the preservative, methylhydroxybenzoate: effects mediated by perturbation of intracellular Ca2+?

The preservative, methylhydroxybenzoate inhibited O2- secretion from human neutrophils activated by both the chemotactic peptide fMet-Leu-Phe and phorbol myristate acetate (PMA): the low level of oxidant secretion activated by the ionophore A23187 was similarly reduced in preservative-treated suspensions. Oxidant secretion was similarly reduced in fMet-Leu-Phe and A23187 treated suspensions in which intracellular Ca2+ was buffered by loading with Quin-2, indicating that methylhydroxybenzoate may exert its effects by perturbation of intracellular Ca2(+)-dependent processes. Methylhydroxybenzoate could mimic EGTA in preventing the Ca2+ dependent enhancement of trypsin activity and could also bind this cation in experiments using a Ca2+ electrode, although the preservative bound Ca2+ more slowly and had a lower affinity than EGTA. These data indicate that methylhydroxybenzoate may exert its effects on neutrophils by perturbation of Ca2(+)-dependent activation pathways and this phenomenon may also explain its other known pharmacological effects. Furthermore, these observations provide an insight into the mechanisms by which intracellular Ca2+ may regulate oxidant secretion.

Preservatives in the vehicle of naloxone: pharmacological effects.

The pharmacological activity of the preservatives methyl and propyl hydroxybenzoate, until recently components of the vehicle of naloxone (Narcan), was investigated in vitro. This vehicle produced reversible, concentration-dependent relaxation of guinea pig trachea, not mediated via adrenergic or cholinergic receptors, prostanoid activity or phosphodiesterase inhibition. Sensitivity of the tissue to calcium-induced contraction was decreased. In single isolated rat hepatocytes, surface receptor stimulation elicits repetitive transient rises in intracellular free calcium measured with the photoprotein aequorin. The vehicle reversibly inhibited these transients. These observations suggest that the effect of hydroxybenzoates may be mediated via a perturbation of intracellular calcium-related processes.

Does naloxone induce relaxation of guinea-pig airway smooth muscle?

The effects of the opiate receptor antagonist naloxone were investigated on isolated preparations of guinea-pig trachealis contracted with either histamine, methacholine or KCl. The commercially available solution of naloxone (Narcan) induced concentration-dependent relaxation of the contracted airway preparations. In stark contrast, aqueous solutions of naloxone were without any significant relaxant effect. Aqueous solutions of the preservatives (methyl and propyl hydroxybenzoate) present in the vehicle used in the commercial formulation of naloxone mimicked exactly the relaxant effects induced by Narcan. Thus, naloxone does not directly induce relaxation of airway smooth muscle. The effects of Narcan can be solely attributed to the activity of the preservatives present in the vehicles. The mechanism underlying the bronchodilator activity of methyl and propyl hydroxybenzoate is unknown but is not related to receptor blockade or to alterations in the intracellular levels of cyclic AMP.
