A marsupial oncovirus?

A virus-like particle was observed in two continuous cell lines derived from the marsupial Sminthopsis crassicaudata (Fat-tailed Dunnart). The development of the particle was similar to the development of D-type oncoviruses. Initially, a crescentof nucleoid material was observed near the nucleus in the region of the Golgi apparatus. This crescent developed into a doughnut-shaped-A-type particle which migrated through the cytoplasm towards the cell membrane where it budded either into a smooth membrane cytoplasmic vacuole or from the cell membrane. Only enveloped A-type particles were observed; no mature B-type, C-type or D-type particles were detected.

Marsupial cells in long-term culture.

Cell lines have been developed from several species of Australian marsupials and studied during long-term growth. Cell lines developed from macropodid skin or heart tissues all had reproducible finite life-spans. However, cell lines developed from dasyurids showed bariable behavior in culture: lines developed from Antechinus stuartii and Dasyurus viverrinus had finite life-spans, while lines developed from Sminthopsis crassicaudata had indefinite life-spans. S. crassicaudata lines usually became heteroplloid, but one was still diploid after 150 population doublings, while another contained a proportion (10%) of haploid cells. Other lines were developed from the peramelid, Perameles nasuta, and the phanlngerid, Trichosurus vulpecula.

Cholesterol metabolism in Papua New Guineans.

Cholesterol metabolism has been studied in a group of healthy males drawn from a New Guinea highland population whose plasma cholesterol concentration is low by Western standards and does not rise with age. Mean values from compartmental studies in six subjects were: pool A mass 289 mg/kg; production rate 1069 mg/day or 17.4 mg kg-1 day-1. The average rate of endogenous synthesis measured by sterol balance in twelve men was 751 mg/day or 12.4 mg kg-1 day-1. There was evidence of probable suppression of synthesis by about 500 mg/day when 1 g/day of egg yolk cholesterol was added to the diet of six subjects for 5 weeks. In the latter group, mean cholesterol absorption was 39% (390 mg/day) but the plasma cholesterol concentration did not rise, possibly reflecting compensatory redution in synthesis. It is suggested that these results were unexceptional, except possibly for the failure of the plasma cholesterol concentration to rise when cholesterol was ingested, despite gross differences in diet and many other factors.

Rapid detection and isolation of mycoplasmas from cell cultures.

An improved medium for the isolation of Mycoplasma species from contaminated cell cultures is reported. A modification of the mycoplasma detection method using the uridine/uracil uptake ratio method is described. Results obtained using this method with mycoplasma contaminated cell cultures and with contaminated cell cultures treated with the antibiotic Lincomycin are presented.

Priscoline and epinephrine in selective pancreatic angiography: a comparison study using high pressure injection, Valsalva maneuver and geometric magnification.

The value of selective, high injection rate, pancreatic pharmacoangiography was assessed in 114 patients. Opacification of the pancreas was graded on a 0-4+ scale. Selective injection of gastroduodenal and small pancreatic arteries gave the best results, 3.7+. Celiac injection rating without drugs was 0.9+, with epinephrine 2.2+, and with Priscoline 2.7+. Average ratings of hepatic artery injections were 2.6+ with epinephrine and 3.0+ with Priscoline; splenic artery injections were 2.5+ and 3.5+, respectively. Although superselective injection was possible in only 50% of the cases, addition of epinephrine and Priscoline resulted in an average grading of 3.4+ (yielding a preoperative diagnostic accuracy of 94%).

Analysis of control methods: mercury and cadmium pollution.

Physical system conceptual models are developed to illustrate the various interconnecting pathways of metal flow. Economic use of mercury and cadmium, as representative toxic heavy metals, is inventoried, and the losses of each from the pathways of economic use are compared. Distinctions are made between high volume consumers and industries that are responsible for a large percent of total emissions or effluent loads. The pathways of the metals in the environment are traced via the conceptual models. A global mass balance is presented for mercury. The problem of high local concentrations vs. global metal flow is reviewed. Impacts of the metals on human health are categorized by type of effect. Available control strategies and abatement measures are evaluated with respect to the effectiveness of each on the problems of metal pollution, as illustrated by the physical models.

Duplex scanning of the thyroid.

High resolution ultrasound scanners are used to image nodular thyroid glands and in the study of thyroid malignancy. There are no specific ultrasonic characteristics of malignant lesions to distinguish them from benign conditions. Preliminary results using a Duplex ultrasound scanner shows that the detection of a high frequency Doppler-shift signal from the superior thyroid artery is indicative of an overactive thyroid. In a study of 23 consecutive patients who were being investigated for possible hyperthyroidism, 15 were found to have normal thyroid function and 8 to be overactive. The normals had a mean systolic peak frequency of 1.77 +/- 1.75 kHz, and the hyperthyroid group 4.08 +/- 1.5 kHz. This result is highly significant, having a p value less than 0.0002 (Mann-Whitney U Test). In another 10 patients with solid lesions, one was diagnosed as follicular carcinoma. High frequency Doppler shift signals, similar to those found in carcinoma of the breast, were detected in the periphery of the lesion but were not found in nine cases of adenoma. Duplex scanning may assist in the investigation of hyperthyroidism and in the differential diagnosis of benign and malignant lesions.
