Lipolytic activity of porcine pancreas lipase on fatty acid esters of dialkylglycerols: a structural basis for the design of new substrates for the assay of pancreatic lipases activity.

For the design of new synthetic substrates for the assay of pancreatic lipases activity, acyl dialkylglycerols of variable chain length were prepared. Titrimetric assay of these substrates showed the highest lipolytic activity of porcine pancreas lipase (pPL) with butanoyl dibutylglycerol. The activity is lower but comparable to that shown by pPL towards the classical substrate tributyrin. The 4-nitrophenylcarbonate of 1,2-di-O-butylglycerol, has been prepared and proposed as synthetic substrate for a new spectrophotometric assay of pancreatic lipases.

Substrate interaction with 5alpha-reductase enzyme: influence of the 17beta-chain chirality in the mechanism of action of 4-azasteroid inhibitors.

A series of steroidal compounds were synthesized in order to evaluate the possible influence of the configuration of a stereocenter in the 17beta-side chain on the inhibitory activity on the enzyme 5alpha-reductase (5AR). For this purpose diastereomerically pure 4-azasteroids epimers at C-22 were prepared (compounds 1-11) and tested as inhibitors of 5AR in "in vitro" tests. The obtained data showed that in most cases the couples of epimers possess a significant difference in their biological activity. We also considered, for the tested molecules, a series of chemico-physical parameters in order to find a possible correlation with their biological activity. The findings allowed us to propose a model of the binding site of 5AR which comprises also, for 4-azasteroid inhibitors, the configurational aspect of the 17beta-side chain.

A practical route for the synthesis of 17 substituted steroidal 3-thioxamides.

A facile method for the synthesis of a series of new steroidal 3-thioxamides from the 3-oxo compound, variously substituted at the 17 position, is described. The "one pot" reaction, using Lawesson's reagent (4-methoxyphenylthionophosphine sulphide dimer) in dichloromethane solution, gives the desired compounds with a high degree of chemoselectivity, in good yields (> 80%).

Open-chain peptides obtained by acidic hydrolytic cleavage of cyclosporin A.

Hydrolysis of cyclosporin A (CsA) was studied in order to clarify the still undefined point of attack of the acidic degradation. Among ether extractable and water-soluble products formed from CsA in HCl, two open-chain peptides were isolated by high-performance liquid chromatography which were identified as the deca- and nonapeptides deriving from CsA through the hydrolytic cleavage of amino acid residue 11 and both residues 11 and 10, respectively. Identification was carried out by fast atom bombardment tandem mass spectrometry.

Studies on analogues of succinic semialdehyde as substrates for succinate semialdehyde dehydrogenase from rat brain.

The methyl ester of succinic semialdehyde (SSA) was examined as a substrate for succinate semialdehyde dehydrogenase (SSADH) from rat brain. It was found that the ester can be oxidized by the enzyme. Values of Km for SSA-Me were higher than for those for SSA, and for this substrate the enzyme showed a substrate-dependent inhibition. This finding suggests that the carboxylate group of SSA is not essential in the process of inhibition of SSADH by the substrate. Cyclopropyl analogues of SSA, cis- and trans-1-formyl-cyclopropan-2-carboxylic acids, were also individually tested as substrates of SSADH. Only the trans isomer was found to be oxidized to the corresponding dicarboxylic acid; it inhibited the enzyme in the same range of concentrations as SSA. The above data suggest that, as for gamma-aminobutyric acid, SSA is present in an unfolded, transoid conformation at the active site of SSADH.

Inversion of the unnatural cis C/D sterol ring junction of 5alpha, 14beta-cholest-7en-3beta-ol by rat-liver enzymes.

Labelled 5alpha, 14beta-cholest-7-en-3beta-ol with the 'unnatural' cis C/D ring junction was synthesized. Incubations of this sterol with rat liver homogenate under aerobic conditions gave radioactive 5alpha-cholest-7-en-3beta-ol, 5alpha-cholest-8(14)-en-3beta-ol andcholesterol indicating the presence of enzymes in the rat liver capable of inverting the C/D cis to a trans configuration. No radioactivity was found associated with added 14beta-cholesterol showing the specificity of the 14alpha configuration for the enzymic conversion of a sterol molecule into cholesterol.
