Isolation of self-recognizing IgG2a monoclonal rheumatoid factors.

Hybridoma cell lines were produced by fusing spleen cells from 11 mice that expressed the lpr mutation to nonsecreting SP2/0 myeloma cells. Five to 20% of the hybridoma cell lines so derived were found to secrete antibodies that recognized autologous IgG2a. Out of 168 anti-IgG2a clones tested, 4 clones produced anti-IgG2a rheumatoid factor (RF) that was itself IgG2a. Two clones recognized self IgG2a while the other two clones each recognized a nonself IgG2a allotypic determinant. These findings indicate that self-recognizing antibodies are a component, albeit a minor one, of the RF repertoire of lpr/lpr mice.

Frequency and epitope recognition of anti-ribosome P antibodies from humans with systemic lupus erythematosus and MRL/lpr mice are similar.

Autoantibodies reactive against a shared, conserved epitope on the ribosomal phosphoproteins P0, P1, and P2 occur in approximately 15% of patients with SLE and are relatively specific for this disease. To determine whether anti-P antibodies occur in murine lupus, serum from MRL/lpr and NZB/W F1 mice were analyzed by immunoblotting as well as by ELISA using a synthetic peptide Ag. Of those analyzed, 4 of 35 (11%) MRL/lpr, 0 of 25 NZB/W F1 and 0 of 13 control NIH/Swiss mice had anti-P antibodies. Anti-P specificity was confirmed by immunoblotting of ribosomal proteins separated by two-dimensional gel electrophoresis and by inhibition of anti-P reactivity on immunoblots with the synthetic peptide Ag. These findings indicate a striking similarity in the frequency and fine epitope specificity of anti-P antibodies in humans and MRL/lpr mice with SLE.

Oligoclonality of rheumatoid factors arising spontaneously in lpr/lpr mice.

Rheumatoid factors (RF) from individual mice of the MRL/lpr and C3H/lpr strains were examined with regard to target specificity and heavy chain class expression. It was found that MRL/lpr-derived RF preferentially recognize IgG2a target antibodies while C3H/lpr-derived RF exhibit a broader range of cross-reactivity. IgM RF represent only a minor proportion of the RF response in both strains. The IgG2a-specific RF produced by the lpr strains were found to consist of relatively equal proportions of IgG1, IgG2b, IgG3, and IgA antibodies. However, most of the RF produced by a single individual expressed the same heavy chain class and antigen-binding specificity. These results indicate that within an individual lpr mouse, circulating RF are derived from extensive expansion of a limited number of antigen-activated precursors.

Selective elimination of non-lpr lymphoid cells in mice undergoing lpr-mediated graft-vs-host disease.

The transfer of lpr BM stem cells into lethally irradiated non-lpr recipients (including the congenic MRL/+ differing only at the lpr locus) causes GVHD characterized by a wasting syndrome. In this study we investigated the interaction between the autoimmune (lpr) and normal (A-Thy) B, T, and RBC cell lineages in two types of radiation chimeras: MRL/lpr plus A-Thy----(MRL/lpr X A-Thy)F1 and MRL/+ plus A-Thy----(MRL/lpr X A-Thy)F1. Analysis of B cell repopulation by competitive RIA of serum Igh-1 allotype showed that both the MRL and the A-Thy donor cells initially engrafted. However, by 2 to 4 mo post-transplantation the normal A-Thy allotype was barely detectable (reduced greater than 2 orders of magnitude), whereas the autoimmune MRL/lpr allotype persisted at normal levels. Similarly, investigation of the donor origin of peripheral blood T cells by two-color flow cytometry showed that by 8 mo post-transplantation normal A-Thy T cells had been eliminated and only MRL/lpr T cells were present in the circulation. In contrast, erythrocytes from both the MRL/lpr and A-Thy donor strains successfully engrafted the F1 recipients and persisted until the termination of the study. Control chimeras transplanted with a mixture of MRL/+ plus A-Thy BM were stably engrafted with both donor strains in both the erythroid and lymphoid populations. Additional experiments in which either B6/lpr or MRL/lpr (and B6/+ or MRL/+ control) BM cells were transferred into (MRL/lpr X B6/+)F1 and (MRL/lpr X B6/lpr)F1 recipients demonstrated that the development of GVHD was not simply due to increased alloreactivity by the lpr donor cells. In these chimeras only the recipients heterozygous (but not homozygous) for the lpr gene developed lpr-GVHD, although both types of recipients had identical genotypes except at the lpr locus.

The lpr gene is associated with resistance to engraftment by lymphoid but not erythroid stem cells from normal mice.

This study demonstrates cell lineage-specific resistance to engraftment involving lymphocytes but not erythrocytes by the spontaneously autoimmune MRL/lpr mouse strain. In these experiments, MRL/lpr mice were lethally irradiated (1000 R) and reconstituted with normal A-Thy bone marrow stem cells. Periodic analysis from 6 wk to 6 mo posttransplantation demonstrated that the T and B cells of these chimeras were derived from the MRL/lpr host. However, in the same A-Thy----MRL/lpr chimeras, erythrocyte repopulation was completely of A-Thy donor origin. In contrast, control MRL/+ (congenic mice that differ from MRL/lpr at the lpr locus and do not develop accelerated autoimmune disease) recipients were successfully repopulated in both the lymphoid and erythroid compartments by the A-Thy donor cells.

A rapid method for comparing monoclonal antibodies by limited proteolysis and electrophoresis.

A rapid and sensitive method for comparing the primary structure of proteins has been adapted to the study of monoclonal antibodies. Samples were digested with alpha-chymotrypsin in the presence of sodium dodecyl sulfate after which peptide fragments were separated into distinctive banding patterns by polyacrylamide gel electrophoresis. This method could easily detect differences in the primary structure of antibodies with related as well as unrelated binding specificities. In addition, antibody molecules derived by somatic diversification from the same germ line gene segments could be distinguished from one another.

Induction of idiotope suppression in the anti-azophenylarsonate response of T-depleted A/J mice.

The homologous, monoclonal antiidiotope, MB, induced idiotope suppression that was remarkably stable and could be transferred by B lymphocytes. Marked depletion of T cell function, confirmed by limiting diluting analysis, did not affect the ability of MB to suppress the corresponding idiotope. Suppression induced by MB appears to result from direct interaction with idiotope-positive B cells, without the intervention of idiotope-specific T suppressor cells.

Specific regulation of c-myc oncogene expression in a murine B-cell lymphoma.

The c-myc oncogene has been implicated in a wide spectrum of B-cell neoplasias. In normal cells, the level of expression of the c-myc gene correlates with growth status. In the present study, we examined the effect of receptor-mediated inhibition of growth on c-myc expression in a B-cell lymphoma. The murine lymphoma line WEHI 231 has been characterized as an early B cell; it bears surface-bound IgM and has unrearranged c-myc genes. Following treatment of a WEHI 231 culture with anti-mouse Ig antiserum, the cells undergo one round of division and further proliferation is inhibited. We observed that this treatment specifically affected cytoplasmic levels of c-myc mRNA. An initial early increase is followed by a precipitous drop such that by 4 hr (after exposure) the amount of c-myc mRNA is below control values by a factor of approximately equal to 10. The drop in c-myc precedes cessation of DNA synthesis. During the 2- to 4-hr period, c-myc mRNA had a maximal half-life of between 20 and 30 min. In contrast, even 24 hr after anti-Ig exposure, the amounts of most major mRNAs, including mu heavy chain and actin, were not significantly altered. These results indicate that expression of an unrearranged c-myc gene can be selectively responsive to receptor-mediated regulatory events.

High-affinity monoclonal antibodies for aflatoxins and their application to solid-phase immunoassays.

Monoclonal antibodies specific for aflatoxin B1, aflatoxin B2, aflatoxin M1, and the major aflatoxin-DNA adducts were obtained following fusion of mouse SP-2 myeloma cells with spleen cells of mice immunized with aflatoxin B1 covalently bound to bovine gamma globulin. The aflatoxin-modified protein used to immunize mice was produced chemically by activating aflatoxin B1 to a 2,3-epoxide derivative, which then covalently bound to the protein. One of the monoclonal antibodies isolated (2B11) was found to be a high-affinity IgM antibody with an affinity constant for aflatoxin B1, aflatoxin B2, and aflatoxin M1 of about 1 X 10(9) liters per mol. In a competitive radioimmunoassay using [3H]aflatoxin B1, 3 pmol (1 ng) of aflatoxin B1, aflatoxin B2, or aflatoxin M1 caused 50% inhibition with this antibody. The antibody also had significant cross-reactivity for the major aflatoxin-DNA adducts: 2,3-dihydro-2-(N7-guanyl)-3-hydroxyaflatoxin B1 and 2,3-dihydro-2-(N5-formyl-2',5', 6'-triamino-4'oxo-N5-pyrimidyl)-3-hydroxyaflatoxin B1. The antibody was also covalently bound to Sepharose-4B and used in a column-based solid-phase immunosorbent assay system. Aflatoxins added in vitro to phosphate buffer, human urine, human serum, or human milk at levels expected to be obtained in human samples acquired from environmentally exposed individuals were quantitatively recovered by applying the mixture to this antibody affinity column purification system. Preliminary studies using urine samples from rats injected with radiolabeled aflatoxin B1 have also indicated that aflatoxin metabolites can be isolated by these methods. Furthermore, we have found that the monoclonal antibody affinity columns can be regenerated for multiple use. Therefore, the monoclonal antibodies and their application to affinity chromatography represents a useful and rapid technique to purify environmentally occurring levels of this carcinogen and some of its metabolites for quantitative measurements.

Fine specificity of idiotope suppression in the A/J anti-azophenylarsonate response.

Two hapten-inhibitable murine monoclonal antiidiotopic antibodies identified two idiotopes expressed by the heavy chain of hybridoma protein 36-65, whose amino acid sequence is encoded in the germ line of A/J mice. Among cross-reactive idiotype-positive hybridoma proteins and p-azophenylarsonate-immune antibodies, the two idiotopes were not always expressed together; some diversified antibodies expressed one idiotope without the other. Suppression that was induced by the two antiidiotopes was idiotope specific and corresponded to the fine specificities of these two reagents.

The genetic basis of antibody production: the dominant anti-arsonate idiotype response of the strain A mouse.

Immunization of the A strain of mice with the hapten p-azophenylarsonate (Ars) results in an immune response in which approximately 50% of the anti-Ars antibodies share cross-reactive idiotypic determinants (IdCR). A gene or genes linked to the heavy chain constant region locus is required for the production of this idiotype. The expressed VH gene from a hybridoma cell line which expresses the IdCR has been cloned. DNA hybridization studies utilizing the VH gene have revealed that there are many related genes in both idiotype-producing and idiotype-nonproducing strains of mice. However, under stringent hybridization conditions, only a single band of 6.4 kb is present in Eco R1-digested A strain DNA. Strains of mice which are phenotypically idiotype-negative either lack this band completely or possess a much weaker one at this position. Utilizing DNA from Igh recombinant strains of mice, it has been shown that the VH locus controlling idiotype expression contains the structural gene information for the idiotype-positive heavy chains. It has also been shown that DNA at this locus appears to be sufficient for the production of the cross-reactive idiotype. Utilizing a DNA probe derived from regions flanking the structural gene has confirmed the relatedness of V genes in a variety of mouse strains and revealed a significant degree of polymorphism at the Igh locus.

Two structurally distinct and independently regulated idiotypic families associated with the A/J response to azophenylarsonate.

At least two distinct subpopulations of A/J anti-azophenylarsonate (Ars) antibodies can be defined on the basis of serological cross-reactivity and sequence homology of heavy and light chains. The most frequently occurring idiotype family, IdCR, was initially identified by a rabbit antiserum and is present only in A/J mice. Examination of several randomly selected hybridoma proteins which failed to express IdCR-associated determinants revealed a second set of closely related molecules. The second idiotype family, Id36-60, is recognized by a rabbit antiserum prepared against the hybridoma protein 36-60. It is present in practically all A/J and BALB/c, but not B10. A Ars immune sera. On average, A/J immune sera contain approximately one-fifth as much Id36-60 as IdCR. A/J mice can be suppressed for subsequent expression of either idiotype by neonatal injection of anti-IdCR or anti-Id36-60 antisera. Suppression of IdCR does not affect the production of Id36-60 and vice versa. Together, these idiotype systems provide two independent markers for monitoring network regulation of the anti-Ars antibody response in A/J mice.

Identification and quantification of the rat hepatocyte asialoglycoprotein receptor.

The asialoglycoprotein receptor from rat liver was purified by solubilization and affinity chromatography on asialoorosomucoid-Sepharose. The preparation yielded four distinct polypeptides of Mr 40,000-120,000. We prepared a monoclonal antibody that both immunoprecipitates solubilized receptor activity and blocks the binding of galactose-terminal glycoproteins to immobilized receptor. The monoclonal antibody and a rabbit antireceptor antiserum immunoprecipitated all four polypeptide species. Peptide analysis by two-dimensional chromatography of the individual 125I-labeled species showed nearly identical patterns, which also suggested that the four polypeptides have a similar primary structure. To identify and quantitate the asialoglycoprotein receptor on the hepatocyte cell surface, intact cells were iodinated with lactoperoxidase, and the solubilized membranes were treated with antireceptor antibody. The Mr 55,000 and Mr 65,000 species were the major species found. Our results suggest that the Mr of the surface receptor is at least 55,000 and that it comprises between 1-2% of the iodinated hepatocyte surface protein.
