Effects of cimetidine on in vitro transformation of peripheral monocytes to macrophages in healthy volunteers and cancer patients.

Monocyte-to-macrophage transformation is a phenomenon which correlates with the absolute number of mononuclear cells, principally lymphocytes, contained in the culture. The addition of cimetidine to cultures of mononuclear cells from the peripheral blood of healthy volunteers enhances monocyte transformation (probably by way of blocking the H2 receptors of suppressors T lymphocytes) regardless of the absolute number of monocytes in the culture or of basal transformation rates. Removal of the lymphocytes from the cultures lowers the basal transformation rate and prevents the effect of cimetidine. The addition of histamine to the cultures causes significant depression of the monocyte transformation rate; this effect is partly offset by the concurrent addition of cimetidine. In the case of cancer patients, mononuclear cell cultures from peripheral blood fail to respond to cimetidine even though basal transformation rates are not significantly different from those of healthy controls; this suggests some intrinsic impairment of monocyte function, possibly mediated by blocking factors produced by the tumor.

Effect of lithium carbonate on monocyte to macrophage maturation in cancer patients.

A decrease in the maturation process of monocytes into macrophage is observed in cancer patients. In this study the Authors examined: the maturation pattern of monocytes into macrophages in 14 patients of both sexes, between 45 and 70 years of age, with solid tumours of the lung, breast and gastrointestinal tract, without any infection; the variations induced on the maturation pattern as results of the administration of lithium carbonate. The results (evaluated statistically) show that in all patients there was a significant increase of monocytes-macrophages maturation from basal values, after 14 days of lithium carbonate treatment. Moreover, we also observed that 14 days after withdrawing of lithium carbonate, the values of monocytes-macrophages maturation return to their basal ones. The increase, as percentage of mean basal values, is higher in patients having low basal values of maturation.

[Protective effect of lithium carbonate in neutropenia induced by antiblastic agents and evaluation of its long-term efficacy].

The association of lithium carbonate and antiblastic drugs has been studied in 22 patients suffering from tumours to see whether it was able to prevent or attenuate the neutropenising effect and establish whether its protection action on leucocytaemia was not just transitory but was maintained for prolonged treatment also. Patients were submitted to four oncolytic treatment cycles during which administration with LiCO3 was associated with the first and third cycles (LNLN sequence) in 14 patients and with the second and fourth (NLNL sequence) in the other 8. Statistical analysis of the results obtained confirmed that lithium carbonate performs a protective action with respect to antiblastic-induced neutropenia and that the action is long term. The drop in monocytes also seems to diminish after administration of lithium carbonate.

[The chordoma. Description of 2 clinical cases and results of oncolytic chemotherapy].

Only seven hundred cases of chordoma approximatively are reported in medical literature from 1850 to date. We now describe two cases recently observed at our Medical Division, one of sacro-coccigeal chordoma and the other developed from clivus of Blumenbach. Their peculiarities are briefly discussed and compared with a rapid revision of the literature on this argument. The results of antiblastic therapy are presented and its opportunity in addition to Roentgenologic and surgical therapy is discussed.

[Albers-Schoenberg disease in adults. Description of a case].

-- A benign form of Albers Schonberg disease (osteopetrosis) is described. The clinical and pathogenetical aspects are discussed and the findings compared with the literature. The diagnosis was made on the basis of radiological findings and histological results of bone biopsy.

Polymerase chain reaction (PCR) amplification of hypervariable genomic sequences.

This paper describes a rapid method for VNTRs (variable number of tandem repeats) typing using polymerase chain reaction (PCR). Three VNTRs (YNZ22, Apo B, MCT118) were amplified and alleles mendelian segregation was confirmed. We also demonstrate their applicability to paternity testing and forensic purposes.

Aneurysms of azygous anterior cerebral artery. Report of two cases treated by surgery.

The occurrence of a so called azygous anomaly of the distal ACA, consisting in a single branch, is rare. Authors report two cases of azygous aneurysm on which surgery was performed successfully, and describe their angiographic characteristics, the frequent association of an aneurysm with azygous and the morphological anomalies of azygous. A literature review is made.
